    인간 피부는 미용 및 성형외과에서 핵심적인 치료 표적이지만, 경피 전달은 각질층 장벽과 여러 기존 전임상 모델의 낮은 인간 적용성으로 인해 제약을 받습니다. 본 학위논문은 방법론적 개요로 시작하며, ex vivo 인간 피부 절편 모델을 단기간 생존이 가능한 시간 창 내에서 표준화된 기능적·조직학적·분자적 지표를 통해 장벽 기능과 조직 리모델링을 평가할 수 있는 적절한 중간 플랫폼으로 제시합니다. 또한 본 분석은 제한된 유지 기간과 관류 및 면역세포 유입의 부재 등 ex vivo 모델의 실질적 한계를 정리하여, 결과 해석과 연구 설계에 반영할 수 있도록 합니다.
    피부 노화와 상처 치유를 동시에 해결하기 위해, 저희는 히알루론산과 콜라겐으로 구성된 용해성 미세침과 이온도입 기술을 통합한 신규 경피 전달 플랫폼인 CELLADEEP 패치를 개발하였습니다. 본 연구는 포괄적인 ex vivo 인간 피부 모델과 in vivo 모델을 활용하여, 피부 재생/리쥬버네이션 촉진 및 상처 치유 가속 효과를 평가하는 것을 목표로 하였습니다.
    피부 리쥬버네이션 평가를 위해, ex vivo 인간 피부 조직을 무처치군, UVB 조사군, CELLADEEP 처리군으로 구분하였습니다. 평가는 피부 수분, 경피수분손실, 탄성, 조직학적 분석, 유전자 발현 프로파일링을 포함하였습니다. 상처 치유 평가를 위해서는 쥐에서 전층 상처를 유도하고 CELLADEEP 처리 유무에 따라 치료하였습니다. 치유 결과는 육안적 관찰, 조직학, 면역형광, 그리고 염증 반응·조직 리모델링·재생 표지를 표적으로 하는 생화학 분석을 통해 평가하였습니다.

    The human skin is a crucial therapeutic target in cosmetic and plastic surgery; nevertheless, transdermal delivery is constrained by the stratum corneum barrier and the inadequate human relevance of several traditional preclinical models. This dissertation commences with a methodological overview that establishes ex vivo human skin explant models as a pertinent intermediate platform for assessing barrier function and tissue remodeling through standardized functional, histological, and molecular endpoints within a viable short-term timeframe. This analysis elucidates the practical limitations of ex vivo models, such as restricted duration and absence of perfusion immune recruitment, to inform suitable interpretation and study design.
    To address both skin aging and wound repair, we developed the CELLADEEP Patch-a novel transdermal delivery platform integrating dissolvable microneedles composed of hyaluronic acid and collagen with iontophoresis technology. This study aimed to evaluate its efficacy in promoting skin rejuvenation and accelerating wound healing using comprehensive ex vivo human skin models and in vivo models.
    For skin rejuvenation, ex vivo human skin tissues were divided into untreated, UVB-irradiated, and CELLADEEP-treated groups. Assessments included skin hydration, transepidermal water loss (TEWL), elasticity, histological analysis, and gene expression profiling. For wound healing, full-thickness wounds were induced in rats and treated with or without CELLADEEP. Healing outcomes were evaluated via macroscopic observation, histology, immunofluorescence, and biochemical assays targeting inflammatory response, tissue remodeling, and regenerative markers.
    CELLADEEP significantly improved hydration and elasticity while reducing TEWL in ex vivo tissues. Histological analysis showed increased collagen fiber density, and gene expression revealed upregulation of COL1A1 and HAS3 and downregulation of MMP-1 and IL-1β, indicating anti-aging and anti-inflammatory effects. In vivo, CELLADEEP treatment accelerated wound closure, reduced wound width, and promoted thicker epidermal regeneration. It also decreased inflammatory mediators (NO, ROS, IL-6, TNF-α) and macrophage activity (CD68, MCP-1, F4/80), while enhancing fibroblast activation and angiogenesis, as shown by increased α-SMA, vimentin, VEGF, collagen I, and TGF-β1 expression.
    The final section synthesizes the extensive preclinical-to-clinical approach established during my PhD, highlighting reproducible outcomes within managed risk, underpinned by biologically interpretable evidence across many contexts. In this context, ex vivo human skin models serve as the primary, human-relevant screening and tissue response platform, while animal and human research is employed to establish safety parameters and validate clinically significant outcomes. This methodological framework is utilized in various translational projects, encompassing exosome-enhanced microneedling, CO₂ cryotherapy combined with EGF delivery, 3D-bioprinted human adipose-derived stem cells (hADSCs) lymph node constructs, noble gas plasma safety assessment, and a randomized controlled trial of Gynostemma pentaphyllum (GP).
