식품 매개 병원성 미생물에 의한 오염은 생산·가공·유통 환경에서 지속적인 위생 문제를 일으키며 특히 식품 접촉 표면에 형성되는 biofilm은 미생물의 생존력과 소독제 내성을 높여 식품 안전 관리에 큰 어려움을 초래한다. 특히 Salmonella spp.는 전 세계 식중독의 주요 원인균으로 보고되고 있으며 다양한 환경과 식품에서 검출되는 대표적인 병원균으로 알려져 있다. 이 중 Salmonella enterica serovar Typhimurium는 강한 병원성을 가진 식중독 세균으로 식품 생산 및 가공환경에서 지속적으로 검출되고 있다. Salmonella enterica Schwarzengrund는 최근 여러 지역에서 검출 빈도가 증가하고 있는 혈청형으로 가금류·축산물 및 다양한 식품에서 반복적으로 검출되고 있으며 항균제 내성 보유 사례가 증가하면서 잠재적 공중보건 위험성이 더욱 강조되고 있다. 식품 접촉 표면에 형성되는 Salmonella biofilm을 효과적으로 제어하기 위해서는 표면 재질에 따른 부착 특성과 혈청형 간 biofilm 형성 능력의 차이를 이해하는 것이 중요하다. 그러나 stainless steel과 polypropylene과 같은 식품 산업 표면에서 표준균주와 환경 분리 균주의 biofilm 특성을 비교하고 소독제 종류 및 처리조건에 따른 단계별 저감 효과를 분석한 연구는 제한적이다. 따라서 본 연구에서는 표준균주 S. Typhimurium과 환경 분리 균주 S. Schwarzengrund의 biofilm 형성 특성을 stainless steel(SS) 및 polypropylene(PP) 표면에서 비교하고, 형성된 biofilm에 대한 sodium hypochlorite과 aqueous chlorine dioxide의 저감 효과를 확인하고자 하였다. 두 균주의 biofilm 양은 배양 시간이 증가함에 따라 SS와 PP표면 모두에서 유의적으로 증가하였다. SS표면에서 3 h에서 3.62–3.95, 12 h에서 7.13-7.14, 48 h에서 7.66–7.67 log CFU/cm² 범위로 나타났으며 PP 표면에서도 3 h 3.42–3.44, 12 h 6.96-7.09, 48 h 7.37–7.64 log CFU/cm² 범위로 나타나 두 균주 모두 SS와 PP표면에서 시간대별 유사한 증가를 보였다. Calcofluor White 염색결과, 두 균주의 SS와 PP 표면 모두에서 배양 시간에 따라 EPS가 증가하였으며 특히 6–12 h 구간에서 증가폭이 가장 크게 나타나 중기 단계에서의 급격한 EPS 축적이 확인되었다. EPS 증가 양상을 바탕으로 3h, 12h, 48h을 각각 초기·중기·성숙 단계로 설정하여 두 소독제의 저감 효과를 평가하였다. Sodium hypochlorite 처리 결과, biofilm의 발달 단계에 따라 저감 효과는 다른 것을 확인하였다. 초기 단계에서는 SS와 PP 모두에서 1–2 log 감소가 나타났으며, 중기 단계에서는 20–100 mg/L 처리 시 3–6 log의 감소가 확인되었다. 반면 성숙 단계에서는 100 mg/L 처리에서도 3–5 log 감소에 그쳐 높은 내성을 보였으며 EPS 잔존량 또한 증가하여 PP에서 더 낮은 수준을 보였다. Aqueous chlorine dioxide 처리 결과 역시 농도와 시간 증가에 따라 저감 효과가 높게 나타났으며, 특히 초기·중기 단계에서 sodium hypochlorite보다 더 빠르고 일관된 저감 효과를 나타냈다. 두 소독제 모두 50–100 mg/L에서는 SS와 PP에서 약 3–5 log 수준의 감소가 나타났으며, 초기 단계에서는 ClO₂가 더 높은저감 효과를 보였다. 이러한 결과를 바탕으로 stainless steel과 polypropylene 표면에 형성된 Salmonella Schwarzengrund biofilm의 단계별 제어를 위해 sodium hypochlorite과 aqueous chlorine dioxide의 적용 가능성을 평가하였고 두 소독제 모두 고농도에서는 효과적인 biofilm 감소를 보였으며 aqueous chlorine dioxide은 초기·중기 단계에서 높은 저감 효과를 나타내 식품 가공환경의 위생 관리에 활용될 수 있음을 확인하였다.

Contamination by foodborne pathogenic microorganisms causes persistent hygiene problems throughout food production, processing, and distribution environments, and particularly, biofilms formed on food contact surfaces enhance microbial survival and sanitizer resistance, creating major challenges in food safety management. Among these pathogens, Salmonella spp. are reported as leading causative agents of foodborne illness worldwide, and are known to be commonly detected in various environments and foods. Salmonella enterica serovar Typhimurium is a highly pathogenic foodborne bacterium that is continuously detected in food production and processing environments. Salmonella enterica serovar Schwarzengrund has shown increasing detection frequencies in recent years across various regions, and has been repeatedly isolated from poultry, livestock products, and diverse foods, with rising reports of antimicrobial resistance, thereby highlighting its potential public health risk. To effectively control Salmonella biofilms formed on food contact surfaces, it is important to understand the differences in attachment properties by surface material and the biofilm-forming abilities among serotypes. However, studies comparing the biofilm characteristics of standard strains and environmental isolates on food industry surfaces such as stainless steel and polypropylene, and analyzing stage-specific reduction effects according to sanitizer type and treatment conditions, remain limited. Therefore, this study compared the biofilm formation characteristics of the standard strain S. Typhimurium and the environmental isolate S. Schwarzengrund on stainless steel (SS) and polypropylene (PP) surfaces, and evaluated the reduction effects of sodium hypochlorite and aqueous chlorine dioxide on the formed biofilms. Biofilm amounts of both strains significantly increased over time on SS and PP surfaces. On SS, biofilm levels ranged from 3.62–3.95 log CFU/cm² at 3 h, 7.13–7.14 at 12 h, and 7.66–7.67 at 48 h, and similarly on PP, 3.42–3.44 at 3 h, 6.96–7.09 at 12 h, and 7.37–7.64 at 48 h, showing comparable time-dependent increases for both strains on both surfaces. Calcofluor White staining showed that EPS levels increased with incubation time for both strains on SS and PP surfaces, with the largest increase observed between 6 and 12 h, indicating rapid EPS accumulation during the mid-stage of biofilm development. Based on EPS patterns, 3 h, 12 h, and 48 h were designated as the early, intermediate, and mature stages of biofilm development, respectively, to evaluate the reduction effects of the two sanitizers. Sodium hypochlorite treatment showed that reduction effects differed according to biofilm developmental stage. In early-stage biofilms (3 h), 1–2 log reductions were observed on both SS and PP. In intermediate-stage biofilms (12 h), 20–100 mg/L treatments produced 3–6 log reductions. In mature biofilms (48 h), even 100 mg/L resulted in only 3–5 log reductions, showing high resistance; EPS residues also increased over time, with lower levels observed on PP than on SS. Chlorine dioxide treatment also showed increased reduction with higher concentrations and longer exposure times and demonstrated faster and more consistent reductions than sodium hypochlorite in early and intermediate stages. At 50–100 mg/L, both sanitizers produced approximately 3–5 log reductions on SS and PP, and chlorine dioxide showed greater reduction in the early stage. Based on these results, the applicability of sodium hypochlorite and aqueous chlorine dioxide for stage-specific control of Salmonella biofilms formed on stainless steel and polypropylene surfaces was evaluated, and both sanitizers showed effective biofilm reduction at high concentrations, while chlorine dioxide demonstrated superior reductions in early and intermediate stages, indicating its potential usefulness for hygiene management in food processing environments.
