    1. 배경 및 목적
    삼차원 바이오프린팅은 조직 재생을 위해 중요한 역할을 하고 있으며 효율적인 바이오구조물의 개발은 더욱 중요해지고 있다. 본 연구는 새로운 바이오프린팅 기법을 사용한 바이오구조물의 조직 재생 가능성을 평가하기 위해 시행되었다.

    2. 방법
    오메가 3 지방산인 Eicosapentaenoic acid(EPA)와 docosahexaenoic acid(DHA)를 포함한 치은 섬유아세포가 있는 콜라겐 바이오잉크를 제작하고, 기계적 신호전달을 강화하기 위한 전단 유도 바이오프린팅 기법(comb-assisted bioprinting)을 적용하였다. 일반 바이오프린팅을 이용한 콜라겐만 포함된 바이오구조물 (Con), EPA/DHA를 함유한 일반 바이오프린팅 구조물 (Con-ColED)과 EPA/DHA 함유한 comb-보조 바이오프린팅 구조물 (S-ColED)을 제작하였다. In vitro 실험에서는 Con과 Con-ColED의 조직 재생 및 혈관 형성 효과를 비교 분석하였다. 또한, 총 30마리의 생쥐 피하조직에 Con, Con-ColED, 그리고 S-ColED를 이식한 후 EPA/DHA 및 comb-보조 바이오프린팅이 조직 재생 및 혈관형성에 미치는 영향을 평가하였다.

    3. 결과
    Con-ColED에서는 Con에 비해 F-actin 형성과 콜라겐 I 발현 그리고 조직 재생 및 혈관 형성과 관련된 유전자 발현이 통계적으로 유의하게 증가하였다(p < 0.001). 또한 관 형성 분석(Tube formation assay) 결과, Con-ColED는 Con에 비해 우수한 혈관 형성 능력을 보였다. In vivo 실험에서는 이식 4주 후에 S-ColED가 Con 및 Con-ColED에 비해 유의하게 증가된 콜라겐 영역을 나타냈다(p < 0.05). 면역형광 분석 결과, Con 및 Con-ColED보다 S-ColED에서 더 증가된 콜라겐 III 침착을 보였다(4주; p < 0.05, 8주; p < 0.01). Con-ColED와 S-ColED 모두 Con에 비해 혈관 형성이 유의하게 증가하였으며(p < 0.01), 특히 S-ColED는 이식 후 8주에 더 많은 모세혈관 침투를 나타냈다(p < 0.05).

    4. 결론

    Background and Objective:
    Three-dimensional (3D) bioprinting has emerged as a key technology in periodontal tissue regeneration and key challenges include the development of optimal biomaterials. This study aimed to evaluate a new bioconstruct with mechanical stimulation for tissue reconstruction.

    Materials and Methods:
    A human gingival fibroblasts (hGFs)-laden collagen bioink incorporating the omega-3 fatty acids, eicosapentaenoic acid (EPA) and docosahexaenoic acid (DHA) was developed and a shear-inducing bioprinting approach was applied to enhance mechanotransduction. Collagen-only bioink with normal bioprinting (Con), EPA/DHA-containing bioconstructs with either normal bioprinting (Con-ColED) or comb-assisted bioprinting (S-ColED) were prepared. In vitro studies were performed to compare the effects of tissue regeneration between Con and Con-ColED. Using thirty nude mice, in vivo subcutaneous implantation of Con, Con-ColED and S-ColED constructs was performed to investigate the effects of EPA/DHA and comb-assisted bioprinting on tissue regeneration and angiogenesis.

    Results:
    Con-ColED significantly enhanced F-actin formation and collagen I expression at 7 days and upregulated pro-regenerative and pro-angiogenic gene expression compared to Con (p < 0.001). Tube formation assay revealed a superior angiogenic performance in Con-ColED compared to that in Con. In vivo experiments revealed a significant increase of collagen deposition in S-ColED compared to Con and Con-ColED at 4 weeks (p < 0.05). Immunofluorescence analysis further showed an increase of collagen type III deposition in S-ColED compared to Con and Con-ColED (p < 0.05 at 4 weeks and p < 0.01 at 8 weeks). Both Con-ColED and S-ColED demonstrated markedly enhanced vessel formation compared to Con (p < 0.01), with the S-ColED exhibiting significantly greater capillary infiltration at 8 weeks (p < 0.05).

    Conclusion:
    These findings indicate that EPA/DHA-assisted bioinks, in conjunction with mechanically stimulatory bioprinting provide a promising strategy for advancing tissue regeneration.
