타액선은 복잡한 신경 및 분자 기전을 통해 타액 분비를 조절함으로써 구강 및 전신 건강 유지에 중요한 역할을 한다. G 단백질 결합 수용체 (G protein–coupled receptors, GPCRs)는 신경 전달, 체액 분비, 혈관 반응 및 면역 반응을 조절함으로써 타액선 생리에서 핵심적인 조절자로 작용한다. 그러나 타액선에서 GPCR의 전반적인 발현 양상과 기능적 역할은 아직 명확히 규명되지 않았다.
본 연구에서는 타액선 기능 조절의 핵심인 신경–타액선 간 신호 전달 경로에 작용하는 GPCR의 발현을 확인하고자 하였다. 이를 위해 인간과 생쥐의 악하선에서 생성된 공개 단일세포 전사체 분석 (single-cell RNA sequencing, scRNA-seq) 데이터 (Gene Expression Omnibus, GEO: GSE199209 및 GSE175649)를 Seurat R 패키지를 이용해 분석하였다. 데이터 전처리 및 균일 매니폴드 근사 투영 (Uniform Manifold Approximation and Projection), t-분포 확률적 이웃 임베딩 (t-distributed Stochastic Neighbor Embedding)을 이용한 차원 축소 후, 기존의 세포 유형 마커를 기반으로 클러스터의 세포 유형을 분석하였다.
분석 결과, 무스카린 아세틸콜린 수용체, 아드레날린 수용체, P2Y2 퓨린 수용체, 장관 혈관 작용성 펩타이드 수용체 2, 타키키닌 수용체 3, 브래디키닌 수용체 2, 엔도텔린 수용체 A/B, 히스타민 수용체 1, 아연 감지 수용체, 아데노신 수용체 (adenosine receptors, ARs), 그리고 오렉신 수용체 1 등 다양한 GPCR이 세포 유형 특이적으로 발현함을 확인하였다.
특히, 본 연구는 scRNA-seq 데이터베이스를 기반으로 타액선에서 ARs의 발현을 최초로 보고하였다. Ars 의 생리적 기능을 규명하기 위하여 인간 악하선 유래 A253 세포에서 기능적 실험을 수행하였다. 세포를 ARs 작용제인 에틸카르복사미도아데노신으로 자극하고, 포스포디에스터레이스 억제제인 이소부틸-메틸잔틴을 함께 처리한 결과, 세포 내 cAMP 농도가 유의하게 증가하였으며 이는 A2B 수용체를 통한 Gs 단백질 연결 신호 전달 경로가 활성화되었음을 시사한다.
이러한 결과는 타액선 기능과 관련된 GPCR의 발현 양상에 대한 통찰을 제공하며, ARs의 발현 및 A2B 수용체에 의한 세포 내 cAMP 증가를 처음으로 규명하였다. 본 연구는 쇼그렌증후군 및 구강건조증과 같은 타액선 관련 질환에서 아데노신 수용체를 치료 표적으로 활용할 수 있는 가능성을 제시한다.

Salivary glands play a critical role in maintaining oral and systemic health by regulating salivary secretion through complex neural and molecular mechanisms. G protein-coupled receptors (GPCRs) are essential regulators of salivary gland physiology, modulating neural communication, fluid secretion, vascular responses, and immune activity. Despite their importance, the comprehensive expression landscape and functional roles of GPCRs in salivary glands have not been fully elucidated.
This study aimed to identify the expression of GPCRs, which play a pivotal role in salivary gland–nerve communication, a key process essential for the regulation of salivary gland functions. We analyzed publicly available single-cell RNA sequencing (scRNA-seq) datasets from human and mouse submandibular glands (GEO: GSE199209 and GSE175649) using the Seurat R package. After quality control and dimensionality reduction using Uniform Manifold Approximation and Projection and t-distributed Stochastic Neighbor Embedding, clusters were annotated based on established cell-type markers. These transcriptomic results revealed the expression of the diverse GPCR subtypes—including muscarinic acetylcholine receptors, adrenergic receptors, P2Y2 purinergic receptor, vasoactive intestinal peptide receptor 2, tachykinin receptor 3, bradykinin receptor 2, endothelial receptor A and B, histamine receptor 1, ZnR/GPR39, adenosine receptors (ARs) and orexin receptor 1—with distinct cell-type specificity.
Importantly, we report for the first time the expression of adenosine receptors in salivary glands based on scRNA-seq database. To confirm this finding and characterize the physiological functions, we performed functional assays in A253 human submandibular gland cells. Cells were stimulated with 10 μM 5'-N-Ethylcarboxamidoadenosine, an adenosine receptor agonist, in the presence of 1 mM isobutylmethylxanthine. Intracellular cAMP concentrations were significantly elevated, indicating active Gs-coupled adenosine signaling, likely mediated by A2B-type receptors.
Together, these findings provide the insights into the expression profile of GPCRs involved in salivary gland function. We report, for the first time, the expression of and adenosine receptors in SMG. Furthermore, we demonstrate that the A2B receptor elicits a physiological response by increasing intracellular cAMP levels. These findings provide a foundation for the potential therapeutic targeting of adenosine receptors in salivary gland–related disorders, such as Sjögren’s syndrome and xerostomia.
