Powdery Mildew (PM), primarily caused by Podosphaera xanthii, is one of the most destructive fungal diseases in cucumbers (Cucumis sativus L.) (Wang et al., 2024; Ayaz et al., 2023). The disease is characterized by white, powder-like lesions on leaf surfaces, which reduce photosynthetic activity, weaken plant vitality, and ultimately lead to significant yield and quality losses (Cui et al., 2022; Cohen et al., 2004; Chen et al., 2021). Due to its rapid spread and severe impact on cucumber production worldwide, PM has long been recognized as an economically important disease in agriculture (Pérez-García et al., 2008). Traditional management strategies rely heavily on chemical fungicides; however, their long-term effectiveness is limited by the development of fungicide resistance, environmental concerns, and regulatory restrictions on chemical usage(Corkley et al., 2022; Riseh et al., 2024). As a result, the search for sustainable and eco-friendly alternatives has become an urgent priority.

In recent years, beneficial microbes have attracted attention as biological control agents capable of reducing plant diseases and promoting crop health(M. Yang et al., 2021). Among them, actinobacteria, particularly the genus Pseudonocardia, have been reported to produce a diverse array of antimicrobial metabolites(Mueller et al., 2010). These secondary metabolites play an important role in microbial interactions, often suppressing the growth of phytopathogens. Furthermore, Pseudonocardia species are frequently associated with protective symbioses in insects and plants, where they contribute to defense against fungal infections(Currie et al., 1999; Li et al., 2022; Sen et al.,2009; Aylward et al., (2014)). Despite these promising traits, relatively few studies have examined the direct application of Pseudonocardia to control cucumber PM, leaving a critical gap in knowledge regarding its biocontrol potential.

Advances in high-throughput sequencing technologies have enabled comprehensive analyses of microbial communities in plant-associated environments. Amplicon sequencing of the 16S rRNA gene using Illumina MiSeq provides insights into how microbial communities shift in response to biocontrol treatments, thereby revealing ecological mechanisms underlying plant disease suppression(Bolyen et al., 2019; Caporaso et al., 2010; Du et al., 2024). In parallel, metabolomic approaches such as LC-MS/MS-Q-TOF allow for the identification of bioactive compounds produced by candidate biocontrol strains(Chen et al., 2020; Wu et al., 2015). Integrating these approaches offers a holistic understanding of both microbial ecology and the chemical basis of biocontrol activity.

The present study was therefore designed to investigate the biocontrol potential of Pseudonocardia strains against cucumber PM (Wu et al., 2023). Specifically, cucumber plants were treated with Pseudonocardia and compared with pesticide-treated and non-treated (NT) controls. Microbiome composition was analyzed using Illumina MiSeq sequencing, while LC-MS/MS-Q-TOF was employed to detect antimicrobial metabolites. Through this integrated analysis, we aim to evaluate whether Pseudonocardia can serve as a sustainable alternative to chemical fungicides and to provide insights into the ecological and biochemical mechanisms underlying PM suppression.
