정확한 수의적 힘 조절은 인간 운동 제어의 핵심 요소이다. 그러나 기존 연구들은 등척성 과제에서 힘을 증가시키는 구간에 비해 힘을 감소시키는 구간에서 수행 정확도가 낮고 변동성이 크다는 점을 일관되게 보고해 왔다. 이러한 비대칭성의 신경학적 기전은 아직 명확히 규명되지 않았다. 그 잠재적 원인 중 하나로 척수 운동뉴런에서 발생하는 지속성 내향전류(Persistent Inward Current, PIC)가 제시된다. PIC는 모노아민성 입력에 의해 매개되는 지속적인 탈분극 전류로, 시냅스 흥분을 증폭시키는 역할을 한다. PIC는 운동단위(motor unit, MU)의 모집과 힘 생성을 촉진하지만, 활성화가 지속될 경우 MU의 탈모집을 지연시켜 힘 감소 구간에서의 정밀한 조절을 방해할 가능성이 있다.
본 연구의 목적은 수의적 등척성 무릎 신전 과제에서 힘 조절 정확도와 PIC 활성 간의 관계를 규명하는 것이다. 참가자들은 최대 수의적 토크(Maximal Voluntary Torque, MVT)의 50%를 목표로 선형적인 램프 업과 램프 다운 구간으로 구성된 삼각형 토크 추적 과제를 수행하였다. 무릎 신전근의 표면 근전도(EMG)는 Delsys Trigno Galileo 시스템을 이용해 측정하였으며, Neuromap 분해 알고리즘을 통해 개별 운동단위를 추출하였다. PIC 크기는 paired motor unit technique(PI 방법)을 통해 추정하였고, 토크 추적 정확도는 목표 토크와 실제 토크 간의 제곱평균제곱근 오차(RMSE)로 정량화하였다.
그 결과, 램프 다운 구간에서의 RMSE가 램프 업 구간에 비해 유의하게 높게 나타나(p = 0.007), 힘 감소 시 정확도가 더 낮음을 확인하였다. 또한 PIC 크기(PI)와 힘 감소 구간의 오차 간에는 유의한 상관관계가 관찰되었으며(r = 0.44, p = 0.015), 특히 FAST 조건에서 가장 강한 상관관계가 나타났다(r = 0.68, p = 0.032). 이러한 결과는 PIC 활성 증가가 운동뉴런의 탈활성을 지연시켜 정밀한 토크 감소를 방해함을 시사한다.
종합하면, 본 연구는 운동뉴런의 내재적 흥분성이 수의적 힘 조절의 행동적 정확도와 직접적으로 연관되어 있음을 실험적으로 입증하였다. PIC는 수축 동안 탈분극을 유지함으로써 힘 생성을 촉진하는 반면, 힘 감소 구간에서는 토크 조절 정확도를 저해할 수 있으며, 이러한 영향은 힘 감소 속도가 증가할수록 더욱 두드러진다. 향후 연구에서는 PIC가 운동 단계에 따라 토크 조절을 어떻게 촉진하거나 방해하는지를 정량적으로 규명할 필요가 있다. 이러한 이중적 역할에 대한 이해는 파킨슨병이나 경직(spasticity)과 같이 모노아민성 입력이 변화된 신경계 질환에서 관찰되는 운동 제어 장애를 설명하는 데 중요한 단서를 제공할 것이다.

Precise modulation of voluntary force is fundamental to human motor control. Yet, previous studies have consistently reported that individuals display reduced accuracy and greater variability when decreasing force compared to when increasing it during isometric tasks. The underlying neural mechanism for this asymmetry remains unclear. One potential contributor is the Persistent Inward Current (PIC) in spinal motoneurons-a sustained depolarizing current mediated by monoaminergic inputs that amplifies synaptic excitation. While PIC facilitates motor unit (MU) recruitment and force generation, its prolonged activation may interfere with MU de- recruitment, thereby imparing fine control during force reduction.
This study aims to examine the relationship between force control accuracy and PIC activation during voluntary isometric knee extension. Participants performed a triangular torque-tracking task consisting of linear ramp-up and ramp-down phases at 50% of their maximal voluntary Torque (MVT). Surface electromyographic (EMG) signals were recorded from the knee extensor muscles using the Delsys Trigno Galileo system, and individual MUs were extracted via the Neuromap decomposition algorithm. PIC magnitude was estimated using the paired motor unit technique (PI method), and torque-tracking accuracy was quantified as the root mean square error (RMSE) between target and torque trajectories.
Results showed that RMSE was significantly higher during the ramp-down phase than during ramp-up (p = 0.007), indicating lower accuracy when decreasing force. Moreover, the relationship between PIC magnitude (PI) and force-release error was significant across conditions (r = 0.44, p = 0.015), with the strongest correlation in the FAST condition (r = 0.68, p = 0.032). These findings suggest that greater PIC activation impairs precise torque reduction, likely due to delayed motoneuron deactivation. Collectively, this study provides experimental evidence linking intrinsic motoneuron excitability to behavioral precision in voluntary force control
These findings indicate that PIC impairs force-release accuracy by delaying motor unit de-recruitment, and this effect becomes more pronounced at higher release speeds. However, PIC may also facilitate force generation by sustaining depolarization during contraction. Future work should aim to quantify how PIC alternately supports or hinders torque modulation across phases of movement. Understanding this dual role could help explain impaired motor control in conditions with altered monoaminergic input, such as Parkinson’s disease or spasticity.
