본 연구는 제주국제공항 내 표면에서 분포하는 미생물의 종류와 출현 특성을 파악하고, 공간·표면·시기적 요인에 따른 미생물 군집 구성의 변동성을 분석하기 위하여 수행되었다. 연구 대상은 국내선 및 국제선의 출·입국장으로 설정하였으며, 2024년 10월부터 2025년 3월까지 매월 동일한 위치에서 손잡이 표면과 바닥 표면을 대상으로 시료를 반복 채취하였다. 채취된 시료는 배양 후 순수 분리한 단일 콜로니를 대상으로 16S rRNA 유전자 분석을 통해 동정하였다. 분석 결과, 총 98종이 확인되었으며, 이들은 46개의 속에 속하는 것으로 나타났다. 월별 종수는 11월에 가장 높게 관찰되었고 이후 감소하였다가 3월에 다시 소폭 증가하는 변동 양상을 보였다. 속 단위에서는 Staphylococcus, Micrococcus, Bacillus, Microbacterium, Kocuria 등이 상대적으로 높은 빈도로 관찰되었으며, 각각은 출현 범위와 지속성에서 차이를 나타내었다. Staphylococcus와 Micrococcus는 조사 기간 전반에 걸쳐 지속적으로 확인된 반면, Bacillus와 Kocuria는 특정 시기에 집중되는 간헐적 출현 양상을 보였다. 또한 일부 속은 국제선 구역에서 상대적으로 많이 확인되었으며, 그중에는 해외 문헌에서 토양·식물 환경 또는 특정 지역에서 처음 보고된 이력이 있는 Micrococcus yunnanensis, Lactobacillus formosensis 등이 포함되었다. 구역별·표면별 비교에서는 손잡이 표면이 상대적으로 안정적인 군집 구성을 보인 반면, 바닥 표면에서는 월별 변동 폭이 더 크게 나타났다. 바닥 표면에서는 Bacillus, Kocuria 등 환경 기원 미생물이 넓은 범위에서 확인되었으며, 이는 외부 환경 노출과 사람 및 수하물 이동이 복합적으로 작용한 결과로 해석된다. 전체적으로 제주국제공항 표면 미생물 군집은 지속형·간헐형의 여러 출현 형태가 공존하는 다층적 구조를 보였으며, 시기적·공간적 특성이 군집 구성에 다양한 영향을 미치는 것으로 나타났다. 본 연구는 국내 국제공항을 대상으로 표면 미생물 군집을 장기간·정량적으로 조사한 자료로서, 공항과 같은 다중이용 교통시설의 위생 관리 체계를 이해하는 데 기초 자료로 활용될 수 있다. 아울러 표면 유형과 군집 변동성에 근거한 방역 장비 도입의 필요성을 제시함으로써, 공항 환경에서의 감염 관리 및 환경 위생 전략 수립에 실질적인 시사점을 제공한다.

This study was conducted to identify the types and occurrence patterns of microorganisms distributed on surfaces within Jeju International Airport and to analyze the variability of microbial community composition according to spatial, surface-specific, and temporal factors. Sampling was performed monthly from October 2024 to March 2025 at fixed locations within the domestic and international arrival and departure halls, targeting chair armrests (hand-contact surfaces) and floor areas contacted by shoe soles. Collected samples were cultured, and single colonies were isolated and identified using 16S rRNA gene sequencing. A total of 98 species belonging to 46 genera were identified. The number of detected species was highest in November, decreased thereafter, and showed a slight increase again in March. At the genus level, Staphylococcus, Micrococcus, Bacillus, Microbacterium, and Kocuria were observed with relatively high frequency, each exhibiting distinct patterns in occurrence range and temporal persistence. Staphylococcus and Micrococcus persisted throughout the study period, whereas Bacillus and Kocuria appeared intermittently, showing more seasonally concentrated patterns. Several genera were detected more frequently in the international terminal, including Micrococcus yunnanensis and Lactobacillus formosensis, which have been initially reported in soil, plant, or region-specific environmental studies. Comparative analysis by area and surface type revealed that hand-contact surfaces exhibited relatively stable community structures, whereas floor surfaces showed greater month-to-month variability. Floor samples contained broader distributions of environmental genera such as Bacillus and Kocuria, likely reflecting combined influences of outdoor air exchange, human movement, and baggage flow. Overall, the surface microbial communities of Jeju International Airport displayed a multilayered structure in which persistent and intermittent occurrence patterns coexisted, and temporal and spatial characteristics contributed to dynamic community variation. This study provides one of the first long-term, quantitative datasets on surface microbial communities within a Korean international airport and offers foundational evidence for understanding hygiene management in large-scale transportation facilities. Furthermore, by illustrating community variability according to surface type and occurrence patterns, the findings present practical implications for the introduction of targeted disinfection equipment and the development of infection control strategies in airport environments.
