갯무(Raphanus sativus) 추출물의 생리활성 효능 및 화장품 기능성 원료로서의 활용성 평가
갯무(Raphanus sativus var. hortensis f. raphanistroides Makino)는 해안 지역에 자생하는 십자화과 식물로, 높은 염분, 강한 바람 등 가혹한 환경에서도 생존하는 강한 내성 식물이다. 갯무는 이러한 척박한 환경에 적응하기 위해 다양한 2차 대사산물을 생성하며, 이는 항산화, 항염, 항암 등 다양한 효과를 나타내는 것으로 보고되었다. 그러나 피부미용학적 관 점에서 갯무의 생리 활성 물질을 탐색한 연구는 미비하다. 따라서 본 연 구에서 갯무의 항산화, 항염, 항주름, 미백 및 수렴 효능을 과학적으로 규 명하고, 이를 기반으로 화장품 기능성 원료로서의 활용성을 평가하고자 하였다.
갯무 꽃, 뿌리 및 잎 부위를 동결건조 후 분말화하여 80% 메탄올로 추출하였다. 각 추출물의 항산화 활성은 DPPH 및 ABTS 라디컬 소거능 과 총 폴리페놀 함량(TPC)을 측정하여 평가하였다. 항염 효능은 RAW264.7 세포에 LPS로 염증을 유도한 후 NO 생성 억제, TNF-α, IL-1β, IL-6 사이토카인 생성 억제, iNOS 및 COX-2 단백질 발현 억제를 측정하였다. 항주름 효능은 elastase 저해활성을 통해 평가하였고, 항멜라 닌 효능은 tyrosinase 및 DOPA 산화 저해 활성을 통해 검증하였다. 수렴 효능은 astringent assay를 이용하여 평가하였다.
갯무 추출물의 모든 부위에서 DPPH 및 ABTS라디컬 소거능과 TPC 가 농도 의존적으로 증가하였으며, 특히, 꽃 추출물이 가장 우수한 활성을 나타냈다. 세포 생존율은 RAW 264.7, NHDF 및 HaCaT 세포 모두에서 대부분 97%의 세포 생존율을 보여 이후 효능 평가에서 안전하게 사용될 수 있었다. 항염 효능에서는 꽃 추출물이 NO 생성과 iNOS 및 COX-2 단 백질 발현 억제에서 가장 높은 활성을 보였으며, 염증성 사이토카인 억제 에서는 잎 추출물이 꽃 추출물과 유사하거나 더 우수한 활성을 나타냈다. 항주름 효능에서는 저농도에서 꽃 추출물이 우수하였으나, 고농도에서는 뿌리 추출물이 가장 높은 활성을 보였다. 항멜라닌 효능에서는 꽃 추출물 이 tyrosinase 및 DOPA 산화 활성을 가장 높게 억제하였으며, 수렴 효능 또한 꽃 추출물이 가장 우수하게 나타났다.
본 연구를 통해 갯무 꽃, 잎 및 뿌리 추출물이 항산화, 항염, 항주름, 항멜라닌, 수렴 등 다양한 생리활성을 나타내는 것을 확인하였다. 특히 꽃 추출물은 전반적인 실험에서 가장 우수한 활성을 보여, 기능성 화장품 원 료로서의 활용 가치가 높을 것으로 판단된다. 잎 및 뿌리 추출물 또한 각 각 특정 효능에서 뛰어난 활성을 보여, 갯무 추출물이 천연 소재로서 활 용 가능성이 높음을 시사한다. 본 연구는 갯무의 피부미용 및 뷰티 산업 응용 가능성을 과학적으로 입증한 최초의 연구라는 점에서 학문적‧산업적 의의가 크다고 사료된다.

Raphanus sativus var. hortensis f. raphanistroides Makino, commonly known as Gaetmu, is a cruciferous plant native to coastal regions characterized by strong tolerance to harsh environmental conditions, such as high salinity and strong winds. To survive in such extreme habitats, Gaetmu produces diverse secondary metabolites reported to exhibit antioxidant, anti-inflammatory, and anticancer activities. However, studies exploring its bioactive compounds from a cosmetic perspective remain insufficient. Therefore, this study aimed to scientifically elucidate the antioxidant, anti-inflammatory, anti-wrinkle, anti-melanogenic, and astringent activities of Gaetmu extracts and evaluate their potential as functional cosmetic ingredients.
The flowers, roots, and leaves of Gaetmu were freeze-dried, powdered, and extracted with 80% methanol. Antioxidant activity was assessed by measuring DPPH and ABTS radical-scavenging capacities and total phenolic content(TPC). Anti-inflammatory effects were evaluated in LPS-stimulated RAW264.7 macrophages by measuring nitric oxide (NO) production, suppression of pro-inflammatory cytokines(TNF-α, IL-1β, and IL-6), and inhibition of iNOS and COX-2 protein expression. Anti-wrinkle activity was determined via elastase inhibition, while anti-melanogenic activity was assessed by tyrosinase and DOPA oxidation inhibitory assays. Finally, the astringent potential of the extracts was examined using an astringent assay.
All Gaetmu extracts exhibited dose-dependent increases in DPPH and ABTS radical-scavenging activities and TPC, with the flower extract showing the strongest antioxidant activity. Cell viability exceeded 97% in RAW264.7, NHDF, and HaCaT cells, confirming the non-cytotoxicity of the extracts for subsequent efficacy experiments. Regarding anti-inflammatory effects, the flower extract most effectively suppressed NO production and iNOS and COX-2 expression, while the leaf extract demonstrated comparable or superior inhibition of inflammatory cytokines compared to the flower extract. In anti-wrinkle assays, the flower extract showed strong elastase inhibition at low concentrations, whereas the root extract exhibited the highest activity at higher concentrations. For anti-melanogenic effects, the flower extract most strongly inhibited tyrosinase and DOPA oxidation and demonstrated the highest astringent activity.
Overall, the flower, leaf, and root extracts of Gaetmu displayed diverse biological activities, including antioxidant, anti-inflammatory, anti-wrinkle, anti-melanogenic, and astringent effects. Notably, the flower extract consistently exhibited the highest efficacy across most assays, indicating its strong potential as a functional cosmetic ingredient. The leaf and root extracts also showed notable potency in specific activities, highlighting the value of Gaetmu as a natural bioactive resource. This study is the first to scientifically demonstrate the potential applications of Gaetmu in the skin care and beauty industry, providing significant academic and industrial insights.
