Maturation of cardiomyocytes is an essential process for heart development and
functional stability, requiring the integration of metabolic, electrical, and structural remodeling.
However, immature cardiomyocytes maintained under in vitro culture conditions exhibit
reduced functional stability compared to adult cardiomyocytes, limiting their utility in disease
modeling and pharmacological testing. In this study, we systematically investigated the effects
of long-chain fatty acids(LCFAs) and the sympathetic neurotransmitter norepinephrine(NE)
on the maturation of neonatal rat ventricular myocytes(NRVMs). SU-8–based
microcantilevers and microelectrode arrays(MEAs) were employed for long-term monitoring
of contractile force and electrophysiological activity, while qPCR, Western blotting,
immunocytochemistry, and holotomography were used to assess metabolic and structural
maturation markers. Our results demonstrated that combined treatment with oleic acid(OA)
and γ-linolenic acid(GLA) significantly enhanced contractile amplitude, contraction rate, field
potential amplitude and duration, and the expression of genes and proteins associated with
fatty acid oxidation(FAO). In contrast, NRVMs treated with LCFAs alone exhibited a
progressive decline in contraction rate during long-term culture; this decline was effectively suppressed when NE was co-administered, resulting in sustained contractile activity. Notably,
co-treatment with NE and OA produced the most pronounced improvements in mechanical
and electrical maturation, and the combination of NE, OA, and GLA showed an additional
upward trend. These findings indicate that LCFAs and NE act through distinct pathways but
converge on the PGC-1α–PPARα regulatory axis to promote integrated metabolic, electrical,
and structural maturation. Collectively, this study demonstrates that co-stimulation with
LCFAs and NE effectively induces functional maturation of immature cardiomyocytes,
providing a strategic basis for advancing hiPSC-CM maturation research, cardiac disease
modeling, drug responsiveness testing, and regenerative medicine applications
