대기 중 이산화탄소 (CO2) 농도의 증가는 기후변화를 야기하며, 식물의 광합성뿐만 아니라 생체량, 번식, 표현형, 그리고 타 생물과의 상호작용에까지 직•간접적인 영향을 미친다. 본 연구에서는 기후변화 환경을 직접 모사하여 생물의 반응성을 확인할 수 있는 open-top Chamber (OTC)를 활용하여 실험을 수행하였다. 한국 고유종은 서식 범위와 생태학적 지위가 제한적이어서 기후변화에 취약할 것으로 예상되나, 실제 기후변화에 대한 반응을 실험적으로 규명한 연구는 미비한 실정이다. 이에 본 연구는 OTC를 활용하여 증가한 CO2 환경에서의 한국 고유종의 반응성을 최초로 정량 평가하고, 다양한 생리•성장•상호작용 지표를 통합한 실험 파이프라인을 구축하고자 하였다. 이를 위해 수확 시점의 생체량과 엽장 등 성장 형질에서 초기값을 뺀 상대생장률을 산출하여 종 및 처리 간 직접 비교가 가능하도록 하였으며, 반복 측정을 통해 성장 궤적을 추적함으로써 데이터의 연속성을 확보하였다. 또한, CO2 반응의 핵심 지표인 동화율과 증산율을 측정하고, CSR (Competitor, stress-tolerator, ruderal) 전략 분석을 통해 식물의 생존 전략을 평가하였다. 나아가 개체 단독 성장뿐만 아니라 경쟁 상황 및 광식성 초식곤충인 담배거세미나방 (Spodoptera litura) 처리에 대한 반응까지 포함하여, 다각적인 시나리오에서 식물의 반응성을 검증하였다. 이러한 실험 방법론은 향후 다양한 초본 식물에 즉각 적용 가능한 범용성과 재현성을 갖춘 파이프라인을 제공한다.
제2장에서는 OTC의 안정적인 운영과 식물 실험 파이프라인 구축에 중점을 두었다. 고산 지역에 분포하며 유사한 생태학적 지위를 공유하는 한국 고유종 산솜다리(Leontopodium leiolepis)와 자생종 가는잎구절초(Dendranthema zawadskii var. tenuisectum)를 동일한 OTC 내에서 재배하였다. 대기 중 CO2 농도 상승은 두 종 모두의 동화율을 증가시켰으나, 수확 시점에는 성장이 포화 상태에 도달하여 생체량의 유의한 차이는 나타나지 않았다. 2주 간격으로 잎의 개수와 길이를 측정하여 성장 추이를 분석한 결과, 산솜다리는 CO2 처리에 따른 유의한 차이를 보이지 않은 반면, 가는잎구절초는 잎 길이가 증가하는 표현형 가소성을 나타냈다. 또한 곤충 구강분비물 (Oral secretion) 처리와 클립케이지를 이용한 실제 섭식 실험을 병행하여 자스몬산 농도를 측정한 결과, 두 종의 상이한 기후변화 반응성과 방어 호르몬 기작을 확인하였으며, 이를 통해 성장•광합성•곤충 섭식을 포괄하는 통합 실험 방법론을 정립하였다.
제3장에서는 개별 종의 성장 반응을 넘어, 서로 다른 생태적 시나리오에서 나타나는 식물 간 경쟁 양상을 평가하였다. 대상종은 습윤한 서식지에 널리 분포하는 고유종 벌개미취 (Aster koraiensis)와 특히 교란지를 중심으로 습윤 지역으로 분포를 확장하고 있는 외래종 큰금계국 (Coreopsis lanceolata)으로 선정하였다. 기후변화로 인한 잠재적 경쟁 심화 가능성을 검토하기 위해 SSP2-4.5 시나리오를 가정하여 종분포모델링을 수행하였다. 그 결과 두 종 모두 대한민국 전역에서 잠재 분포가 확대되고 서식지 중첩이 증가하는 것으로 예측되었다. 이러한 동시 출현 가능성 증가 예측을 바탕으로, OTC 실험을 통해 기후변화 조건에서 두 종의 경쟁 상호작용을 직접 검증하였다. 실험은 단독 성장, 종내 경쟁, 종간 경쟁의 세 조건으로 구성하였다. 이를 바탕으로 (1) 벌개미취 자생지에 큰금계국이 침입하는 조건, (2) 교란지에서 두 종이 경쟁하는 조건, (3) 경쟁 상황 하에서의 담배거세미나방 섭식 조건을 모사하였다. 벌개미취 자생지 침입 시나리오에서 유묘 단계의 큰금계국은 종내•종간 경쟁 조건 모두에서 뿌리줄기 유래 벌개미취 개체보다 빠른 생체량 및 초장 성장을 보였다. 큰금계국은 CO2 상승 및 경쟁 조건에서 증산율을 감소시켜 수분 이용 효율을 높였으나, 이는 생체량 수준 변화로 연결되지 않았고, 벌개미취와의 생체량 차이에도 뚜렷한 영향을 미치지 않았다. 두 종 모두 유묘 단계에서 경쟁시킨 실험에서는 CO2 농도와 경쟁 조건에 관계없이 큰금계국이 생체량과 잎 개수에서 벌개미취보다 우위를 점하였다. 생체량에는 뚜렷한 영향을 미치지 않았지만, CO2 상승 조건에서 큰금계국은 현재 CO2 조건과 비교하여 유의미한 동화율의 증가를 보였고, 벌개미취보다 일관되게 높은 값을 보였다. 두 종 모두 CO2 상승 및 단독 재배 조건에서 최저 증산율을 보였다. CSR 전략 분석 결과, 유묘 단계에서는 R/CR 전략을, 이후 생장이 진행된 단계에서는 빠른 성장에 유리한 C/CR 전략을 보여 이는 선행 연구와 일치하였다. 담배거세미나방은 식물 종이나 CO2 처리에 따른 뚜렷한 섭식 선호를 보이지 않았으나, 경쟁 조건에서 벌개미취의 생체량만 유의하게 낮았다. 이러한 다층적 실험 설계를 통해 보다 현실적인 생태 시나리오를 모사함으로써 식물의 경쟁 및 초식 반응을 정밀하게 평가할 수 있었다.
종합하면, 본 연구는 OTC를 활용하여 한국 고유종의 기후변화 반응성을 정량적으로 제시한 초기 사례로서, 식물 종에 따라 기후변화 반응이 상이함을 보여준다. 실험은 제한된 환경과 단순화된 군집 구조에서 이루어졌기 때문에 실제 서식지에서의 상호작용 강도가 과소평가되었을 가능성이 있다. 이러한 한계는 향후 더 많은 종과 다양한 경쟁•초식 시나리오 조합을 포함하는 실험을 통해 보완될 수 있을 것이다. 본 연구에서 제시한 OTC 기반 실험 파이프라인은 다양한 초본 식물에 확장하기 용이하며, 산출된 정량화된 데이터는 기후변화 취약성 평가와 종 보전 관리 전략 수립에 직접적으로 기여할 것으로 기대된다.

Elevated atmospheric carbon dioxide (CO2) concentrations are a primary driver of climate change, exerting direct and indirect effects on plant photosynthesis, growth, biomass, phenology, and interactions with other organisms. In this study, experiments were conducted using open-top chambers (OTCs) to simulate future climate CO2 conditions and quantify biological responses. Although Korean endemic species are expected to be vulnerable to climate change due to their restricted habitats and narrow ecological niches, their responses to climate drivers remain largely unquantified experimentally. Here, the first quantitative assessment of the responsiveness of Korean endemic species to elevated CO2 using OTCs is presented, together with an experimental pipeline encompassing physiological, growth, and interaction metrics. To account for initial size differences, relative growth rate was calculated by subtracting initial values from harvest traits such as biomass and leaf length, facilitating direct comparisons among species and treatments. Repeated measurements were employed to track growth trajectories, allowing for longitudinal data analysis. Additionally, assimilation and transpiration rates-key indicators of CO2 response-were measured, and survival strategies were evaluated based on Grime's CSR (Competitor, stress-tolerator, ruderal) framework. Finally, plant responses were examined across diverse ecological scenarios, including solitary growth, intra- and inter-specific competition, and herbivory by the generalist insect Spodoptera litura. This methodology offers a versatile and reproducible framework readily applicable to a wide range of herbaceous plant species.
Chapter 2 focuses on optimizing OTC operation and establishing the experimental pipeline. Leontopodium leiolepis (Korean endemic) and Dendranthema zawadskii var. tenuisectum (Korean native), which share similar ecological niches in alpine regions, were co-cultivated within the same OTCs. Elevated CO2 increased assimilation rates in both species; however, as growth reached saturation by the final harvest, no significant differences in biomass were observed. Growth trajectories were tracked via bi-weekly measurements of leaf number and length. L. leiolepis showed no significant response to CO2 treatments, whereas D. zawadskii var. tenuisectum exhibited phenotypic plasticity, evidenced by increased leaf length under elevated CO2. To simulate herbivory, oral secretion treatments and direct feeding assays using clip cages were conducted, followed by the measurement of jasmonic acid concentrations. These experiments revealed species-specific plasticity in response to climate change and defense hormone dynamics, establishing an integrated framework covering growth, photosynthesis, and herbivory.
Chapter 3 extends the evaluation to plant competition under varying ecological scenarios. The Korean endemic Aster koraiensis, which is widely distributed in moist habitats, and the alien species Coreopsis lanceolata, which is expanding into moist areas-particularly in disturbed sites-were targeted. To evaluate whether climate change could increase potential competition, species distribution models were built under the SSP2-4.5 scenario. The models predicted range expansion for both species across the Republic of Korea and increased habitat overlap. Motivated by this predicted increase in co-occurrence, OTC experiments were conducted to directly test climate change driven competitive interactions between the two species. Experimental conditions included solitary growth, intra-specific competition, and inter-specific competition. These setups simulated three scenarios: (1) invasion of A. koraiensis native habitats by C. lanceolata, (2) competition in disturbed habitats, and (3) herbivory by S. litura under competition. In the invasion scenario, seedling-stage C. lanceolata exhibited faster biomass accumulation and vertical growth than rhizome-derived A. koraiensis under both competition types. Under elevated CO2, C. lanceolata improved water-use efficiency by reducing transpiration during competition, yet this physiological adjustment did not further exacerbate the biomass disparity with A. koraiensis. In seedling-stage competition, C. lanceolata consistently outcompeted A. koraiensis in biomass and leaf number, regardless of CO2 levels or competition treatment. Although biomass was not significantly affected, C. lanceolata consistently exhibited higher assimilation rates than A. koraiensis under elevated CO2. Under competitive conditions, C. lanceolata significantly increased its assimilation rate compared to ambient CO2 levels. For both species, transpiration rates were lowest under elevated CO2 in solo stands. CSR analysis indicated a shift from an R/CR strategy at the early seedling stage to a C/CR strategy at later stages, consistent with previous literature. S. litura showed no distinct feeding preference regarding plant species or CO2 treatment, and biomass patterns of A. koraiensis under competition remained significantly lower regardless of herbivory. This multi-layered design allowed for a precise evaluation of competitive and herbivore responses within ecologically realistic scenarios.
In summary, this study provides one of the first quantitative assessments of the climate-change responsiveness of Korean endemic plants using OTCs, highlighting species-specific variations in response to elevated CO2. While the controlled environment and simplified community structure may underestimate the complexity of natural interactions, future research incorporating a broader range of species and interaction scenarios can address these limitations. Nevertheless, the OTC-based experimental pipeline developed herein is readily adaptable to diverse herbaceous species, providing standardized data essential for climate change vulnerability assessments and conservation strategy development.
