Sensorineural hearing loss (SNHL), including noise-induced hearing loss (NIHL) and age-related hearing loss (ARHL), is closely associated with excessive production of reactive oxygen species (ROS), which initiates irreversible damage to cochlear hair cells and auditory neurons. In this study, we investigated the otoprotective potential of Arachis hypogaea shell extract (AHE), a phytochemical-rich natural resource with strong antioxidant activity, against oxidative cochlear injury across multiple experimental models. In a NIHL mouse model, AHE reduced ROS accumulation, mitigated outer hair cells (OHC) loss and apoptosis, and significantly preserved auditory thresholds as assessed by auditory brainstem response (ABR) measurements. Importantly, in an ARHL mouse model, long-term oral administration of AHE effectively attenuated age-dependent elevation of ABR thresholds, particularly in high-frequency regions (8–24 kHz) that are most vulnerable to ARHL. Notably, higher-dose AHE administration resulted in more pronounced preservation of auditory function, demonstrating a dose-responsive protective tendency during aging. In UB-OC1 auditory cells, AHE activated the Nrf2/HO-1 antioxidant signaling pathway and upregulated downstream cytoprotective genes, including HO-1 and SOD, thereby enhancing cellular resistance to oxidative stress. LC–MS-based metabolite profiling identified Eriodictyol (ERD) as a major bioactive constituent of AHE. ERD exhibited Nrf2-activating potential by disrupting the KEAP1–Nrf2 interaction and promoting transcriptional upregulation of antioxidant defenses. ERD also effectively suppressed intracellular ROS and protected auditory cells under chemically induced oxidative stress, supporting its role as a principal mediator of AHE’s bioactivity. Furthermore, in an H2O2-induced cellular senescence model, AHE reduced senescence-associated β-galactosidase activity and p53 accumulation, indicating attenuation of oxidative stress–driven senescence responses. Collectively, AHE and its major active constituent ERD exert protective effects against oxidative cochlear injury by activating Nrf2-based antioxidant pathways, suppressing ROS accumulation, preserving cochlear cellular integrity, and delaying age-related auditory functional decline. These mechanisms—redox balance regulation, enhancement of antioxidant defense systems, and prevention of hair cell damage—are fundamental to multiple forms of SNHL. Therefore, AHE and ERD hold promising potential as preventive or adjunctive therapeutic candidates for oxidative stress–mediated auditory disorders, including both NIHL and ARHL. Keywords: Sensorineural hearing loss (SNHL); Arachis hypogaea shell extract (AHE); Oxidative stress; Nrf2; Eriodictyol (ERD)
