Phytochromes are well-characterized photoreceptors sensing red and far-red light. Previously, reduced responses to blue light of the phytochrome-deficient mutants were shown compared with wild-type Arabidopsis, suggesting that phytochromes also play roles in blue light signaling. However, the molecular mechanism underlying phytochrome-mediated blue light signaling remains largely unknown. In the first topic of this study, a key finding is that blue light-induced degradation of PHYTOCHROME-INTERACTING FACTOR 3 (PIF3) depends on red/far-red photoreceptor phytochromes A and B (phyA and phyB), but not on blue photoreceptor cryptochromes, as evidenced by the phenotypes of exaggerated hypocotyl elongation in phyA and phyAphyB mutants under blue light. Subsequent analyses demonstrated that blue light is able to activate phyA and phyB, which induces their nuclear localization and interaction with PIF3, resulting in PIF3 degradation. Under low-intensity blue light, phyA predominantly mediates PIF3 degradation. In addition, likely in cooperation with cryptochromes, phyA also promotes HY5 accumulation through inactivation of the COP1/SPA E3 ligase complex. On the other hand, under high-intensity blue light, the functional role of phyB in PIF3 degradation and cryptochrome- mediated HY5 accumulation becomes more prominent, revealing a complementary but non- redundant functional relationship among these photoreceptors across a broad range of light intensities. Moreover, the reduction in blue light serves as a signal to trigger senescence in mature leaves, in which phyA delays chlorophyll degradation by limiting PIF3 accumulation, resulting in the transcriptional suppression of SAG12 but the maintenance of CAB transcript levels. Collectively, these findings indicate that phyA and phyB regulate PIF3 degradation and HY5 accumulation under blue light in coordination with cryptochromes, thereby extending their regulatory roles in blue light signaling from seedling photomorphogenesis to leaf senescence across various blue light intensities. CONSTITUTIVE SHADE-AVOIDANCE 1 (CSA1) encodes a Toll/Interleukin-1 receptor- nucleotide binding site-leucine-rich repeat (TIR-NBS-LRR) immune receptor. The csa1 mutant, generated by a T-DNA insertion, produces a truncated form (ΔCSA1) that leads to phenotypes similar to those of the phyB-deficient mutant, including elongated hypocotyls and early flowering under long-day conditions. However, the molecular signaling mechanism through which ΔCSA1 induces phyB-like phenotypes remains elusive. In the second topic of this study, a key finding is the direct interaction between ΔCSA1 and PIFs, which play critical roles in phytochrome signaling. In particular, among PIFs, ΔCSA1 interacted most strongly with PIF4, which is normally phosphorylated and degraded upon light exposure during photomorphogenic development. Although ΔCSA1 does not disrupt the interaction between phyB and PIF4, it inhibits phyB-mediated phosphorylation of PIF4 in vitro, which prevents red light-induced degradation in plants. Consequently, the expression of ΔCSA1 in plants elevates the levels of PIF4 under red light, thereby enhancing the transcription of PIF4-responsive genes such as YUC8 and IAA19, which can induce constitutive shade-avoidance phenotypes, including elongated hypocotyls and early flowering. Furthermore, in transgenic plants harboring a constitutively active phyB mutant (YVB), ΔCSA1 expression suppresses the light-independent constitutive photomorphogenic phenotype of the YVB plant. Additionally, the hypocotyl lengths of csa1 and ΔCSA1-expressing plants are significantly longer than those of the control plants (Col-0 and Ler), but shorter than those of the phyB-deficient mutants, due to HY5 accumulation under red light. These findings reveal a novel molecular mechanism in which the TIR-containing truncated form of CSA1 regulates phyB signaling by interacting with and stabilizing PIF4 in plants, and suggest that light-induced regulation of both PIF4 degradation and HY5 accumulation is important for the successful establishment of seedlings. BLUE LIGHT INHIBITORS OF CRYPTOCHROMES 1 and 2 (BIC1 and BIC2) are known as negative regulators of blue light signaling that suppress the activation of cryptochromes 1 and 2 (CRY1 and CRY2), and previous studies have also reported that BIC1 functions as a transcriptional coactivator through its interaction with PIF4. In addition, different subcellular localizations of BICs have been reported. However, the interconnection between their function and subcellular localization has not been studied. In the third topic of this study, key findings include distinct subcellular distribution patterns of BICs and the association of their differential localization with roles in cryptochrome inhibition and PIF4 regulation. BIC1 was localized to both the cytoplasm and the nucleus, whereas BIC2 was predominantly confined to the nucleus. Under blue light, BIC1:NLS and BIC2:NLS plants exhibited reduced responsiveness similar to BIC1- and BIC2-overexpressing plants, indicating that cryptochrome repression persists when BICs are restricted to the nucleus. In contrast, BIC1:NES and BIC2:NES plants, in which BICs accumulate in the cytoplasm, inhibit the nuclear import of CRYs. In addition to their roles in blue light signaling, BICs also function in temperature-responsive growth. CRY1 suppresses thermomorphogenesis by interacting with PIF4 and reducing its transcriptional activity, whereas BIC1 and BIC2 promote hypocotyl elongation at high-temperature by disrupting the CRY1-PIF4 interaction. Conversely, excessive nuclear accumulation of BIC2 in BIC2:NLS plants enhanced its association with PIF4, leading to repression of PIF4 target gene expression and consequently reducing hypocotyl elongation under high-temperature. Collectively, these results suggest that the distinct subcellular localizations of BIC1 and BIC2 contribute to the functional separation of cryptochrome inhibition and PIF4 regulation. Therefore, BICs may act as integrative regulators for growth responses to light and temperature signals under variable environmental conditions by modulating the balance between cryptochrome signaling and PIF4-mediated transcriptional activity.
