급격한 기후변화와 국제교류의 증가로 산림 병해충 피해의 종류와 규모가 증가하고 있으며 이는 산림의 건강성과 생태계 균형에 위협이 되고 있다. 효과적인 병해 관리 체계를 마련하기 위해서는 병원균의 생물학적, 생태학적 특성에 대한 정확한 이해를 돕는 과학적 정보 축적이 반드시 선행되어야 한다. Taphrina 속 곰팡이는 전 세계 주요 과수 및 산림 수종의 열매, 잎, 가지에 기형과 조직 비대를 일으켜 활력과 생산성을 저해하는 주요한 식물병원성 곰팡이 중 하나이다. 우리나라에는 현재까지 9개의 수종에서 Taphrina 속 병원균 8종의 발생이 기록되어 있다. 그러나 이러한 기록의 대부분은 병징 발현 모습, 병원균의 형태적, 유전적, 생리적 특성 등에 대한 체계적 분석 없이 국외 문헌에 의존한 결과이며 구체적 발생 자료와 병원균 동정 근거는 실질적으로 매우 부족한 실정이다. 특히 자두나무 주머니병은 과수 수확량과 품질을 저하시키는 중요한 병해임에도 아직까지 충분히 연구되지 않았으며 병원균 규명에 관한 기존 연구 또한 재검토가 필요하다. 따라서 본 연구에서는 우리나라에서 자생하거나 재배되고 있는 벚나무속(Prunus) 4종에서 주머니병을 일으키는 Taphrina 속 병원균의 형태학적, 분자생물학적, 생리학적 특성을 종합적으로 분석하고 분류학적 위치를 규명하였다. 형태적 특징으로는 자낭과 병세포, 자낭포자, 출아포자의 모양과 크기를 비교 분석하였다. 분자생물학적 계통분석에서는 핵 리보솜 DNA(Ribosomal DNA, rDNA)의 비번역 구간인 Internal Transcribed Spacer (ITS) 영역과 리보솜 대단위체 RNA 유전자 (Large subunit ribosomal RNA gene, LSU)의 D1/D2 도메인(domain), 그리고 미토콘드리아 DNA(mtDNA)의 리보솜 소단위체 RNA(SSU rRNA, rns) 등 3개 유전자 영역을 기반으로 다중 유전자 염기서열(multi-locus sequence) 분석을 수행하였다. 생리학적 특성 검정에는 효모류 분류에 사용하는 탄소원 및 질소원 동화능력, 비타민 요구성, 요소 가수분해 능력, 전분 형성 능력, 항생제 내성, Diazonium Blue B(DBB) 반응을 조사했다. 그 결과 우리나라 벚나무속 핵과류 수목에서 주머니병을 일으키는 원인균으로는 Taphrina neopruni sp. nov., T. haraeana sp. nov., 그리고 T. padi 가 확인되었다. T. neopruni sp. nov. 와 T. haraeana sp. nov.는 본 연구를 통해 신종 병원균으로 보고한다. 이 연구의 결과는 국내 벚나무속 핵과류 주머니병균의 다양성을 체계적으로 이해하는데 기여할 수 있고 Taphrina 속 병원균에 의한 병해의 정확한 진단 및 효과적인 관리 방안 마련뿐만 아니라 국가 간 식물검역을 위한 과학적 근거로 활용될 것으로 기대된다.

Rapid climate change and increasing international trade have increased the diversity and severity of forest diseases and insect pest outbreaks, posing growing risks to forest health and ecosystem stability. Among the causative agents, species of Taphrina, which induce malformation and tissue hypertrophy in a wide range of fruit and forest trees, constitute an important group of plant-pathogenic fungi. Although eight Taphrina species have been reported from nine host tree species in South Korea, most records lack detailed morphological, molecular, and physiological evidence. In this study, we investigated Taphrina species associated with fruit pocket disease on four Prunus hosts native to or cultivated in Korea. Their taxonomic placement was clarified using morphological characterization and multi-locus phylogenetic analyses. Physiological traits were further examined through standard yeast taxonomic tests, including assimilation of diverse carbon and nitrogen sources, vitamin requirements, urea hydrolysis, starch formation, antibiotic resistance, the diazonium blue B (DBB) color reaction, and growth responses across different temperatures. These integrated analyses identified Taphrina neopruni sp. nov., T. haraeana sp. nov., and T. padi as the causal agents of fruit pocket diseases on Korean Prunus species. Especially, T. neopruni and T. haraeana were described as new species and T. padi is the first report in Asia. Overall, our findings refine the current understanding of the diversity of Taphrina species in Korea and provide essential information for accurate disease diagnosis and the development of effective management strategies for Taphrina associated diseases.
