연골은 무혈관성, 무림프성, 낮은 세포 밀도를 가진 조직으로 자체적인 조직 재생이 제한적이므로 손상 시 재생을 위해서는 치료적 요법이 요구되는 조직이다. 연골 조직 재생을 위해 결손 부위에 이식되는 하이드로겔은 하중을 견뎌야 하기에 높은 기계적 강도와 연골의 특성을 모방하기 위한 점탄성을 가져야 하지만 높은 기계적 강도를 가지는 지지체의 경우 대체적으로 낮은 탄성 특성을 나타낸다. 젤라틴 메타크릴로일은 높은 기계적 강도를 가지지만 연골 재생용 지지체로 사용하기에는 탄성 계수가 낮아 한계가 존재한다. 이를 보완하기 위해 세포 외 기질의 주 구성성분인 히알루론산을 첨가하였고 이와 함께 연골 재생에 기여하는 폴리데옥시리보뉴클레오타이드를 첨가하였다. 히알루론산의 낮은 기계적 물성을 보완하기 위해 산화 히알루론산과 아디프산 디히드라자이드 개질 히알루론산으로 개질하여 쉬프 염기 반응 및 젤라틴 메타크릴로일의 광가교를 통한 이중 가교 하이드로겔을 제조하였다. 또한 PDRN의 높은 초기 방출을 제어하고 지속적인 약물 방출을 유도하기 위해 키토산과의 정전기적 인력을 이용한 PDRN-폴리플렉스를 합성하여 하이드로겔 지지체에 탑재하였다.
제조된 하이드로겔 지지체와 PDRN-폴리플렉스의 화학적, 물리학적 특성은 NMR, FT-IR, 팽윤율, 분해율 및 기계적 강도 분석을 통해 평가되었으며 그 결과 GelMA@HA-HYD 하이드로겔은 GelMA 하이드로겔보다 향상된 점탄성 특성을 보이며 기계적 강도가 유지되는 것을 확인하였다. 또한 PDRN-폴리플렉스를 하이드로겔 지지체에 탑재하고 동일 농도의 PDRN과의 약물 방출 프로파일을 분석한 결과 PDRN-폴리플렉스가 초기 급격한 방출을 제어할 수 있는 것을 확인하였다. 최종적으로 hAMSCs를 활용한 세포 분화 실험에서 히알루론산이 첨가된 GelMA@HA-HYD 하이드로겔과 폴리플렉스를 탑재한 GelMA@HA-HYD@PP 하이드로겔은 SOX9, AGG, COL2와 같은 연골 특이적 유전자에 대해 대조군보다 효과적인 연골 세포 분화도를 보이며 연골 재생용 하이드로겔 지지체로의 적용 가능성을 확인하였다.

Due to its avascular, alymphatic nature, and low cellular density, arcticular cartilage has limited self-regenerative capacity, necessitating therapeutic interventions for effective repair upon injury. Hydrogel scaffolds ,implanted into defect sites to support cartilage regenerataion, require sufficient mechanical strength to withstand physiological loads and viscoelastic properties that mimic native cartilage. However, scaffolds with high mechanical strength often display compromised elastic characteristics. While gelatin methacryloyl(GelMA) is a widely researched biocompatible material offering high mechanical strength, it shows a relatively low elastic modulus that limits its application for cartilage regeneration.
In this study, we used a Hyaluronic acid, a major component of the extracellular matrix(ECM), to enhance the elastic modulus of GelMA hydrogel and mimic the microenvironment of ECM. Along with Polydeoxy ribonucleotide(PDRN) to promote cartilage regeneration. To overcome the low mechanical properties of Hyaluronic acid, Oxidized Hyaluronic acid(OHA) and Adipic dihydrazide modified hyaluronic acid(HA-ADH) were synthesized to fabricate a dual -crosslinked hydrogel with Gelatin Methacryloyl via the dynamic Schiff base reaction between OHA and HA-ADH and photo-crosslinking of GelMA. Furthermore, to control the initial burst release of PDRN and induce sustained drug release, PDRN-Polyplexes were synthesized using electrostatic interactions with Chitosan and loaded into the hydrogel scaffold.
The physicochemical properties of the fabricated hydrogels and PDRN-Polyplexes were evaluated using NMR, FT-IR, swelling ratio, degradation rate, and mechanical strength analysis. The results indicate that the GelMA@HA-HYD hydrogel exhibited improved viscoelastic properties compared to pure GelMA hydrogels while maintaining mechanical integrity. Additionally, analysis of drug release profiles shows that the PDRN-Polyplex can control the initial burst release effectively compared to free PDRN. Finally, in vitro differentiation studies and RT-PCR using hAMSCs, confirming the expression of cartilage-specific genes such as SOX9, AGG, and COL2, indicate that the GelMA@HA-HYD hydrogel and the Polyplex-loaded GelMA@HA-HYD hydrogel significantly enhanced chondrogenic differentiation compared to the control group. Consequently, these findings suggest the potential of the developed hydrogel system as an effective scaffold for cartilage tissue engineering.
