연구목적: 본 연구는 황련(Coptis chinensis, 黃連)의 대사질환 치료 효능을 단일 성분이 아닌 다성분·다표적(Multi-component, Multi-target) 관점에서 규명하기 위해 수행되었다. 특히 기존의 베르베린(Berberine) 중심의 환원주의적 해석이 갖는 한계를 극복하고자, 네트워크 약리학과 분자 도킹 분석을 통합한 통합적 네트워크 약리학 방법을 적용하여 황련 복합 알칼로이드의 시스템적 상호작용과 약리 기전을 체계적으로 규명하고자 하였다.
연구방법: TCMSP 데이터베이스 및 문헌 고찰을 통해 황련의 화학 성분을 수집하고, 정량적 약물 유사성(QED) 기준을 적용하여 4종의 주요 활성 알칼로이드(Berberine, Coptisine, Palmatine, Jatrorrhizine)를 선별하였다. 선별된 성분의 예측 표적 유전자와 대사질환 관련 유전자의 교집합을 기반으로 성분-표적-질환(C-T-D) 네트워크를 구축하였다. 이어 단백질-단백질 상호작용(PPI) 및 MCODE 분석으로 핵심 기능 모듈을 도출하고, GO 및 KEGG 풍부도 분석을 통해 주요 생물학적 경로를 해석하였다. 마지막으로 분자 도킹 시뮬레이션을 수행하여 핵심 성분과 표적 단백질 간의 물리화학적 결합 타당성을 검증하였다.
연구결과: C-T-D 네트워크 및 위상학적 분석 결과, 베르베린이 가장 높은 연결성을 보이며 허브(Hub) 역할을 수행하였으나, 콥티신과 팔마틴 등 공존 성분들이 독자적인 네트워크를 형성하며 상호 보완적으로 작용함을 확인하였다. 주요 핵심 표적으로는 TNF, STAT3, PPARγ, AMPK, SIRT1 등이 도출되었으며, 이들은 염증 반응, 지질 대사, 인슐린 감수성 조절과 밀접하게 연관되어 있었다. KEGG 분석 결과, ‘당뇨 합병증에서의 AGE-RAGE 신호전달 경로’, ‘지질 및 죽상동맥경화’ 등 대사성 염증 및 합병증과 직결된 경로가 유의하게 풍부화되었다. 분자 도킹 분석에서는 4종의 주요 알칼로이드가 PPARγ, AMPK 등 핵심 표적의 활성 부위에 –5.0 kcal/mol 이하의 에너지로 안정적으로 결합하여, 예측된 기전의 구조적 타당성을 뒷받침하였다.
결론: 본 연구는 황련의 대사 조절 효능이 베르베린의 단독 작용이 아닌, 다양한 알칼로이드의 약력학적 시너지와 상보적 작용을 통해 대사질환 네트워크를 통합적으로 제어함으로써 발현됨을 입증하였다. 이는 ‘청열조습(淸熱燥濕)’이라는 황련의 전통적 효능을 현대 시스템 생물학 관점에서 ‘대사성 염증 억제 및 에너지 대사 불균형의 회복’으로 재해석한 결과이며, 향후 복합 성분 기반 천연물 신약 개발의 중요한 학문적 근거가 될 것이다.

Objectives: This study was conducted to elucidate the therapeutic efficacy of Coptis chinensis on metabolic diseases from a “Multi-component, Multi-target” perspective, moving beyond the limitations of single-component analysis. Specifically, to overcome the reductionist approach centered on Berberine, this study aimed to systematically investigate the systemic interactions and pharmacological mechanisms of Coptis complex alkaloids by applying a systems pharmacology framework that integrates network pharmacology and molecular docking simulation.
Methods: Chemical constituents of Coptis chinensis were collected via the TCMSP database and literature review. Based on Quantitative Estimate of Drug-likeness (QED) criteria, four major active alkaloids (Berberine, Coptisine, Palmatine, and Jatrorrhizine) were selected. A Compound-Target-Disease (C-T-D) network was constructed based on the intersection between the predicted targets of these alkaloids and metabolic disease-related genes. Subsequently, Protein-Protein Interaction (PPI) and MCODE analyses were performed to identify core functional modules, and GO and KEGG enrichment analyses were conducted to interpret key biological pathways. Finally, molecular docking simulations were carried out to validate the physicochemical binding plausibility between core components and target proteins.
Results: The C-T-D network and topological analysis revealed that while Berberine exhibited the highest connectivity acting as a primary Hub, co-existing components such as Coptisine and Palmatine formed distinct networks, functioning in a complementary manner. Key hub targets identified included TNF, STAT3, PPARγ, AMPK, and SIRT1, which are closely associated with inflammatory response, lipid metabolism, and insulin sensitivity regulation. KEGG analysis showed significant enrichment in pathways directly linked to metabolic inflammation and complications, such as the “AGE-RAGE signaling pathway in diabetic complications” and “Lipid and atherosclerosis”. Molecular docking analysis confirmed that the four major alkaloids stably bind to the active sites of core targets like PPARγ and AMPK with binding energies lower than –5.0 kcal/mol, supporting the structural validity of the predicted mechanisms.
Conclusion: This study demonstrates that the metabolic regulatory efficacy of Coptis chinensis is not solely due to Berberine but is realized through the integrated control of the metabolic disease network via the pharmacodynamic synergy and complementary actions of various alkaloids. These findings scientifically reinterpret the traditional efficacy of “Clearing Heat and Drying Dampness” (Cheong-Yeol-Jo-Seup) as the “suppression of metabolic inflammation and restoration of energy metabolic imbalance” from a modern systems biology perspective. Furthermore, this study provides a significant academic basis for the future development of multi-component botanical drugs.
