Functional and structural mechanisms that govern the PilQ secretin of the type IV pilus (T4P) system in Pseudomonas aeruginosa were refined in this study. Through a combination of genetic analyses and visualization through cryo-electron microscopy, we demonstrate that SlkA and SlkB function as luminal plug that block disengaged PilQ pores and preserve outer-membrane integrity, revealing that secretin gating is maintained by a multilayered stabilization rather than by PilQ alone. Comparative analyses of PilQ-associated accessory components—including the periplasmic scaffold TsaP, the outer-membrane ring protein PA0359, and the AMIN domains within the PilQ N-terminus—show that these factors contribute in distinct and non-redundant ways to secretin positioning, stability, phage susceptibility, and T4P-dependent motility. AMIN1 emerged as an essential determinant of secretin’s polar localization and phage entry, whereas PA0359, despite its well-defined ring architecture, may perform environment- or matrix-dependent functions that were not captured under the tested conditions. Together, this study establishes PilQ as a multifunctional structure supported by diverse periplasmic and outer-membrane elements and provides an insight of mechanism for understanding how secretin-associated networks enable the surface behaviors of P. aeruginosa.
