본 연구는 호주 아열대 우림 자생종인 핑거라임(Citrus australasica)의 가공 부산물인 과피와 가시줄기, 그리고 과육과 종자 추출물을 대상으로 항산화, 항균 및 미백 효능을 평가하고, 분자 기전을 규명함으로써 기능성 화장품 원료로서의 활용가능성을 검토하고자 하였다. 항산화 활성 평가 결과, 과피 추출물은 가장 높은 총 플라보노이드 함량(131.43 mg QE/g)을, 가시줄기 추출물은 가장 높은 총 폴리페놀 함량(24.55 mg GAE/g)을 각각 나타내었다. 두 추출물 모두 5 mg/mL 농도에서 80% 이상의 우수한 DPPH 및 ABTS 라디칼 소거 활성을 나타내어, 비식용 부위인 가시줄기 역시 과피와 유사한 수준의 항산화 효능을 보유함을 보여주었다. 항균 활성 평가에서는 과육과 종자 추출물이 피부 상재균인 S. epidermidis에 대해 최소억제농도(MIC) 및 최소살균농도(MBC) 모두 17 mg/mL로 나타나, 시트러스 유래 천연 보존제로서 경쟁력 있는 활성을 보였다. 미백 활성 평가 결과, 과피 추출물(0.1 mg/mL)은 세포 내 멜라닌 생성을 72.51% 수준으로 억제하여 우수한 미백 효능을 나타내었으며, 가시줄기 추출물(0.1 mg/mL)은 세포 내 티로시네이즈 활성을 82.54% 억제하여 높은 효소 저해 효과를 보였다. 이러한 미백 지표의 차이는 분자 기전 분석을 통해 부위별로 상이한 작용 경로가 존재함을 보여주었다. 과피 추출물은 멜라닌 생성의 핵심 전사 인자인 유전자 발현을 75.26% 억제하여 전사 조절(transcriptional regulation) 기전을 나타내는 반면, 가시줄기 추출물은 유전자 발현 수준에서는 상대적으로 미미한 억제 효과를 보였지만 TRP-2 단백질 발현을 82.28%까지 감소시켜 단백질의 안정성 감소 또는 분해 촉진을 통한 단백질 번역 후 조절(post-translational regulation) 기전을 시사했다.
결론적으로 핑거라임 부산물 은 항산화 및 항균 활성을 포함한 전반적인 생리활성과 함께, MITF 매개 전사 억제와 TRP-2 단백질 안정성 조절이라는 이원화된 기전(dual-mechanism)을 통해 미백 효능을 발휘함이 확인되었다. 이러한 결과는 기존에 핑거라임 부산물을 고부가가치 기능성 화장품 소재로 활용할 수 있는 과학적 근거를 제시한다.

This study aimed to evaluate the antioxidant, antimicrobial, and whitening activities of extracts obtained from processing byproducts of finger lime(Citrus australasica), an endemic subtropical rainforest species of Australia, including peel, thorny stem, pulp, and seeds, and to elucidate their molecular mechanisms, thereby examining their applicability as functional cosmetic ingredients. In antioxidant activity assessments, the peel extract exhibited the highest total flavonoid content(131.43 mg QE/g), while the thorny stem extract showed the highest total polyphenol content(24.55 mg GAE/g). Both extracts demonstrated strong DPPH and ABTS radical scavenging activities exceeding 80% at a concentration of 5 mg/mL, indicating that the non-edible thorny stem possesses antioxidant efficacy comparable to that of the peel. In antimicrobial activity evaluations, the pulp and seed extracts showed minimum inhibitory concentration(MIC) and minimum bactericidal concentration(MBC) values of 17 mg/mL against the skin-resident bacterium Staphylococcus epidermidis, and exhibited competitive activity as citrus-derived natural preservatives. In whitening activity assays, the peel extract(0.1 mg/mL) suppressed intracellular melanin production to 72.51%, exhibiting notable whitening efficacy, whereas the thorny stem extract(0.1 mg/mL) inhibited intracellular tyrosinase activity by 82.54%, showing a strong enzyme inhibitory effect. These differences in whitening indicators showed that distinct action pathways exist depending on the tissue site, as revealed by molecular mechanism analyses. The peel extract suppressed MITF gene expression, a key transcriptional regulator of melanogenesis, by 75.26%, indicating a transcriptional regulation mechanism, whereas the thorny stem extract showed relatively minor suppression at the gene expression level but reduced TRP-2 protein expression by up to 82.28%, thereby implicating a post-translational regulation mechanism mediated by decreased protein stability or enhanced protein degradation.
In conclusion, finger lime byproducts exhibited overall physiological activities, including antioxidant and antimicrobial effects, and exerted whitening efficacy through a dual-mechanism involving MITF-mediated transcriptional suppression and TRP-2 protein stability regulation. These findings provide scientific evidence for the utilization of previously discarded finger lime byproducts as high-value functional cosmetic ingredients.
