화피산(樺皮散)은 『동의보감』을 비롯한 전통 의서에 수록된 처방으로, 피부 표층에 발현되는 은진(癮疹), 풍은진(風癮疹), 담마진 등 가려움과 발진을 특징으로 하는 병증에 활용되어 온 처방으로, 화피(樺皮), 행인(杏仁), 형개(荊芥), 지각(枳殼), 감초(甘草)로 구성되어 있다(Her, 2025). 국내 한의 처방 DB에서도 화피산(樺皮散)은 은진(癮疹), 풍은진(風癮疹), 담마진(蕁麻疹) 등과 같이 ‘갑작스러운 가려움과 두드러짐’이 특징인 피부 증상군에 적용되는 처방으로 소개된다. 이와 관련한 연구로, 화피 및 구성 약재에 대한 항산화, 항염 효과 등은 보고된 바 있으나, 화피산 처방 추출물에 대한 연구는 이루어진 바 없다. 이에 화피, 행인, 형개, 지각, 감초로 구성된 화피산 처방 추출물에 대한 항산화, 항노화 및 세포독성 효과를 측정하여 한약재 혼합 추출물의 생리활성에 대한 기능성을 검증하여 기능성 제품 및 유효 성분 활용을 위한 기초 근거 자료를 제공하고자 한다.
1. 화피산 처방을 물과 에탄올을 용매로 하여 환류 추출한 추출수율은 물 추출물(HRW)가 45.10%, 에탄올 추출물(HRE) 28.73%의 고형분 수율을 나타내었다.
2. 화피산 물과 에탄올 추출물의 환원당 환원당은 HRW 332.48 mg/g, HRE 264.43 mg/g으로 환원당을 함유하는 것으로 나타났으며, HRW가 HRE보다 약 1.2배 많은 환원당을 함유하는 것으로 분석되었다. 기능성 물질을 만드는 효소로서 작용할 수 있는 수용성 단백질의 함량은 HRE에서 88.05 mg/g으로 62.60 mg/g의 함량을 나타낸 HRW보다 더 많은 수용성 단백질을 함유하였다.
3. 총 폴리페놀 함량은 HRE에서 95.00 mg/g으로 70.39 mg/g의 함량을 나타낸 HRW보다 더 높은 함량을 보였다. 총 플라보노이드 화합물 함량 또한 HRE 18.82 mg/g으로 HRW(14.62 mg/g)보다 더 많은 플라보노이드 화합물을 함유하는 것으로 나타났다
4. ABTS radical HRW는 125 μg/mL에서 85.34%의 소거능을 나타내었으며, 2,000 μg/mL에서는 104.50%의 활성을 보였고, HRE는 125 μg/mL에서 66.95%로 HRW에 비해 유의적으로 낮은 소거능을 보였으나, 농도가 증가함에 따라 활성이 증가하여 1,000 μg/mL 이상에서는 99% 이상의 소거능을 나타내었다
5. DPPH radical 소거율을 측정한 결과, 125~2,000 ug/mL의 농도에서 HRW 47.76~76.43, HRE 67.55~75.46%의 소거활성을 보였다
6. 유해한 superoxide를 제거하는 효소와 유사한 효과에 대해 pyrogallol에 대한 자동산화 반응으로 측정한 결과, 2,000 ug/mL의 농도에서 화피산 에탄올 추출물인 HRE 9.79%의 결과를 보였으며, 화피산 물 추출물인 HRW는 2.35%로 낮은 SOD 유사활성을 나타내었다
7. 아질산염 소거효과를 상이한 pH 조건에서 측정한 결과, pH 1.2에서는 2pH 1.2 조건에서는 HRW는 19.51~94.55%였으며, HRE은 5.12~79.87%의 아질산염 소거율을 나타내었다. pH 3.0의 조건에서는 HRW는 3.68~22.42%였으며, HRE은 5.70~30.62%의 소거작용 나타내었다
8. 피부에서 미백효과 및 식품의 갈변화를 저해할 수 있는 tyrosinase에 대한 게걸무와 가을무 추출물에 대한 저해효과를 측정한 결과, 2,000 ug/mL의 농도에서 HRW 57.46%, HRE 47.19%의 저해율을 나타내어, 에탄올 추출물보다는 물 추출물에서 더 우수한 tyrosinase 저해율을 보였다.
9. 피부 진피조직의 탄력을 유지시키는 기질 단백질인 elastine 활성 저해 효과를 확인한 결과, 2,000 ug/ml의 농도에서 HRW는 38.87%, HRE는 12.49%로 에탄올 추출물보다는 물 추출물에서 더 우수한 tyrosinase 저해율을 보였다.
10. MTT assay를 이용한 세포독성을 측정한 결과, 마우스 대식세포주인 RAW264.7 세포에서 모든 추출물이 세포 독성을 보이지 않았으며, 처리 농도 전 구간에서 세포 생존율이 90% 이상으로 유지되는 것으로 확인되었다
11. 대식세포인 RAW264.7 세포에 LPS(lipopolysaccharide)를 처리하여 생성된 NO의 양을 측정한 결과, HRW는 14~16%의 NO 저해 효과를 나타내었으며, HRE는 9~19%의 NO 생성율이 감소하였다.
화피산 물 추출물(HRW)은 에탄올 추출물(HRE)에 비해 고형분 수율이 높고 환원당 함량이 풍부하고, HRE는 수용성 단백질 함량과 총 폴리페놀 및 총 플라보노이드 화합물 함량이 높게 나타나 추출 용매에 따라 화피산 추출물의 성분 조성이 뚜렷하게 구분됨을 확인하였다. 항산화 활성 평가에서 HRW는 ABTS radical 소거능에서 저농도부터 매우 높은 활성을 나타내었으며, DPPH radical 소거능 또한 HRW와 HRE 모두에서 유의한 효과를 나타내었다. 또한 HRW는 elastase 및 tyrosinase 저해 활성에서 HRE보다 우수한 결과를 나타내었다. RAW264.7 대식세포에 대한 MTT assay 결과, HRW와 HRE 모두 전 농도에서 유의적인 세포 독성을 나타내지 않았으며, LPS로 유도된 RAW264.7 대식세포에서 nitric oxide(NO) 생성을 효과적으로 억제하였다.
이에, 전통적으로 피부의 열을 내리고 가려움증을 완화하는 처방으로 활용되어 온 화피산은 본 연구 결과를 통해 항산화, 항노화 및 항염 기능성을 확인할 수 있었으며, 추출 용매에 따라 실험 결과에 차이를 보이는 것이 확인되었다. 이에 따라 화피산 추출물은 천연 항산화제 및 피부 노화 개선을 목적으로 한 기능성 식품 또는 화장품 소재로의 활용 가능성이 높을 것으로 사료되며, 향후 활성 성분 규명 및 작용 기전에 대한 추가 연구가 이루어진다면 산업적 응용 가치가 더욱 확대될 것으로 판단된다.

Hwapisan (樺皮散) is a traditional herbal prescription documented in classical Korean medical texts, including Donguibogam, and has historically been used to treat skin disorders characterized by pruritus and eruptions, such as Eunjin (癮疹), Pung-eunjin (風癮疹), and urticaria. This prescription is composed of Betulae Cortex (樺皮), Armeniacae Semen (杏仁), Schizonepetae Spica (荊芥), Aurantii Fructus (枳殼), and Glycyrrhizae Radix (甘草) (Her, 2025). In domestic Korean medicine prescription databases, Hwapisan is similarly described as a formula applied to a group of skin conditions characterized by sudden itching and wheal-like eruptions, including Eunjin, Pung-eunjin, and urticaria. Although previous studies have reported antioxidant and anti-inflammatory activities of Betulae Cortex and individual constituent herbs, studies evaluating the biological activities of Hwapisan extracts as a combined herbal prescription are currently limited. Therefore, the present study aimed to investigate the antioxidant, anti-aging, and cytotoxic effects of Hwapisan extracts prepared from Betulae Cortex, Armeniacae Semen, Schizonepetae Spica, Aurantii Fructus, and Glycyrrhizae Radix. The findings of this study are expected to provide fundamental scientific evidence for the physiological activities of mixed herbal extracts and to support the potential utilization of Hwapisan as a functional material andsource ofbioactive compounds.
1. The extraction yields of Hwapisan obtained by reflux extraction using water and ethanol as solvents are presented as solid yields. The water extract of Hwapisan (HRW) exhibited a higher extraction yield of 45.10%, whereas the ethanol extract (HRE) showed a yield of 28.73%, indicating that water extraction was more effective in recovering total solid components from the formula.
2. The reducing sugar contents of the Hwapisan extracts were 332.48 mg/g for the water extract (HRW) and 264.43 mg/g for the ethanol extract (HRE), with HRW showing a higher reducing sugar content than HRE. The content of water-soluble protein was higher in the ethanol extract of Hwapisan (HRE), with a value of 88.05 mg/g, compared to 62.60 mg/g in the water extract (HRW).
3. The total polyphenol content was higher in the ethanol extract of Hwapisan (HRE, 95.00 mg/g) than in the water extract (HRW, 70.39 mg/g). Similarly, the total flavonoid content of HRE (18.82 mg/g) was higher than that of HRW (14.62 mg/g).
4. The ABTS radical scavenging activity of HRW was 85.34% at 125 μg/mL and increased to 104.50% at 2,000 μg/mL. HRE showed a lower scavenging activity of 66.95% at 125 μg/mL compared with HRW; however, its activity increased in a concentration-dependent manner, exceeding 99% at concentrations of 1,000 μg/mL and above.
5. The DPPH radical scavenging activity ranged from 47.76% to 76.43% for HRW and from 67.55% to 75.46% for HRE at concentrations of 125–2,000 μg/mL.
6. The superoxide dismutase (SOD)-like activities of HRW and HRE increased with increasing concentration, and at 2,000 μg/mL, HRE and HRW showed activities of 9.79% and 2.35%, respectively.
7. The nitrite scavenging activities of HRW and HRE were evaluated under different pH conditions. At pH 1.2, the scavenging activities ranged from 19.51% to 94.55% for HRW and from 5.12% to 79.87% for HRE. At pH 3.0, HRW showed scavenging activities of 3.68–22.42%, while HRE exhibited activities of 5.70–30.62%.
8. At a concentration of 2,000 μg/mL, the tyrosinase inhibitory activities were 57.46% for HRW and 47.19% for HRE, with HRW showing higher inhibitory activity than HRE.
9. At a concentration of 2,000 μg/mL, the elastase inhibitory activities were 38.87% for HRW and 12.49% for HRE, with HRW showing higher inhibitory activity than HRE.
10. Cell viability was evaluated using the MTT assay in RAW264.7 cells, and no cytotoxicity was observed for any extract. Cell viability remained above 90% across all tested concentrations.
11. NO production was measured in RAW264.7 cells stimulated with LPS. HRW showed 14–16% inhibition of NO production, while HRE reduced NO production by 9–19%.
As a result, the water extract of Hwapisan (HRW) showed higher extraction yield and reducing sugar content, whereas the ethanol extract (HRE) contained higher levels of water-soluble protein, total polyphenols, and total flavonoids. HRW exhibited strong ABTS radical scavenging activity at low concentrations, and both extracts showed notable DPPH radical scavenging activity. In addition, HRW showed higher inhibitory activities against elastase and tyrosinase. Neither extract exhibited cytotoxicity in RAW264.7 cells, and both suppressed nitric oxide production in LPS-stimulated macrophages. In conclusion, Hwapisan extracts may be considered potential natural sources with antioxidant, anti-aging, and anti-inflammatory activities, and may be applicable as functional ingredients for food or cosmetic products.
