흑색종은 가장 공격적이고 사망률이 높은 피부암 중 하나이다. Casein Kinase 2 (CK2)는 세포 생존, 증식, 세포 주기 조절 등 다양한 세포 기능에 관여하는 serine/threonine kinase 로, 수백 개의 기질 단백질을 인산화한다. CK2 의 과발현은 다양한 암에서 보고되었으며, 흑색종에서도 높게 발현된다. CK2 저해제인 CX-4945 는 CK2α에 ATP-경쟁적으로 결합하여 활성을 억제한다.
Histone Deacetylase (HDAC) 역시 여러 암에서 과발현되며, CK2 매개 인산화에 의해 활성이 조절되는 것으로 알려져 있다. 본 연구에서는 melanoma 세포주(A375, PMWK, SK-MEL-2, HMCB)를 대상으로 CX4945 의 효과를 분석하였다. 암 유전체 아틀라스(TCGA) 데이터 분석 결과, 흑색종 조직에서 정상 피부 대비 CK2α 발현이 증가된 것으로 나타났다. CX4945 처리 시 용량 의존적으로 HDAC(HDAC1/2/3)의 인산화 및 활성이 현저히 감소하였다. CX-4945 는 흑색종 세포주에서 세포 생존율, 콜로니 형성 및 이동을 억제하고 세포 사멸을 유도하는 것으로 확인되었다. 기전 연구에서 CX-4945 는 mRNA 및 단백질 수준에서 BAX, BAD, PUMAα와 같은 세포사멸 촉진 유전자의 발현을 상향 조절하고 JNK 신호전달 경로를 활성화했다. 또한 siRNA 또는 Vorinostat 을 이용한 CK2 와 HDACs 의 병용 억제는 단일 요법보다 세포 사멸을 증가시키고 흑색종 세포 성장을 더 효과적으로 억제하였다. 결론적으로, 본 연구는 CK2 억제제 CX-4945 HDAC 활성을 억제하고 흑색종 세포에서 세포 사멸을 유도함을 입증하여, Vorinostat 와 같은 HDAC 억제제와의 병용 사용 가능성을 시사한다.

Melanoma is one of the most aggressive and treatment-resistant forms of skin cancer. Although targeted therapies such as BRAF/MEK inhibitors and immune checkpoint blockade have improved clinical outcomes, their efficacy is often limited by intrinsic or acquired resistance. Casein Kinase 2 (CK2) is a serine/threonine kinase involved in various cellular processes, including cell survival, proliferation, and cell cycle regulation, and it phosphorylates a wide range of substrate proteins. Overexpression of CK2 has been reported in various cancers, including melanoma. CX-4945 is the first CK2 inhibitor to enter clinical trials, acting by competitively inhibiting the ATP-binding site of the CK2α subunit. Histone deacetylases (HDACs) are also frequently overexpressed and are known to be regulated by CK2-mediated phosphorylation in cancer. In this study, we investigated the growth inhibition and cell death of the CK2α inhibitor CX-4945 in melanoma cell lines (A375, PMWK, SK-MEL-2, HMCB). CX-4945 inhibited HDAC phosphorylation, leading to induced apoptosis through the reduction of HDAC activity in melanoma cells. Analysis of The Cancer Genome Atlas (TCGA) data revealed elevated expression of CK2α in melanoma tissues compared with normal skin. CX-4945 treatment significantly reduced phosphorylation and activity of class I HDACs (HDAC1/2/3) in a dose-dependent manner. Functional studies demonstrated that CX-4945 suppressed cell viability, colony formation, and migration, in addition to inducing apoptosis across multiple melanoma cell lines. Mechanistically, CX-4945 upregulated pro-apoptotic genes such as BAX, BAD, and PUMAα at both the mRNA and protein levels, and activated the JNK signaling pathway. Notably, the effects of CX-4945 were less pronounced in PMWK cells, suggesting that genetic context may influence drug sensitivity. Moreover, combinatorial inhibition of CK2 and HDACs using siRNA or vorinostat synergistically enhanced apoptotic cell death and suppressed melanoma cell growth more effectively than monotherapy. In conclusion, this study demonstrates that the CK2 inhibitor CX-4945 suppresses class I HDAC activity and induces apoptosis in melanoma cells, supporting its potential use in combination with HDAC inhibitors such as Vorinostat.
