연구배경
비만으로 인한 근육 내 지방 축적은 지질독성(Lipotoxicity)과 인슐린 저항성을 초래한다. 이러한 대사 이상은 골격근의 기능 저하와 대사 질환 발생의 주요 원인이다. 근육의 지방독성 기전을 규명하고 이를 완화할 수 있는 새로운 기능성 소재를 탐색하는 것이 중요하다.
연구목적
본 연구는 골격근 세포에서 지방 축적 및 지질독성에 미치는 영향과 그 작용 기전을 밝히고 천연화합물인 이소람네틴(Isorhamnetin)이 이 어떠한 영향을 미치는 지 조사하고자 한다. 이를 통해 근육 내 지질 대사 이상으로 발생한 대사 질환 예방 및 개선을 위한 후보물질로서 이소람네틴의 가능성을 평가하고자 한다.
연구방법
Oleic acid 를 처리한 C2C12 지방 축적 모델을 구축하고, 이소람네틴을 병용 처리하였다. 이소람네틴의 세포 수준에서 생리적 보호 효과를 평가하였다. 세포 내 지질 축적은 AdipoRed 염색 과 Confocal microscopy 로 분석하였고, 세포 생존율은 MTT 분석법으로 확인하였다. 전사체 분석(RNA-seq)을 통해 에너지 대사, 지질독성 관련 신호 전달 경로, 인슐린 저항성에 영향을 미치는 유전자 발현 변화를 탐색하였다. 주요 경로에 속하는 유전자들의 발현은 PT-PCR 으로 검증하였다.
연구결과
이소람네틴은 Oleic acid 로 유도된 C2C12 근관세포에서 세포 생존율에 영향을 미치지 않으면서, 세포 내 지질 방울 형성을 농도 의존적으로 유의하게 감소시키는 효과를 보였다. 또한, AdipoRed 및 Confocal 이미징 분석을 통해 이소람네틴 처리군에서 세포 내 지질 축적이 현저히 완화됨을 확인하였다. 이러한 결과는 이소람네틴이 골격근 세포 수준에서 지질 독성을 완화하고, 세포의 대사 항상성 유지에 기여할 가능성을 시사한다.

Excessive lipid accumulation within skeletal muscle is a primary contributor to obesity-associated metabolic disorders, frequently precipitating lipotoxicity and insulin resistance. Such disruptions in cellular energy homeostasis impair myocyte function and are critical factors in the pathogenesis of metabolic diseases. Consequently, identifying novel functional agents capable of mitigating muscle lipotoxicity is of significant clinical importance. While isorhamnetin, a naturally occurring flavonol, is recognized for its anti-obesity and anti-inflammatory properties, its specific regulatory effects on skeletal muscle lipid metabolism and lipotoxicity have not been fully elucidated.
Objective
The primary objective of this study was to evaluate the impact of isorhamnetin on lipid retention and lipotoxicity in skeletal muscle cells. Furthermore, this research aimed to define the underlying molecular mechanisms, specifically targeting signaling pathways regulating energy metabolism and insulin sensitivity, to assess the potential of isorhamnetin as a therapeutic candidate for metabolic diseases.
Methods
To simulate lipid overload in vitro, C2C12 myotubes were treated with oleic acid and subsequently co-treated with isorhamnetin. Intracellular lipid content was quantified using AdipoRed staining followed by confocal microscopy, and cell viability was assessed via MTT assays. To delineate the molecular landscape, transcriptomic profiling was conducted using RNA sequencing, focusing on gene expression changes related to metabolic regulation and lipotoxicity. Finally, the expression levels of key genes identified from the sequencing data were validated using quantitative real-time PCR (RT-qPCR).
Results
Isorhamnetin treatment significantly reduced lipid droplet formation in a dose-dependent manner in oleic acid-induced C2C12 myotubes, without compromising cell viability. Consistent with these findings, AdipoRed staining and confocal imaging analyses confirmed a marked attenuation of intracellular lipid accumulation in the isorhamnetin-treated group. Collectively, these results suggest that isorhamnetin effectively alleviates lipotoxicity and contributes to the maintenance of metabolic homeostasis in skeletal muscle cells.
Conclusion
In conclusion, this study demonstrates that isorhamnetin effectively attenuates lipid accumulation and lipotoxicity in skeletal muscle cells through the modulation of genes governing lipid metabolism and metabolic signaling. These findings indicate that isorhamnetin contributes to the preservation of metabolic homeostasis in skeletal muscle. Consequently, isorhamnetin holds potential as a functional agent for the prevention or amelioration of metabolic disorders associated with muscle lipotoxicity.
