Cardiovascular diseases remain the leading cause of mortality worldwide, and the development of new therapeutics requires substantial time and cost, particularly due to stringent safety and efficacy evaluations. A significant proportion of early-stage drug candidates fail because of cardiotoxicity, underscoring the need for reliable in-vitro models. In this study, we propose an SU-8–based cantilever array capable of precisely quantifying the contractile behavior of cardiomyocytes, and we comprehensively evaluate its mechanical and electrical characteristics. Conventional cantilever sensors often suffer from limited fabrication stability and reproducibility due to manual release steps. To address these limitations, we employed a Deep Reactive Ion Etching (DRIE) process to simultaneously define the cantilever and supporting body, thereby minimizing post-processing damage and ensuring high uniformity and yield. A metal strain sensor was integrated onto the SU-8 cantilever to enable stable conversion of cellular contractions into measurable electrical signals. The fabricated cantilever array was validated through a series of assessments, including signal stability under temperature and culture medium conditions, resistance and voltage changes with displacement, and cell culture experiments. The results demonstrate that the proposed DRIE-based cantilever array provides a robust tool for accurately evaluating the mechanical responses of cardiomyocytes and establishes a foundation for future systems capable of real-time electrical monitoring of cellular contractile activity. These characteristics support the cantilever array’s potential applicability in cardiotoxicity screening, drug response monitoring, and mechanistic studies related to cardiac diseases.
