ADC(Antibody-Drug Conjugate)
(All other drugs), (2) ADC
Group(Zemtuzumab ozogamicin, Brentuximab vedotin, Trastuzumab emtansine, Trastuzumab deruxtecan, Enfortumab vedotin, Sacituzumab govitecan,
Tisotumab vedotin, Polatuzumab vedotin, Mirvetuximab soravtansine, Datopotamab deruxtecan,) (3) Immune Therapy Group(Blinatumomab,

Inotuzumab ozogamicin is an antibody–drug conjugate (ADC) composed of
    a CD22-targeting monoclonal antibody linked to a calicheamicin-derived
payload that induces DNA strand breaks, and it is used for the treatment of
relapsed or refractory acute lymphoblastic leukemia. By exploiting the antigen
specificity of the antibody component, the drug is designed to preferentially
deliver the cytotoxic payload to malignant cells while limiting exposure of
normal tissues, thereby enhancing antitumor efficacy and reducing off-target
    toxicity.
Despite its substantial therapeutic benefit, serious hepatic adverse reactions,
including veno-occlusive disease (VOD), have been reported in both clinical
trials and post-marketing safety assessments. Given the clinical severity of
VOD and its implications for patient outcomes, there is a clear need to evaluate
the hepatic risk of inotuzumab ozogamicin under real-world treatment
conditions. Accordingly, the present study aimed to detect liver-related safety
signals associated with inotuzumab ozogamicin using the World Health
Organization global pharmacovigilance database, VigiBase, and to assess its
relative risk in comparison with multiple reference drug groups.
This analysis included all liver-related adverse events of interest reported
to VigiBase up to January 2025 that were classified under Standard MedDRA
    Queries (SMQs) 20000008, 20000009, 20000010, 20000013, and 20000084.
Hepatic risk was evaluated through disproportionality analyses using the
reporting odds ratio (ROR) and information component (IC), with three
comparator groups: (1) all other drugs; (2) an ADC group comprising
gemtuzumab ozogamicin, brentuximab vedotin, trastuzumab emtansine,
trastuzumab deruxtecan, enfortumab vedotin, sacituzumab govitecan, tisotumab
vedotin, polatuzumab vedotin, mirvetuximab soravtansine, and datopotamab
deruxtecan; and (3) an immune therapy group including blinatumomab,
    brexucabtagene autoleucel, and tisagenlecleucel.
The analysis demonstrated a statistically significant disproportionality signal
for hepatic adverse events associated with inotuzumab ozogamicin compared
with all other drugs, with an adjusted reporting odds ratio (aROR) of 6.38. When
compared with the ADC group, inotuzumab ozogamicin also showed a higher
relative reporting frequency of liver-related adverse events (aROR 2.23).
Similarly, comparison with the immune therapy group revealed a significant
signal, with an aROR of 3.90. Consistent signals were further observed across
SMQ-based subgroup analyses and multiple sensitivity analyses, supporting the
robustness of the identified hepatic safety signals for inotuzumab ozogamicin.
