트리칼슘 실리케이트은 칼슘 실리케이트계 생체재료로 치과 및 정형외과 분야에서 생체적합성과 생체활성이 우수하여 골대체재의 가능성을 주목받고 있다. 특히 트리칼슘 실리케이트은 MTA 의 주요 성분으로서 우수한 밀폐성과 조직 친화적 특성으로 널리 사용되어 왔다. 그러나 수화 과정에서의 다량의 칼슘 이온과 수산화 이온을 방출하여 높은 알칼리 환경(pH > 12)을 형성하는데, 이는 항균 효과를 가지지만, 한편 세포 독성을 유발할 수 있다는 단점이 있다. 기존 연구들은 트리칼슘 실리케이트의 수화물이 인산을 포함한 용액과 접촉할 경우 표면에 수산화인회석 혹은 칼슘결핍 수산화인회석이 형성될 수 있음을 보고하였으나, 대부분 연구가 수화과정 후 생성된 결정과의 상호작용에 국한되어 있었다. 이에 본 연구는 트리칼슘 실리케이트의 수화 과정에서 직접 인산 용액을 도입함으로써 결정상 변화, 용액 화학적 특성, 세포 수준에서의 초기 골분화 반응을 종합적으로 평가하였다.
연구에서는 트리칼슘 실리케이트과 인산이칼륨 용액을 사용하였으며, 트리칼슘 실리케이트 분말과 인산이칼륨 용액을 혼합하여 시편을 준비하였다. 준비된 시편은 수화 및 모의 체액 침적 후, X 선 회절법, 주사전자현미경, 유도 결합 플라즈마 분광법, 그리고 pH 측정을 통해 결정상 변화, 경화 시간, 압축 강도, pH 차이 및 이온 방출 양상을 분석하였다. 또한 세포 수준에서의 반응을 확인하기 위하여 실시간 중합효소 연쇄 반응을 통해 골분화 관련 유전자 발현 평가를 수행하였고, 미세 유체 기반 시스템을 이용하여 관류 조건에서의 세포 생존력 및 이동 양상을 관찰하였다.
실험 결과, 인산 용액을 첨가한 경우 트리칼슘 실리케이트은 수화 과정에서 칼슘 결핍 수산화인회석의 전환이 이뤄졌으며, 증류수 조건과 비교하여 뚜렷한 결정상 차이를 나타냈다. X 선 회절법 및 주사전자현미경 분석에서는 침적 시간이 증가할수록 칼슘 결핍 수산화인회석 결정 피크 강도가 뚜렷하게 증가하였다. 또한 인산 함유량이 증가할 수록 경화 시간 및 용해도가 증가하였지만, 압축강도는 낮아지는 결과를 확인할 수 있었다. 유도 결합 플라즈마 분광법 분석 결과에서는 인산 함량이 높을수록 칼슘 및 인의 방출은 억제되었으나 실리콘 방출은 증가하였다. 특히 TP10, TP15, TP20 군에서는 용액 내 칼슘 농도가 2–6 mM 범위에 유지되었는데, 이는 선행 연구에서 보고된 골아세포의 분화 및 성장에 유리한 범위와 일치하였다. 또한 pH
측정 결과, 인산 함량이 증가할수록 알칼리성 완화 경향을 보였으며, 이는 칼슘 실리케이트 하이드레이트 형성 지연 및 실리콘 방출 증가와 연관된 것으로 해석된다.
세포 실험에서 TP15, TP20 은 TP0 에 비해 골분화 관련 초기 전사인자(RUNX2, OSX, COL1A1)의 발현이 증가하고 중기 마커인 OPN 의 발현은 차이가 없었다. 이는 인의 도입이 세포 독성을 완화하고 초기 골분화 방향으로 환경을 조성함을 시사한다. 더 나아가 미세 유체칩 기반 관류 실험에서는 TP20 에서 세포가 시편 표면 방향으로 뚜렷하게 이동하는 양상이 확인되었으며, 이는 정적 배양에서는 확인되지 않았던 현상으로, 인이 풍부한 조성이 관류 환경에서 세포 친화성을 높여 빠른 계면 피복을 유도할 수 있음을 보여준다.
결론적으로, 본 연구는 트리칼슘 실리케이트 수화 과정에 인산 용액을 직접 도입하는 것이 결정상 전환을 촉진하고, 용액 내 이온 농도와 pH 를 조절하며, 초기 세포 반응을 개선하는 효과적인 방법임을 입증하였다. 이러한 결과는 인산이 도입된 트리칼슘 실리케이트이 골이식재로서 임상적 요구에 따라 빠른 통합에서 장기적 안정성까지 조절 가능한 잠재력을 지니고 있음을 보여준다. 또한 본 연구에서 활용한 미세 유체칩은 생리학적으로 유사한 흐름 조건에서 세포–재료 간 동적 상호작용을 분석할 수 있는 새로운 도구로서, 향후 생체재료 평가에 중요한 역할을 할 수 있음을 확인하였다.

Tricalcium silicate (TCS) is a calcium silicate-based biomaterial. TCS has been studied in the field of dental and orthopedic surgery. TCS had possibility of bone marrow because of its excellent bioactivity. In particular, TCS has been widely used as a major component of mineral trioxide aggregates (MTA). TCS had an excellent strength and antibacterial properties. During the hydration process, a large amount of calcium ions and hydroxide ions are released to form a high alkaline environment (pH>12), which has an antibacterial effect and has the disadvantage of causing toxicity to cells. Existing studies have reported that hydroxyapatite (HAp) or calcium-deficient hydroxyapatite (CDHAp) may be formed on the surface when a hydrate of TCS comes into contact with a solution containing phosphate ion, but most of them were limited to interactions after hydration was completed. Therefore, this study to verify comprehensively evaluating the crystal phase change, solution chemical properties, and initial bone differentiation reaction at the cell level by directly introducing a phosphate solution during the hydration process of TCS.
Hence, this study is to investigate the effect of phosphate-assisted hydration on the mineralization behavior and biological potential of TCS. Specifically, this study examines how phosphate-containing solutions influence calcium ion release, CDHAp formation, and the physicochemical environment during hydration, with the goal of enhancing the materials suitability.
Commercial TCS and potassium dihydrogen phosphate (KH2PO4) were purchased and used without further modification. The phosphate solution was prepared by dissolving KH2PO4 in deionized water. Solutions were formulated at weight ratios (w/w) of KH2PO4 to deionized water as 0:100, 5:95, 10:90, 15:85, and 20:80. The phosphate solution was then mixed with TCS at a liquid- to-powder ratio of 0.12 mL per 300 mg. To confirm CDHAp formation, the mixtures were soaked in simulated body fluid (SBF) for 7, 14, 21, and 28 days, followed by characterization using X-ray diffraction (XRD). And after soaked in SBF for 28 days specimens were conducted X-ray photoelectron spectroscopy (XPS), and scanning electron microscopy (SEM). pH was measured after 7 days of immersion in SBF. Inductively coupled plasma optical emission spectrometer (ICP- OES) was used to confirmed the ion released of TCS specimens. To evaluate osteogenic potential, reverse transcription quantitative polymerase chain reaction (RT-qPCR) was conducted, and cell compatibility was assessed using a lab-on-a-chip model. The results of each test were statistically analyzed with one-way analysis of variance (ANOVA) followed by Tukey’s post hoc statistical test (p < 0.05).
By the XRD and XPS analysis CDHAp was confirmed. The intensity of CDHAp peaks increased with longer immersion times in SBF, indicating progressive mineralization. After soaking in SBF for 28 days, SEM revealed characteristic crystalline morphologies of CDHAp. ICP-OES analysis showed that higher phosphate concentrations led to a reduction in calcium and phosphate ion release, while silicon ion release increased. Especially, in the 10-20 wt% phosphate groups, the calcium ion concentration remained within the optimal range for cellular differentiation (2-6 mM), which correlated with enhanced expression of early osteogenic markers as confirmed by RT-qPCR analysis. pH measurements revealed a decrease in alkalinity with increasing phosphate content. The phosphate solution appeared to affect the hydration process of TCS, delayed formation of calcium silicate hydrate (C-S-H). That could increase silicon ion release. Less alkalinity environment not only favored CDHAp growth but also provided cell friendly conditions for osteoblast activity. Cell compatibility and distribution was confirmed by lab-on-a-chip model. That demonstrated cells extended and migrated along to the specimens.
In conclusion, the incorporation of phosphate solution into TCS promoted the formation of CDHAp and improved biological performance. Phosphate-assisted hydration regulated calcium ion release within a favorable range for osteogenesis, increased silicon ion release, and created a mildly alkaline environment that supported mineralization and cell activity. These findings suggest that phosphate-assisted hydration of tricalcium silicate enhances hydroxyapatite formation, effectively enhance the physicochemical and biological properties.
