    수면장애와 주요 우울 장애 (Major Depressive Disorder, MDD)의 양방향 인과관계는 잘 알려져 있으나, 수면 시간과 MDD의 인과관계는 여전히 불명확하다. 본 논문은 양방향 멘델 무작위화를 적용하여, single nucleotide polymorphisms (SNPs)을 도구변수로 사용해 유전적으로 예측된 수면 시간이 MDD 발생을 유발하는지, 반대로 유전적으로 예측된 MDD 발생이 부적절한 수면 시간을 유발하는지 검정하였다.
    연구 결과, 짧은 수면은 MDD 발생과 인과적 연관성을 보였으나 (OR 1.739, 95% CI 1.117–2.708, P = 0.019), 긴 수면은 유의하지 않았다 (OR 1.526, 95% CI 0.669–3.480, P = 0.315). 반대로, MDD는 긴 수면을 유발하는 결과를 보였다 (OR 1.015, 95% CI 1.010–1.021, P = 1.002 × 10-7).

    Background: Although the bidirectional relationship between sleep disturbances and major depressive disorder (MDD) is well established, the causal relationship between sleep duration and MDD remains unclear.
    Methods: The present study applied bidirectional Mendelian randomization (MR) using single nucleotide polymorphisms (SNPs) as instrumental variables to evaluate whether genetically proxied sleep duration causally influences the risk of developing MDD, and conversely, whether genetic liability to MDD contributes to aberrant sleep duration.
    Results: Short sleep duration showed evidence of a positive association with MDD risk (OR: 1.739, 95% CI: 1.117–2.708, P = 0.019), whereas long sleep duration did not demonstrate a causal effect (OR: 1.526, 95% CI: 0.669–3.480, P = 0.315). In the reverse MR analyses, genetic liability to MDD was found to be associated with long sleep duration (OR: 1.015, 95% CI: 1.010–1.021, P = 1.002×10-7).
    Conclusion: By minimizing bias from sample overlap, this study provides the first genetic evidence indicating that short sleep duration may be a potential risk factor for MDD, while also showing that MDD may contribute to prolonged sleep duration. These findings, grounded in genetic causal inference analyses, suggest that improving insufficient sleep to an optimal duration may serve as a potential target for the primary prevention of MDD in the general population, and monitoring long sleep duration may support screening and early detection of MDD.
