    수서 생물종 대상 중금속 및 미세플라스틱의 복합 독성 연구 수서 생태계에서 여러 오염물질의 공존은 수서 생물종에 대해 심각한 영향을 미치며, 각각의 물질들로 인한 독성의 합으로는 설명하기 어려운 복합 독성을 유발한다. 플라스틱은 자외선, 물리적 풍화, 합성 섬유의 세탁, 타이어 마모와 같은 일상적 활동으로 파편화되어 미세플라스틱을 형성한다. 미세플라스틱은 1 µm에서 5 mm 사이 크기를 가지는 합성 고체 플라스틱 입자 및 고분자 매트릭스로 정의되며, 작은 입자 크기, 화학적 안정성, 넓은 비표면적과 같은 특성으로 인해 생태계에 장기간 잔류할 수 있다. 생태계에 잔류하며 지속적인 독성을 유발하는 미세플라스틱과, 인류의 기술 발전에 따라 축적된 중금속의 공존은 실제 생태계에서 충분히 발생할 수 있는 시나리오임에도 불구하고, 기존 연구들은 각각의 오염 물질의 개별 독성 영향에 초점을 맞춰, 복합 노출에 따른 독성학적 영향과 그 기작에 대해서는 아직 일관된 결론이 도출되지 않았다. 본 연구는 수서 생물종을 대상으로 수행된 미세플라스틱과 중금속의 복합 노출 관련 132편의 선행 연구 문헌을 수집하여, 생물종, 미세플라스틱의 크기 및 소재, 중금속 섭취량 변화를 고려하여 다양한 생물종에 공통으로 적용할 수 있는 다섯 가지 종말점(산화스트레스, 발달, 번식, 성장, 치사)을 중심으로 복합 독성의 발생 여부를 평가하고 분류하였다. 그 결과 총 84편의 문헌에서 중금속과 미세플라스틱의 복합 노출이 각각의 오염물질에 의한 개별 독성의 합을 초과하는 시너지 효과를 나타냄을 확인할 수 있었다. 이러한 시너지 효과는 다수의 문헌에서 미세플라스틱이 중금속의 운반체로 작용한 뒤 생명체의 체내에서 탈착되어 생체이용률을 증가시키는 트로이 목마 효과(Trojan horse effect)에 기인한 것으로 제시했고, 그 결과 각각의 종말점에서 중금속 독성이 증폭되는 경향이 보고되었다. Daphnia magna를 대상으로 한 폴리에스터 미세섬유와 구리(Cu2+)의 48시간 복합 노출 사례 연구 결과, EC50가 단독 노출 대비 약 46.9% 감소해 급성 독성이 증가하는 경향을 확인하였다. 활성산소종 분석 결과, 복합 노출군에서 활성산소종 농도가 단독 노출군 대비 최대 31.17% 증가하였으며, catalase레이스 활성 또한 구리 복합 노출 조건에서 효소 활성 감소를 통한 항산화 체계 붕괴를 관찰할 수 있었다. 이는 선행 연구 분석을 통해 도출된 복합 독성 양상과 부합하는 결과로 해석된다. 종합하자면, 미세플라스틱과 중금속의 복합 노출 결과 개별 오염물질 노출에 비해 추가적인 독성이 발생하며, 미세플라스틱에 대한 중금속의 흡착과 생체이용률 증가가 주된 원인으로 확인하였다. 다만 선행 연구에서 활용된 미세플라스틱 소재와 실험 종의 제한으로, 본 분석에서 확인된 추가 독성은 실제 생태계에서 나타날 복합 독성을 정확하게 반영되지 않았을 가능성이 있다. 따라서 두 물질의 복합 독성을 명확히 규명하기 위해 보다 다양한 생물종과 미세플라스틱 소재를 활용한 후속 연구가 필요하다.

    Exposure to multiple pollutants in aquatic ecosystems can trigger toxic responses that cannot be explained by the sum of the toxicities of individual contaminants. Plastics can be fragmented into microplastics through UV radiation, physical weathering, and biological processes, or directly released via routine activities such as laundering of synthetic fibers and tire abrasion. Microplastics are defined as synthetic solid plastic particles or polymer matrices ranging in size from 1 µm to 5 mm. Due to their small size, chemical stability, and large surface area, microplastics are widely dispersed and spread in the environment. Although simultaneous exposure of aquatic organisms to microplastics and heavy metals can be a plausible scenario in real ecosystems given their respective accumulation patterns, much of the existing research has focused primarily on the individual toxicities of each pollutant.
    This study analyzed 132 scientific publications that comprehensively examined the exposure of aquatic organisms to microplastics and heavy metals. Common toxicological endpoints—oxidative stress, development, reproduction, growth, and mortality—were assessed across aquatic organisms, taking into account the type and size of microplastics, as well as changes in heavy metal uptake and metallothionein expression. Eighty-four of the analyzed studies reported synergistic toxicity effects, where combined exposures elicited responses greater than the sum of the individual toxicities. This synergy is often attributed to a "Trojan horse effect," where heavy metals adsorbed to microplastics are transported within organisms and then desorbed, thereby enhancing bioavailability and amplifying toxicity across various endpoints.
    In addition, a case study using Daphnia magna demonstrated that combined exposure to polyester microfibers and copper (Cu2+) increased acute toxicity, with a 46.9% decrease in the 48-hour EC50 compared to single exposure. Reactive oxygen species (ROS) analysis revealed that ROS levels in the combined exposure group increased by up to 31.17% compared to the single treatment group. Furthermore, catalase (CAT) activity decreased under high-concentration copper exposure conditions in the combined treatment, suggesting a breakdown of the antioxidant defense system. These results are consistent with the toxicological mechanism derived from the literature analysis.
    This study aims to comprehensively analyze the various toxicological interactions resulting from combined exposure to microplastics and heavy metals, complementing literature-based insights with experimental evidence. Consequently, the results of this study provide a scientific basis for improved ecotoxicological assessments and environmental risk assessments in multi-contaminant exposure scenarios.
