    보상행동은 생존과 적응을 위해 진화적으로 보존되어 온 가장
    근본적인 신경생물학적 메커니즘 중 하나로, 동기부여, 목표 지향적
    행동 및 의사결정 과정을 뒷받침한다. 그러나 이러한 보상 회로는
    다양한 정신질환에서 이중적으로 이상 조절되며, 과활성화될 경우
    중독과 같은 병적 보상추구 행동을 유발하고, 저활성화될 경우
    무쾌감증과 의욕저하 등 우울증의 핵심 증상으로 나타난다. 본 연구는
    중독과 동기조절에서 핵심적인 역할을 하는 측좌핵(nucleus accumbens,
    NAc)의 두 가지 주요 기전을 규명하는 데 목적이 있다.
    첫째, 강력한 중추신경 자극제인 코카인 중독에서 TRPA1 이온
    채널의 역할을 규명하였다. 기존 연구는 TRPA1이 통각과
    말초신경계에서의 기능에 주로 초점을 맞췄으나, 최근 중추신경계
    전반에 걸친 광범위한 발현과 신경활동 조절 가능성이 제시되고 있다.
    본 연구에서는 TRPA1 채널이 코카인에 의한 흥분성 시냅스 전달 및

    The neurobiological substrates underlying reward processing and motivational behavior constitute fundamental mechanisms for understanding the pathophysiology of substance use disorders and mood disorders. Despite their distinct clinical phenotypes, these conditions exhibit convergent neural circuitry and shared molecular signaling cascades. Within the reward circuit, the nucleus accumbens (NAc) serves as a critical computational hub that translates motivational salience into behavioral output. Through the integration of diverse neurotransmitter systems—including dopaminergic, glutamatergic, GABAergic, and cholinergic inputs—the NAc orchestrates reward-related behaviors and represents a key neuroanatomical locus implicated in psychiatric disorders characterized by dysregulated motivation and affective processing.

    This dissertation elucidates two previously underexplored molecular mechanisms within the NAc that govern reward-related behavior: the role of the transient receptor potential ankyrin 1 (TRPA1) channel in substance uses disorders, and cholinergic signaling in depression-related motivational anhedonia.
    In the context of substance use disorders, targeted knockdown of the cation channel TRPA1 within the NAc produced significant attenuation of cocaine-induced addictive behavior as long as cocaine-induced excitatory synaptic transmission. These findings implicate TRPA1 as a critical modulator of glutamatergic reward signaling, potentially through regulation of D1 dopamine receptor-expressing medium spiny neurons (MSNs) and their downstream effector pathways.

    To investigate cholinergic regulation of motivational processes, intra-NAc micro-infusion of the non-selective acetylcholine receptor agonist carbachol resulted in decreased willingness to exert effort for high-value rewards, as quantified using a progressive ratio reinforcement schedule. Subsequent electrophysiological characterization revealed that this behavioral phenotype could be recapitulated through selective activation of M1 muscarinic receptors and was mediated by bidirectional modulation of excitatory and inhibitory synaptic transmission onto MSNs, thereby establishing a mechanistic framework for acetylcholine-induced suppression of motivational drive.

    Collectively, these investigations reveal novel molecular determinants within NAc circuitry that regulate distinct dimensions of reward processing and motivational behavior. The identification of TRPA1 as a modulator of cocaine reward and cholinergic signaling as a regulator of effort-based motivation advances our mechanistic understanding of the neurobiological foundations of addiction and mood disorders, while simultaneously identifying promising therapeutic targets for pharmacological intervention. These findings highlight the functional heterogeneity of NAc microcircuits and demonstrate how discrete signaling pathways within a single brain region can contribute differentially to the spectrum of neuropsychiatric conditions characterized by dysregulated reward processing and motivational dysfunction.
