최근 제약산업에서는 인공지능(AI), 클라우드(Cloud), 빅데이터(Big Data) 등의 기술이 의약품 제조 및 품질관리 영역의 주요 전산시스템(LIMS, QMS, WMS, ELN, EDMS 등)에 빠르게 도입되고 있다. 이에 따라 시스템의 신뢰성 및 데이터 완전성(Data Integrity)을 확보하기 위한 컴퓨터 시스템 밸리데이션(Computerized System Validation, CSV)의 중요성이 더욱 강조되고 있다. 그러나 이러한 기술적 고도화는 전통적인 문서 중심의 밸리데이션 접근 방식으로는 복잡한 알고리즘과 비정형 데이터 흐름을 충분히 검증하기 어렵다는 한계를 드러내고 있다. 본 연구는 AI 기반 전산시스템이 도입된 제약산업 환경을 대상으로, GAMP5(2nd Edition)와 미국 식품의약청(FDA)의 Computer Software Assurance(CSA) 가이드라인을 중심으로 최신 CSV 수행 방안을 체계적으로 분석하고자 하였다. 연구 방법으로는 문헌 연구(Literature Review)와 국제 규제 비교 분석을 병행하였으며, EudraLex Volume 4 Annex 11, 21 CFR Part 11, PIC/S PI-011 등 주요 글로벌 규제 문헌을 참조하였다. 또한 Validation Life Cycle을 구성하는 IA(Impact Assessment), VP(Validation Plan), RA(Risk Assessment), URS(User Requirement Specification), DQ(Design Qualification), IQ(Installation Qualification), OQ(Operational Qualification), PQ(Performance Qualification), VSR(Validation Summary Report) 각 단계의 상호 연계성을 GAMP5의 V-Model을 기반으로 분석하였다. 연구 결과, AI 기반 전산시스템의 밸리데이션 수행 시에는 기존 CSV Life Cycle을 단순히 적용하기보다, 위험 기반 접근(Risk-based Approach)을 중심으로 한 테스트 자동화, 요구사항 기반 검증, 문서화 효율화가 필수적임을 확인하였다. 특히 CSA 접근법은 기존의 문서 중심 CSV 절차를 보완하며, 실제 시스템의 위험 수준에 따라 품질보증(Quality Assurance) 활동을 최적화할 수 있는 합리적 대안으로 평가되었다.
따라서 본 연구는 제약산업의 AI 기반 전산시스템 환경에 적합한 CSV 수행 프레임워크를 제안함으로써, 향후 GxP 전산시스템의 합리적 밸리데이션 수행 체계 구축 및 규제 대응 전략 수립에 기여 할 수 있을 것으로 기대된다.

In the pharmaceutical industry, emerging technologies such as Artificial Intelligence (AI), Cloud Computing, and Big Data have been widely implemented in core computerized systems, including LIMS, QMS, WMS, ELN, and EDMS. These advancements enhance data integrity and quality assurance; however, they also expose the limitations of conventional document-centric Computerized System Validation (CSV) approaches in verifying the reliability of complex, within regulated environments. This study aims to propose an optimized CSV framework suitable for AI-driven computerized environments within the pharmaceutical sector, focusing on the principles of GAMP5 (2nd Edition) and the FDA’s Computer Software Assurance (CSA). The research methodology combines literature review and comparative analysis of global regulatory frameworks, including EudraLex Volume 4 Annex 11, 21 CFR Part 11, and PIC/S PI-011. The interrelationships among validation life cycle stages—IA (Impact Assessment), VP (Validation Plan), RA (Risk Assessment), URS (User Requirement Specification), DQ (Design Qualification), IQ (Installation Qualification), OQ (Operational Qualification), PQ (Performance Qualification), and VSR (Validation Summary Report)—were systematically analyzed and structured based on the GAMP5 V-Model. The findings indicate that, in AI-based computerized environments, an optimized CSV approach requires shifting from traditional procedural validation toward a risk-based methodology emphasizing automated testing, requirement-driven verification, and efficient documentation practices. In particular, the CSA framework serves as a practical alternative that mitigates documentation inefficiencies and optimizes assurance activities according to actual system risk levels. This research proposes an integrated optimization model for CSV applicable to AI-driven pharmaceutical information systems, contributing to the advancement of quality assurance and regulatory compliance in future GxP computerized environments.
