과학수사 분야에서는 미량의 시료로부터 신뢰성 있는 정보를 얻는 것이 매우 중요하다. 피부 마이크로바이옴은 주변 환경과 숙주의 상태에 따라 민감하게 변화하는 양상을 보이며, 이러한 특성은 피부 마이크로바이옴이 수사지원정보 획득에 활용될 수 있는 가능성을 보인다. 이에 본 연구에서는 과학수사 지원을 위해 피부 마이크로바이옴을 기반으로 인구학적 정보, 생물학적 정보, 생활 습관과 관련된 수사지원정보의 예측 가능성을 평가하였으며, 해외 공공데이터를 활용하여 거주지역에 따른 피부 마이크로바이옴의 차이에 대해 분석하였다.
그 결과, 피부 부위, 피부 유분도 및 수분도, 성별, 연령, 체질량지수(BMI), 동거인 또는 배우자 유무, 반려동물 유무, 음주 및 흡연 습관, 화장품 사용, 야외 활동 시간, 프로바이오틱스 섭취, 거주 지역에 따라 피부 미생물 구성에 차이가 확인되었다. 이 요인들 중 피부 부위, 성별, 연령, 동거인 또는 배우자 유무, 일일 흡연량, 일일 프로바이오틱스 섭취 여부, 국가 내 및 국가 간 지리적 위치에 대해 유의한 머신러닝 기반 예측 모델이 개발되었다. 모델의 접근성과 실용성을 높이기 위해, 개발된 모든 모델은 MATLAB 기반 어플리케이션으로 통합하였다.
전반적으로, 이 연구는 포렌식 마이크로바이옴 연구 분야를 발전시키고, 피부 마이크로바이옴이 과학수사에 있어서 보조적인 역할을 할 수 있음을 시사한다. 이 연구 결과는 미량 DNA의 활용 가능성을 확장하고, 과학수사 목적으로 피부 마이크로바이옴 데이터를 활용하기 위한 프로토타입 분석 모델을 제공한다.

In forensic science, obtaining reliable information from trace biological evidence is of critical importance. The skin microbiome exhibits sensitive responses to environmental conditions and host-related factors, indicating its potential value as a source of investigative information. In this study, skin microbiome patterns in a Korean population was compared to establish a standardized approach for forensic applications. The feasibility of predicting investigative information—including demographic characteristics, biological factors, and lifestyle habits—was further evaluated using skin microbiome data. Additionally, publicly available international datasets were analyzed to assess differences in the skin microbiome according to residential area.
As a result, differences in skin microbial composition were observed according to skin site, skin oil and moisture levels, biological gender, age, body mass index (BMI), presence of cohabitants or a spouse, companion animal ownership, drinking and smoking habits, cosmetic use, time spent outdoors, probiotics intake, and residential information. Among these factors, machine learning–based prediction models were successfully developed for skin site, biological gender, age, presence of cohabitants or a spouse, daily smoking amount, daily probiotics intake, and both inter-national and cross-national geolocation. To enhance accessibility and practical applicability, all developed models were integrated into a MATLAB-based application.
Overall, this study advances the field of forensic microbiome research and highlights the potential utility of the skin microbiome as a supplementary forensic tool. The findings extend the scope of forensic analysis in situations involving trace DNA and provide prototype analytical models for the forensic application of skin microbiome data.
