    Taste perception is a vital sensory modality that guides organisms toward nutritious substances and away from harmful compounds, playing an essential role in both survival and hedonic behavior. While recent advances have identified taste receptors and elucidated key components of the gustatory signaling pathway, a fundamental limitation persists: current methodologies cannot simultaneously capture the functional responses and molecular identities of individual taste cells within their native spatial context. This disconnect impedes a comprehensive understanding of how taste qualities are encoded at the cellular level. To address this challenge, we developed a correlative imaging platform that integrates in vivo functional imaging with post hoc molecular profiling at a single-cell resolution. We performed calcium responses of individual taste cells to defined taste stimuli under intact physiological conditions. Spatial indexing via near-infrared branding and fluorescent reporters enabled precise re-identification of the same cells in subsequent histological procedures. Functional and molecular data were subsequently registered using an image alignment pipeline, enabling single-cell resolution mapping of taste cell function to transcriptomic identity within the intact taste bud. This integrated correlative approach provides a novel framework for decoding the functional and molecular architecture of the peripheral taste system. It opens new avenues for understanding how spatial organization, cell-type diversity, and intercellular interactions contribute to the encoding of taste signals.
