Deep geological repositories for high-level radioactive waste are expected to become hyperalkaline (pH > 13) due to the influence of cement, and elevated temperatures (up to ~100 °C) as a result of radioactive decay. Over geological timescales of hundreds of thousands to millions of years, steel corrosion and radiolysis of water can lead to the accumulation of hydrogen (H2), and may cause a risk to repository stability. Therefore, microorganisms capable of oxidizing H2 while reducing and immobilizing metal ions and radionuclides may play an important role in enhancing the long-term safety of a repository system.
The objectives of this study were to investigate the diversity of hydrogen-oxidizing iron-reducing bacteria (IRB) and sulfate-reducing bacteria (SRB) enriched from deep-groundwater and tidal-flat sediments in Korea, and to evaluate their growth characteristics and metal reduction under alkaline pH conditions (pH 8-10). Furthermore, examined their ability to reduce and immobilize selenium (Se), a redox-sensitive radionuclide.
Samples were collected from deep groundwater (280-340 m depth) at the KAERI Underground Research Tunnel (KURT) and from tidal-flat sediments (20 cm depth) in Muan (MA), Korea. Hydrogen-oxidizing IRB were enriched using ferric citrate as the electron acceptor and a H2/N2 gas mixture (20%/80%) as the electron donor, while SRB were enriched using ferrous ammonium sulfate as the electron acceptor. Iron-reduction experiments were conducted using iron oxyhydroxide (akaganeite) and hydrogen gas, and microbial activity was evaluated under different alkaline pH conditions (pH 8, 9, and 10). To assess radionuclide immobilization under repository-relevant conditions, microorganisms were incubated in media containing heat-treated bentonite with selenium(IV) [Se(IV)] added as sodium selenite. Identical experiments were performed using both growth media and distilled-water-based equilibrium water to compare the effects of microbial activity. Microbial community changes were analyzed using next-generation sequencing (NGS), while geochemical variations were monitored through Eh-pH measurements and inductively coupled plasma optical emission spectrometry (ICP-OES). The mineralogical characteristics of precipitates were identified using X-ray diffraction (XRD) and field-emission transmission electron microscopy (FE-TEM).
Enriched IRB cultures from deep-groundwater and tidal-flat sediments changed yellow to colorless within 7 and 1 days, respectively, whereas enriched SRB cultures produced black precipitates within 7 and 11 days. NGS results showed Paludirhabdus as the dominant genus in KURT_IRB and Desulforamulus in KURT_SRB. In contrast, tidal-flat enrichments (MA_IRB and MA_SRB) showed diverse microbial communities, including Citrobacter, Terrisporobacter, Paraclostridium, Shewanella, Romboutsia, Methanosarcina, and Lacrimispora. Among IRB cultures, MA_IRB was capable of reducing akaganeite to magnetite within 4 days and demonstrated active growth and reduction at pH 8-9, whereas activity was strongly inhibited at pH 10. Both KURT_SRB and MA_SRB formed amorphous FeS precipitates at pH 8-9.
Selenium reduction experiments showed that in equilibrium water, Se removal efficiencies were 33.7% (control), 39.2% (KURT_IRB), and 43.1% (MA_IRB), whereas in growth media, the efficiencies were 82.9% (control), 53.8% (KURT_IRB), and 79.8% (MA_IRB). For SRB, Se removal efficiencies in equilibrium water were 49.3% (control), 48.1% (KURT_SRB), and 75.7% (MA_SRB), while all SRB cultures in growth media showed nearly complete removal (≈100%). These results indicate that microbial activity significantly enhances reduction and immobilization, thereby decreasing the mobility of redox-sensitive radionuclides.
Hydrogen-oxidizing metal-reducing microorganisms—such as Paludirhabdus, Desulforamulus, and Shewanella—could contribute to stabilizing the near-field environment of high-level radioactive waste repositories (pH < 10). By oxidizing hydrogen and reducing iron, sulfate, and selenium, these microorganisms are expected to promote radionuclide immobilization and enhance the long-term geochemical stability of deep geological disposal systems.
