본 연구는 Burkholderia sp. O33의 Botrytis cinerea에 대한 항진균 활성을 평가하고, Burkholderia의 엽면 처리에 따른 상추의 생리적·대사적 변화를 종합적으로 분석하기 위하여 수행되었다. 기내 항진균 실험에서 Burkholderia는 농도 의존적으로 강한 억제 효과를 나타냈으며, ≥5.9 × 10⁹ CFU/mL에서 효과적인 병원균 억제가 확인되었다. 종자 발아 시험 결과, ≤5.9 × 1010 CFU/mL 농도 범위에서 식물 독성은 나타나지 않아 Burkholderia의 식물 적용 가능성을 확인하였다.
포트에서 수행한 엽면 처리 실험에서는 fludioxonil 및Burkholderia 처리구 모두에서 상추의 생육(생체중, 엽수)이 개선되었으며, 이는 병 발생 억제 및 미생물 유래 생육 촉진 효과가 복합적으로 작용한 것으로 판단된다. 잎 분리 접종 실험에서도 처리 직후 Burkholderia 고농도 처리구에서 병반 확산이 감소하여 단기적 생체균의 길항 효과가 확인되었다.
대사체 분석 결과, 처리별로 뚜렷한 생리·대사 반응 차이가 나타났다. 스테롤 분석에서는 Burkholderia 처리 시 초기 증가 후 빠르게 정상화되는 경향을 보였으나, fludioxonil은 지속적인 스테롤 감소를 유도하였다. 지방산 분석에서는 fludioxonil이 포화지방산 증가 및 불포화지방산 감소를 유도한 반면, Burkholderia은 반대로 불포화지방산 증가를 유도하여 막 유동성과 스트레스 적응성을 높이는 방향으로 대사 재편이 일어났다.
PCA·PLS-DA·HCA 기반 1차 대사체 분석에서는 Burkholderia 처리 시 TCA 회로, 아미노산 대사, 탄수화물 대사, 산화환원 관련 대사가 폭넓게 재편되는 것이 확인되었다. 특히 고농도 Burkholderia 처리에서는 GABA, ornithine, 5-oxoproline 등 스트레스·적응 관련 대사가 강하게 활성화되어 대사적 프라이밍(metabolic priming)이 유도되었다. 반면 저농도 처리에서는 초기 변화 후 7일차에 대사 항상성이 회복되는 경향을 보였다.
종합적으로, Burkholderia sp. O33은 병원균 억제뿐 아니라 상추의 에너지·질소 대사 및 산화환원 균형을 향상시키는 등 식물의 대사 기반 방어력과 생육을 동시에 강화하는 효과를 나타냈다. 본 연구는 Burkholderia sp. O33이 상추 재배에서 지속 가능한 미생물 기반 병해 관리 소재로 활용될 수 있음을 체계적으로 제시하였다.

In this study, the antifungal activity of Burkholderia sp. O33 against Botrytis cinerea was evaluated, the physiological and metabolomic responses of lettuce following foliar application of Burkholderia were comprehensively analyzed. In vitro assays demonstrated that Burkholderia exhibited strong, concentration-dependent inhibition of B. cinerea, with effective suppression observed at ≥5.9 × 10⁹ CFU/mL. Seed germination tests further confirmed that no phytotoxicity occurred within the concentration range of ≤5.9 × 10¹⁰ CFU/mL, validating the applicability of Burkholderia to plant systems. In port-based foliar application trials, both fludioxonil and Burkholderia treatments improved lettuce growth, particularly in fresh weight and leaf number, suggesting a combined contribution of disease suppression and microbe-induced growth promotion. Detached leaf assays further revealed that high-density Burkholderia treatment reduced lesion expansion immediately after inoculation, confirming a short-term antagonistic effect of viable bacterial cells. Metabolomic analyses revealed clear treatment-dependent differences in physiological and metabolic responses. Sterol profiling indicated that Burkholderia induced an initial increase followed by rapid normalization, whereas fludioxonil caused sustained reductions in sterol content. Fatty acid profiling showed that fludioxonil increased saturated fatty acids while decreasing polyunsaturated fatty acids, whereas Burkholderia induced the opposite trend, shifting lipid metabolism toward enhanced membrane fluidity and stress adaptability. Primary metabolite profiling based on PCA, PLS-DA, and HCA revealed broad restructuring of the TCA cycle, amino-acid metabolism, carbohydrate metabolism, and redox-associated pathways following Burkholderia treatment. In particular, high Burkholderia density strongly activated stress- and adaptation-related metabolites such as GABA, ornithine, and 5- oxoproline, indicating metabolic priming. In contrast, low-density treatments produced transient alterations followed by a return to metabolic homeostasis by day 7. Overall, Burkholderia sp. O33 enhanced both disease suppression and metabolic resilience in lettuce by improving energy and nitrogen metabolism as well as redox balance. This study provides systematic evidence that Burkholderia has strong potential as a sustainable, microbe-based disease management agent for lettuce production.
