목적 : 본 연구는 적층가공 방식으로 제작된 의치상 수지의 기계적, 표면적, 생물학적 특성에 대한 제조 조건 및 후중합 조건의 영향을 평가하고자 하였다. 이를 위해 세 가지 개별 실험을 수행하였다. 첫 번째 실험에서는 수지의 점도, 적층 두께(layer thickness, LT), 출력 각도(build angle, BA)가 표면 특성과 미생물 부착에 미치는 영향을 분석하였다. 두 번째 실험에서는 동일한 인자들이 기계적 물성과 진도(trueness)에 미치는 영향을 평가하였다. 세 번째 실험에서는 단일 고점도 수지를 이용하여 후중합 시간과 후중합 대기 조건이 굴곡 강도, 비커스 경도, 중합도(degree of conversion, DC), 세포 독성, 잔류 단량체 방출에 미치는 영향을 조사하였다.
방법 : 첫 번째 실험에서는 점도가 상이한 두 종류의 적층가공 치과용 수지(고점도 및 저점도)를 사용하여, 적층 두께(50 및 100 μm)와 출력 각도(0°, 45°, 90°)를 조합한 디스크 형태의 시편을 제작하였다. 각 시편의 표면 거칠기 및 접촉각(n = 10)을 측정하였으며, Streptococcus oralis와 Candida albicans에 대한 미생물 부착은 흡광도(n = 6)를 통해 정량적으로 평가하였다. 통계 분석은 3요인 분산분석(three-way ANOVA) 및 사후 검정을 통해 수행하였고, 흡광도 자료에 대해서는 정렬 순위 변환(Aligned Rank Transform, ART) 기반의 비모수 분산분석을 적용하였다. 두 번째 실험에서는 동일한 수지와 제작 조건을 적용하여 디스크형, 바형, 의치상형 시편을 제작하였고, 각각의 비커스 경도(n = 10), 굴곡 강도(n = 10), 진도(trueness, n = 10)를 측정하였다. 세 번째 실험에서는 고점도 수지만을 이용하여, 후중합 대기조건(postpolymerization atmosphere; PPA, 공기 및 질소)과 후중합 시간(postpolymerization time; PPT, 5, 10, 20분)을 조합한 여섯 가지 조건 하에서 시편을 제작하였다. 이후 굴곡 강도(n = 15), 비커스 경도(n = 15), 중합도(DC, n = 10), 세포독성(n = 6), 잔류 단량체 함량(n = 5)을 각각 측정하였고, FTIR, MTT assay, HPLC를 이용하여 분석하였다. 모든 결과는 정규성 검정을 거친 후, 비모수 요인분석을 통해 통계 처리되었다(α = 0.05).
결과 : 시편의 표면 거칠기와 접촉각은 적층 두께와 출력 각도에 의해 유의하게 영향을 받았으며, 이러한 영향은 수지의 점도에 따라 달라지는 경향을 보였다(P < 0.001). 미생물 부착에 대해서는 3요인 간의 상호작용은 유의하지 않았으나(P > 0.05), 점도 × 출력 각도 및 적층 두께 × 출력 각도 간의 2요인 상호작용은 통계적으로 유의하였다(P < 0.05).
비커스 경도는 수지 점도, 적층 두께, 출력 각도 간의 3요인(P = 0.027) 및 2요인 상호작용에 의해 유의한 영향을 받았으며, 저점도 수지가 고점도 수지보다 일관되게 높은 경도 값을 나타냈다(P < 0.001). 굴곡 강도는 적층 두께(P = 0.003)와 출력 각도(P < 0.001)의 주효과에 의해 유의하게 영향을 받았으나, 변수 간의 상호작용은 유의하지 않았다(P = 0.212). 진도는 점도와 출력 각도 간의 상호작용에서 유의한 차이를 보였으며(P = 0.001), 45° 및 90° 출력 각도에서 저점도수지는 더 우수한 정확도를 나타냈다(P < 0.001).
후중합 조건에서는 비커스 경도에 대해 후중합 대기조건, 후중합 시간 간의 상호작용이 유의하게 나타났다(P < 0.001). 반면, 굴곡 강도, 중합도(DC), 세포독성에서는 유의한 상호작용이 관찰되지 않았다(P = 0.826, P = 0.786, P = 0.563). 질소 대기조건에서 후중합 시간이 길어질수록 경도는 유의하게 증가하였다(P < 0.001). 중합도는 후중합 대기조건에 따라 유의한 차이를 보였으나(P = 0.012), 굴곡 강도 및 세포독성에는 유의한 영향을 미치지 않았다(P = 0.500, P = 0.299). 후중합 시간은 세포독성에 유의한 영향을 주었으나(P < 0.001), 굴곡 강도와 중합도에는 영향을 주지 않았다(P = 0.482, P = 0.167). 후중합 10분 이후에는 잔류 단량체가 검출되지 않았다.
결론 : 표면 거칠기는 출력 각도가 0°일 때 가장 낮았으며, 이는 수지의 점도와 적층 두께와 무관하게 일관된 경향을 보였다. 적층 두께가 50 μm인 시편에서는 출력 각도 45° 조건에서 접촉각이 가장 작았으며, 수지 점도에 관계없이 동일한 결과를 나타냈다. Streptococcus oralis의 부착은 모든 조건에서 0° 출력 각도에서 최소화되었으며, Candida albicans의 부착은 수지 점도가 높고 적층 두께가 작을수록 감소하였다. 기계적 특성과 진도는 제작 조건의 영향을 받았으며, 낮은 점도의 수지, 50 μm 적층 두께, 45° 출력 각도를 조합한 조건에서 높은 경도와 진도가 확인되었다. 또한, 질소 대기조건에서 10분 이상 후중합을 수행할 경우 경도와 중합도가 향상되고 세포독성과 잔류 댠량체가 효과적으로 제거되어, DLP 방식으로 제작된 의치상 수지의 임상 적용에 적합함을 시사하였다.
임상적 의의 : 본 연구는 수지 점도, 적층 두께, 출력 각도, 후중합 조건과 같은 제조 인자들을 임상 상황에 따라 조절함으로써, 적층가공 방식의 의치 베이스 제작을 최적화할 수 있도록 임상적으로 유의미한 지침을 제시한다. 구강 감염에 취약한 환자의 경우, 고점도 수지를 사용하고 50 μm 적층 두께 및 0도 출력 각도로 출력하는 것이 미생물 부착을 줄이는 데 도움이 될 수 있다. 반대로, 우수한 물성 및 높은 치수 정확도가 요구되는 경우에는 저점도 수지와 50 μm 적층 두께, 45도 출력 각도의 조합이 권장된다. 특히, 질소 분위기에서 최소 10분 이상 후중합을 수행하는 것은 생체적합성을 높이고 임상적 안전성을 확보하는 데 효과적이다.

Purpose: This study aimed to investigate the effects of manufacturing and postpolymerization parameters on the mechanical, surface, and biological properties of dental polymers fabricated using additive manufacturing (AM). To achieve this, three separate experiments were conducted. In the first experiment, the effects of resin viscosity, layer thickness (LT), and build angle (BA) on surface characteristics and microbial adhesion were examined. The second experiment evaluated the influence of these parameters on mechanical properties and dimensional accuracy (trueness). In the third experiment, a single high-viscosity resin was used to assess the effects of postpolymerization time and atmospheric conditions on flexural strength, Vickers hardness, degree of conversion (DC), cytotoxicity, and residual monomer release.
Materials and methods: In the first experiment, disk-shaped specimens were printed using two AM resins of distinct viscosities (high and low), with LT values of 50 and 100 μm and BA settings of 0°, 45°, and 90°. Surface roughness and contact angle were measured (n = 10), and microbial adherence was quantitatively assessed based on absorbance measurements of Streptococcus oralis and Candida albicans (n = 6). Statistical analysis was performed using a three-way analysis of variance (ANOVA) and post hoc tests; for absorbance data, a nonparametric aligned rank transform (ART) ANOVA was used. In the second experiment, the same resins and manufacturing parameters were used to fabricate disk-, bar-, and denture base-shaped specimens for the evaluation of Vickers hardness (n = 10), flexural strength (n = 10), and trueness (n = 10). A three-way ANOVA was conducted with significance set at P < 0.05. In the third experiment, only a high-viscosity resin was used to fabricate specimens under six conditions defined by combinations of two postpolymerization atmospheres (air, nitrogen) and three postpolymerization times (5, 10, and 20 minutes). Flexural strength (n = 15) and Vickers hardness (n = 15) were tested, and DC (n = 10) was measured via Fourier transform infrared (FTIR) spectroscopy. Cytotoxicity (n = 6) was assessed using human gingival fibroblasts in an MTT assay, and residual monomer content (n = 5) was analyzed by high-performance liquid chromatography (HPLC). A nonparametric factorial ANOVA was applied after confirming non-normality using the Shapiro–Wilk test (α = 0.05).
Results: LT and BA significantly influenced the surface roughness and contact angle of the specimens, with these effects varying according to resin viscosity (P < 0.001). Although no significant three-way interaction was found in microbial adhesion data (P > 0.05), two-way interactions involving viscosity × BA and LT × BA were significant (P < 0.05).
Vickers hardness was significantly affected by three-way (P = 0.027) and two-way interactions involving resin viscosity, LT, and BA, with low-viscosity resin consistently exhibiting higher hardness than the high-viscosity counterpart (P < 0.001). Flexural strength was significantly influenced by the main effects of LT and BA (P = 0.003 and P < 0.001, respectively), while no significant interaction effects were observed (P = 0.212). Regarding trueness, a significant interaction between viscosity and BA was found (P = 0.001), and the low-viscosity resin demonstrated superior accuracy at 45° and 90° BAs (P < 0.001).
The postpolymerization parameters revealed a significant atmosphere × time interaction for hardness (P < 0.001), whereas strength, DC, and cytotoxicity showed no such interaction (P = 0.826, P = 0.786, P = 0.563). Under nitrogen, extended postpolymerization significantly increased hardness (P < 0.001). The atmosphere had a significant effect on DC (P = 0.012) but not on strength or cytotoxicity (P = 0.500 and P = 0.299, respectively). Postpolymerization time significantly influenced cytotoxicity (P < 0.001) but did not significantly affect flexural strength or DC (P = 0.482 and P = 0.167, respectively). The residual monomer was undetectable after 10 minutes of postpolymerization.
Conclusions: Surface roughness was lowest in disks fabricated at a 0° BA, regardless of resin viscosity or LT. For 50-μm LT specimens, those printed at a 45° BA exhibited the smallest contact angle, regardless of resin viscosity. The adhesion of S. oralis was minimized at 0° BA under all conditions. The adhesion of C. albicans was reduced by increased resin viscosity and decreased LT, both of which contributed to this effect. Mechanical properties and trueness were affected by the manufacturing parameters. The optimal parameters for achieving superior hardness and dimensional trueness included low-viscosity resin, a 50-μm LT, and a 45° BA. Postpolymerization under nitrogen for at least 10 minutes enhanced both hardness and DC. In addition, it effectively eliminated cytotoxicity and residual monomers, supporting its suitability for the clinical use of AM-fabricated denture bases.
Clinical significance: This study provides practical guidance for optimizing denture base fabrication through AM by adjusting parameters such as resin viscosity, LT, BA, and postpolymerization conditions according to clinical needs. For patients susceptible to oral infections, using a high-viscosity resin printed at a 50-μm LT and a 0° BA may help reduce microbial adhesion. Conversely, when superior mechanical properties and high dimensional accuracy are required, a low-viscosity resin combined with a 50-μm LT and a 45° BA is recommended. In particular, postpolymerization under a nitrogen atmosphere for at least 10 minutes effectively enhances biocompatibility and ensures clinical safety.
