Drynariae Rhizoma (骨碎補), literally meaning “to mend broken bones,” is a traditional herbal medicine widely used in East Asian medicine. In Korea, it is prepared from the rhizome of Drynaria mariesii T. Moore ex Baker, a perennial herb belonging to the family Davalliaceae. In this study, Drynariae Rhizoma was extracted using water and 80% ethanol as solvents through reflux and hot-water extraction methods. The contents of bioactive compounds were determined, and antioxidant activities were evaluated using ABTS and DPPH radical scavenging assays, along with nitrite scavenging activity. In addition, SOD-like activity, xanthine oxidase inhibitory activity, and anti-aging effects were investigated to assess the physiological activities of the extracts.
1. The water-soluble protein contents of the three extracts of Drynariae Rhizoma were highest in the high-pressure hot water extract (HWDR) 127.95 mg/g, followed by the water reflux extract (WDR) 105.28 mg/g and the ethanol reflux extract (EDR) 75.62 mg/g.
2. The reducing sugar contents were highest in the EDR 336.03 mg/g, followed by the HWDR 291.55 mg/g, and the WDR 290.67 mg/g. No significant difference was observed between the HWDR and WDR extracted using water as the solvent.
3. The total flavonoid contents were highest in EDR 9.25 mg/g, followed by HWDR 8.79 mg/g and WDR 5.13 mg/g.
4. The total polyphenol contents were highest in HWDR 121.76 mg/g , followed by WDR 108.28 mg/g and EDR 57.84 mg/g.
5. The ABTS radical scavenging activities of the three extracts measured at concentrations of 62.5∼2,000 ug/mL were 79.21∼98.95% for HWDR, 72.53∼99.46% for WDR, and 57.95–100.67% for EDR, showing higher activities than the positive control, L-ascorbic acid (51.00∼99.31%). In particular, WDR and HWDR extracted using water as the solvent exhibited ABTS radical scavenging activities exceeding 93% even at 125 ug/mL.
6. The DPPH radical scavenging activities at 2,000 ug/mL ranged from 77.13 to 87.47%, with WDR exhibiting the highest scavenging activity among the extracts.
7. The nitrite scavenging activities at 2,000 ug/mL under pH 1.2 were 96.91% for WDR, 92.65% for HWDR, and 81.89% for EDR. At pH 3.0, the activities were 96.95% for WDR, 79.61% for HWDR, and 69.17% for EDR. Notably, WDR showed no significant difference in nitrite scavenging activity between pH 1.2 and pH 3.0.
8. The SOD-like activities at 2,000 ug/mL ranged from 10.28 to 18.10%, with HWDR exhibiting the highest activity among the three extracts.
9. The xanthine oxidase (XO) inhibitory activities at 2,000 ug/mL were 44.87% for WDR, 39.68% for EDR, and 17.76% for HWDR.
10. The tyrosinase inhibitory activities at 2,000 ug/mL were highest in EDR 34.00% > WDR 26.03% > HWDR 22.89%.
11. The elastase inhibitory activities, representing anti-wrinkle effects, indicated that HWDR showed the highest inhibition 30.90% at 2,000 ug/mL, while WDR and EDR exhibited less than 10% inhibitory activity at the same concentration.
Based on these results, Drynariae Rhizoma, traditionally used in Korean medicine and folk remedies as a bone-strengthening herbal material, exhibited DPPH radical scavenging activity comparable to that of the natural antioxidant L-ascorbic acid, confirming its strong antioxidant potential. In addition, the extracts showed excellent nitrite scavenging activity. In particular, the water reflux extract (WDR) demonstrated minimal variation in nitrite scavenging activity depending on pH, suggesting its potential applicability as a food additive under low-acidic to near-neutral conditions.
Therefore, Drynariae Rhizoma, traditionally used as a bone-strengthening herbal medicine in Korean medicine and folk remedies, demonstrates strong antioxidant and nitrite scavenging activities, along with anti-inflammatory, whitening, and anti-wrinkle effects, suggesting its potential application as a natural functional ingredient in the food industry for health promotion, as well as a promising natural resource for functional cosmetic and inner beauty products.

골쇄보(骨碎補; Drynariae Rhizoma)는 “부서진 뼈를 붙인다’ 하여 명명된 한방생약재로 우리나라에서는 넉줄고사리과(Davalliaceae)의 다년생 초본식물인 넉줄고사리(D. mariesii T. Moore ex Baker)의 뿌리줄기를 골쇄보로 사용한다. 이에 골쇄보를 물과 80% 에탄올을 용매로 환류 추출 및 열수 추출하여 각 추출물에 대한 유용성분의 함량과 ABTS 및 DPPH 시약을 이용한 free radical 소거능과 아질산염 소거 활성을 측정하고, SOD 유사 활성, xanthine oxidase 저해 및 항노화 효과를 확인하였다.
1. 골쇄보 3가지 추출물의 수용성 단백질은 고압 열수 추출물(HWDR) 127.95mg/g>환류 물 추출물(WDR) 105.28mg/g>환류 에탄올 추출물(EDR) 75.62mg/g을 함유하였다.
2. 환원당은 EDR 336.03mg/g>HWDR 291.55mg/g>WDR 290.67mg/g으로 물을 용매로 추출한 HWDR과 WDR은 유의적 차이가 없었다.
3. 플라보노이드 화합물 함량은 EDR 9.25mg/g>HWDR 8.79mg/g>WDR 5.13mg/g을 함유하였다.
4. 폴리페놀 화합물 총 함량은 HWDR 121.76mg/g>WDR 108.28mg/g>EDR 57.84mg/g을 함유하였다.

6. DPPH radical 소거 활성은 2,000ug/mL에서 77.13∼87.47%로 WDR에서 가장 높은 소거효과를 나타내었다.
7. 아질산염 소거 활성을 측정한 결과, pH 1.2의 2,000ug/mL에서 WDR 96.91%>HWDR 92.65%>EDR 81.89%의 활성을 나타내었으며, pH 3.0에서는 WDR 96.95%>HWDR 79.61%>EDR 69.17%로 WDR은 pH 1.2와 3.0에서의 아질산염 소거효과에 차이가 없었다.
8. SOD 유사 활성은 2,000ug/mL에서 10.28∼18.10%로 골쇄보 3가지 추출물중 HWDR에서 가장 높은 활성을 나타내었다.
9. Xanthine oxidase(XO) 저해효과는 2,000ug/mL에서 WDR 44.87%, EDR 39.68%, HWDR 17.76%의 활성을 나타내었다.
10. Tyrosinase 저해 활성은 2,000ug/mL에서 EDR 34.00%>WDR 26.03%>HWER 22.89%의 순으로 나타났다.
11. 주름개선 효과를 elastase 저해 활성을 측정한 결과, 2,000ug/mL에서 HWDR은 30.90%로 가장 우수한 저해 효과를 나타내었으나 WDR과 EDR은 10% 미만의 저해효과를 나타내었다.
이상의 결과, 한방과 민간에서 뼈에 좋은 한방생약재로 사용되는 넉줄고사리 뿌리, 골쇄보는 천연항산화제인 L-ascorbic acid와 유사한 DPPH radical 소거 활성을 나타내어 우수한 항산화 효과가 있음을 확인하였으며, 아질산염 소거효과도 우수하였다. 특히 환류 물 추출물은 pH에 따른 변화가 적어 산성도가 낮거나 중성에 가까운 식품 첨가제로 활용 가능성이 있음을 확인할수 있었다. 또한 항염증, 미백, 주름개선 효과를 확인하여 한방생약재 이외에 건강 증진을 위한 식품산업에서 기능성 원료로 활용 가능성이 있으며, 미백, 주름개선을 위한 기능성 화장품 원료나 이너뷰티 소재로서도 활용 가능한 천연유래 한방생약재인 것으로 판단된다.
