Three-dimensional (3D) microphysiological systems that merge microfluidics with accessible fabrication routes provide a controllable way to model metabolic liver disease in vitro while retaining essential multicellular interactions. This thesis develops a MSG to assemble reproducible hepatocyte–stellate co-cultures and to orchestrate stepwise perturbations that emulate key features of metabolic dysfunction–associated steatohepatitis. The platform stabilizes hanging-drop cultures, supports uniform spheroid formation, and permits non-disruptive, temporally precise reagent exchanges suited to longitudinal studies and comparative assays. Within this ecosystem, steatosis, oxidative stress, inflammatory signaling, and matrix remodeling are profiled through imaging and transcript readouts to yield an integrated view of disease-relevant
phenotypes. Methodologically, the work emphasizes manufacturability, workflow robustness, and compatibility with standard plate formats to promote adoption, reduce operator variance, and enable parallel condition testing. Conceptually, it positions microfabricated liver spheroids as a bridge between oversimplified two-dimensional (2D) cultures and resource-intensive
animal models, highlighting advantages for dissecting timing, dose, and sequence of stimuli as well as for evaluating combination interventions. The closing perspective outlines extensions to patient-derived inputs, gradient generators for microenvironment control, and automation for medium-throughput screening, underscoring the potential of this approach to accelerate discovery and prioritization of anti-steatotic, anti-oxidative, and anti-fibrotic strategies while reducing cost and ethical burdens.
