Mutations in the tumor suppressor gene STK11 (Serine-Threonine Kinase 11), also known as LKB1 (Liver Kinase B1), are frequently observed in non-small cell lung cancer (NSCLC), particularly when co-occurring with KRAS mutations. This co-mutation is strongly associated with poor clinical prognosis and resistance to therapies, including immune checkpoint inhibitors and KRAS-targeted therapies. Despite its clinical importance, the functional consequences of individual STK11 variants remain poorly understood. In this study, we first established five STK11 knockout NSCLC cell lines using CRISPR-Cas9 genome editing. Drug response profiling of these knockout models showed altered sensitivity to KRAS inhibitors, confirming the biological impact of STK11 loss. To systematically evaluate the phenotypic effects of STK11 mutations, we constructed a site-saturation variant library (SSVL) comprising 8,660 unique variants covering all codons of the STK11 coding sequence. This library will be introduced into STK11-deficient NSCLC cells and screened under pharmacologic and proliferative stress to identify functionally significant mutations. Our research will provide a functional mapping of STK11, forming a foundation for identifying clinically actionable variants and advancing precision oncology approaches in NSCLC.
