알츠하이머병 (Alzheimer’s disease, AD)은 치매의 약 80%를 차지하는 대표적인 만성 신경퇴행성 질환으로, 주로 고령층에서 발병률이 높다. 알츠하이머병의 주요 병리적 원인으로는 아밀로이드 전구체 단백질 (amyloid precursor protein, APP)이 βsecretase와 γ-secretase에 의해 연속적으로 절단되어 생성된 아밀로이드 베타 (amyloid beta, Aβ) 펩타이드의 과도한 응집과 이로 인한 신경독성이 지목된다. 그러나 현재까지 승인된 치료제는 소수에 불과하며, 이마저도 일부 중증 환자의 증상 완화에 그칠 뿐 질병의 진행을 근원적으로 억제하는 치료제는 부재한 실정이다. 이에 따라 부작용이 적으면서도 예방과 치료가 가능한 새로운 기전의 소재 발굴이 시급하다. 최근 장내 미생물 군집 (gut microbiota)이 장-뇌 축 (gut-brain axis, GBA)을 통해 뇌의 기능 및 신경 퇴행성 질환의 병리에 영향을 미친다는 연구 결과가 보고됨에 따라, 프로바이오틱스 (probiotics) 섭취가 알츠하이머병을 포함한 신경 질환의 예방 및 치료에 핵심적인 역할을 할 수 있다는 가능성이 대두되고 있다. 이에 본 연구에서는 아밀로이드 베타의 과발현을 유도하여 알츠하이머병 병리를 모사한 예쁜꼬마선충 (Caenorhabditis elegans) 모델을 이용하여 프로바이오틱스의 신경보호 효능과 작용 기전을 규명하고자 하였다.연구 수행 과정은 단계적인 스크리닝 방식을 채택하였다. 첫째, 총 33종의 프로바이오틱스 균주를 대상으로 마비 (paralysis) 지연 효과를 확인하여 항알츠하이머 효능을 가진 잠재적 후보군 5종 (LAB #9, #13, #14, #15, #29)을 1차 선별하였다. 둘째, 선별된 5종의 균주를 대상으로 행동학적 분석 (먹이 탐식 능력 및 주화성 분석)과 형광 현미경을 이용한 생체 내 Aβ 응집 억제 효능 평가를 수행하였다. 그 결과, 주화성 지수를 유의하게 향상시키고 응집체 감소에서 가장 우수한 경향성을 보인 LAB #15를 최종 후보 균주로 선정하였다. 마지막으로, 최종 선별된 LAB #15가 Aβ 독성 환경에서 수명을 유의하게 연장시킴을 확인하였으며, 실시간 중합효소연쇄반응 (RT-qPCR)을 통해 핵심 전사 인자인 daf-16과 항산화 유전자 gst-4의 발현을 상향 조절함으로써 신경 보호 효과를 나타냄을 규명하였다. 본 연구 결과는 프로바이오틱스가 알츠하이머병 예방 및 치료를 위한 기능성 소재로서 활용될 수 있는 과학적 근거를 제시한다.

Alzheimer’s disease (AD) is a representative chronic neurodegenerative disorder accounting for approximately 80% of all dementia cases, with a high incidence rate particularly among the elderly. The primary pathological cause of AD is attributed to the excessive aggregation and subsequent neurotoxicity of amyloid-beta (Aβ) peptides, which are generated through the cleavage of amyloid precursor protein (APP). Recently, as research has shown that the gut microbiota influences brain function via the gut-brain axis (GBA), the consumption of probiotics has emerged as a new alternative for the prevention and treatment of AD. Therefore, this study aimed to investigate the neuroprotective efficacy and mechanisms of action of probiotics using a Caenorhabditis elegans model that mimics AD pathology through Aβ overexpression. A stepwise screening strategy was employed for the research process. First, primary screening was conducted on a total of 33 probiotic strains to identify potential candidates with anti-Alzheimer’s efficacy by evaluating their effects on delaying Aβ-induced paralysis, leading to the selection of five superior candidates (LAB #9, #13, #14, #15, and #29). Second, the selected five strains were subjected to behavioral analyses, such as food race and chemotaxis assays, as well as an assessment of their in vivo inhibitory effects on Aβ aggregation using fluorescence microscopy. Consequently, LAB #15 was selected as the final candidate strain, as it significantly improved the chemotaxis index and exhibited the most superior trend in reducing aggregates. Finally, it was confirmed that the selected LAB #15 significantly extended the lifespan of the C. elegans model under Aβ toxic conditions. Furthermore, through real-time quantitative PCR (RT-qPCR), it was elucidated that LAB #15 exerts neuroprotective effects by upregulating the expression of the key transcription factor daf-16 and the antioxidant gene gst-4. The results of this study provide scientific evidence that selected probiotics can be utilized as functional materials for the prevention and treatment of Alzheimer's disease in the future.
