    미호종개(Cobitis choii)는 금강 수계 저서역에 제한적으로 분포하는 멸종위 기 담수어류로, 수문 구조물 설치와 서식지 단편화, 개체군 감소 등 다양한 환경 적 압력을 받고 있다. 본 연구에서는 약 10년 간격으로 수집된 미호종개 개체군의 microsatellite 유전자형 자료를 이용하여 유전다양성, 개체군 구조, 병목 신호를 종합적으로 평가하고, 동시에 표피성 진균 감염의 병리학적 특징을 규명함으로써 본 종의 보전상 취약성을 통합적으로 분석하였다. 세 개체군(갑천, 지천, 미호강)의 유전다양성 지표(NA, HO, HE)는 이전 연구와 유사한 수준으로 유지되었으며, 평균 이형접합도는 높은 값을 보였다. 개체군 간 유전적 분화는 매우 낮은 범위(FST = 0.005–0.041)에 머물렀고, STRUCTURE 및 DAPC 분석 역시 단일 군집 구조를 지지하였다. 병목 검정에서는 모델 의존적이지 만 최근 규모 축소를 시사하는 약한 신호가 관찰되었으나, M-ratio 및 대립유전자 빈도 분포는 중·장기적 병목의 증거를 보이지 않았다. 이러한 결과는 최근의 국 지적 개체수 감소가 장기적인 유전적 침식으로 이어지지 않았음을 시사하나, 현재 의 유전적 안정성이 지속적으로 유지된다고 보기에는 이른 단계임을 의미한다. 조직병리 분석에서는 Saprolegnia spp.에 의한 표피 감염이 1년생 미만 개체 에서 특히 취약하게 나타났으며, 표피 파괴로 인한 삼투압 장애가 치사율을 증가 시키는 주요 기전으로 확인되었다. 환경적 스트레스(퇴적물 증가, 유량 변동 등)는 감염 위험을 추가적으로 높이는 요인으로 평가되었다. 본 연구는 미호종개가 단일 유전단위를 이루며 아직 심각한 유전적 붕괴는 경험하지 않았으나, 최근의 개체군 감소와 질병 취약성이 결합될 경우 향후 유전 적 침식과 개체군 지속성 저하가 나타날 수 있음을 제시한다. 따라서 본 종의 보 전을 위해서는 서식지 질 유지, 표피성 감염 관리, 그리고 3–5년 주기의 지속적 유전 모니터링이 필수적이다.

    The Miho spined loach Cobitis choii is an endangered benthic freshwater fish restricted to tributaries of the Geum River basin in South Korea. Despite persistent habitat degradation and hydrological modification, no temporal reassessment of its population genetic status has been conducted for more than a decade. This study integrates a decadal comparison of microsatellite variation with an analysis of naturally occurring Saprolegnia infections to evaluate the current conservation status of C. choii. Population genetic analyses of 3 tributaries (Gap, Ji, and Miho Streams) revealed that overall genetic diversity remained comparable to levels reported approximately ten years earlier, with consistently high heterozygosity, Interpopulation differentiation was very low (FST = 0.005– 0.041), and clustering analyses (STRUCTURE and DAPC) supported a single genetic unit with minimal spatial subdivision. Bottleneck tests produced model dependent weak signals of recent demographic reduction, but M-ratio values (Mobs > Mc) and the preservation of L-shaped allele frequency distributions indicated no evidence of long term or severe historical bottlenecks. Histopathological examination demonstrated that Saprolegnia spp. infections were confined to the epidermis, where epithelial disruption caused osmotic imbalance and contributed to mortality. Juvenile C. choii were particularly susceptible, and infection risk increased under environmental stress conditions such as sediment accumulation and flow variability. Although internal organs showed no direct pathology, the combination of disease susceptibility and small or declining population sizes may exacerbate demographic vulnerability. Taken together, these results indicate that C. choii currently maintains moderate genetic diversity and a single population unit despite hydrological fragmentation, but also suggests that early signs of demographic stress and disease vulnerability warrant caution. Sustained genetic monitoring at 3–5 year intervals, maintenance of habitat quality, and management of cutaneous fungal infections are essential to prevent future genetic erosion and ensure the long term persistence of this endangered species.
