    소 유전자 공학은 장기간의 임신 기간, 제한된 수정란 수, 그리고 높은 유지 비용과 같은 고유한 한계에 직면해 있다. 이러한 제약을 극복하기 위해, 최적화된 트랜스포존 시스템, 생식세포를 통해 유전자가 전달되는 Cas9 발현 형질전환 소, 배아줄기세포 유사 세포(ESC-like cells), 그리고 조직 유래 오가노이드 모델을 결합한 통합 플랫폼이 구축되었다. 세 가지 비바이러스성 트랜스포존 시스템(PiggyBac, Sleeping Beauty, Tol2)을 체계적으로 평가한 결과, PiggyBac(PB)은 소 체세포에서 98.4%의 형질전환 효율과 63.9%의 안정적인 유전자 통합 효율을 보여 우수한 성능을 나타냈으며, Sleeping Beauty(SB)와 Tol2는 배아 내에서 더 높은 GFP 발현을 보였다. 그러나 배 발달 능력을 종합적으로 고려했을 때, PB 시스템이 체세포와 수정란 모두에서 가장 최적의 유전자 도입 수단으로 평가되었다.

    이러한 Cas9 발현 자원의 활용도를 확장하기 위해 체세포 및 생식세포 기반 편집 플랫폼이 추가로 평가되었다. Cas9 발현 섬유아세포를 이용한 체세포 유전자 편집은 높은 돌연변이 유도 효율과 함께 체외에서의 발달 능력을 유지했으며, 이는 체세포핵이식(SCNT) 후 형성된 포배 발달을 통해 확인되었다. 또한 Cas9 발현 정자는 유전자 제거(knockout)와 삽입(knock-in) 모두에 활용 가능하였고, 소혈청알부민(BSA) 좌위에 대해 최대 54.2%의 표적 삽입 (knock-in) 효율을 달성하였다.

    Genome engineering in cattle faces inherent challenges such as prolonged gestation, limited embryo yield, and high maintenance costs. To address these constraints, a comprehensive platform has been established combining optimized transposon systems, germline-transmissible Cas9-expressing cattle, embryonic stem-like (ESC-like) cells, and tissue-derived organoid models. Three non-viral transposon systems—PiggyBac (PB), Sleeping Beauty (SB), and Tol2—were systematically evaluated. PiggyBac (PB) exhibited superior transfection (98.4%) and stable integration (63.9%) efficiencies in bovine somatic cells, whereas Sleeping Beauty (SB) and Tol2 showed higher GFP expression in embryos. However, when considering overall embryonic developmental competence, PB was determined to be the most optimal system across both somatic cells and embryos.
    Based on these findings, the PB system was selected for in vivo application, and Cas9-expressing cattle were generated through cytoplasmic microinjection of PiggyBac-based (PB-based) all-in-one vectors. Transgene integration and expression were verified by genomic polymerase chain reaction (PCR), reverse transcription PCR (RT-PCR), Western blotting (WB), and fluorescence-based analyses (FBA). Germline transmission was confirmed in both IVF-derived and naturally bred offspring, with successful editing of the PRNP locus.
    To further expand the applicability of these Cas9-expressing resources, both somatic and germline platforms were subsequently evaluated. Somatic cell-based gene editing using Cas9-expressing fibroblasts demonstrated high mutation efficiencies and preserved developmental competence, as evidenced by blastocyst formation following somatic cell nuclear transfer (SCNT). Cas9-expressing sperm enabled both knockout and knock-in editing, achieving targeted knock-in at the bovine serum albumin (BSA) locus with up to 54.2% efficiency.
    Building upon these successful editing outcomes, downstream derivation and characterization of stem and trophoblast cell lineages were performed. Importantly, embryonic stem-like cells (ESC-like cells) were successfully derived from edited blastocysts. These ESC-like cells maintained stable transgene expression, pluripotency marker SOX2, and Cas9 protein across multiple passages, confirming their utility for downstream functional studies and regenerative applications. Additionally, trophoblast cell cultures were established from genome-edited embryos and maintained under defined conditions, providing a tractable in vitro system for studying placental development and gene function.
    To extend this platform to tissue-derived models, gene editing capabilities were also assessed in organoid systems. Bovine endometrial organoids (EOs) supported efficient ex vivo gene editing. Lentiviral vectors encoding single guide RNAs (sgRNAs) targeting PRNP and BLG achieved indel frequencies of 73% and 59%, respectively, validating CRISPR-mediated functional studies in a 3D epithelial context.
    Altogether, a comprehensive platform has been established combining optimized transposon systems, germline-transmissible Cas9-expressing cattle, embryonic stem-like (ESC-like) cells, and tissue-derived organoid models. This modular genome engineering system offers scalable, inheritable, and cell-type adaptable tools to advance bovine genetic studies, disease modeling, and reproductive biotechnology.
