    쉬나무(Tetradium daniellii (Benn.) T.G. Hartley)는 한국과 중국에 자생하며, 전통적으로는 약재로 이용되어 왔고, 우수한 화밀 분비량으로 인해 밀원수로서의 가치가 높다. 쉬나무속 식물은 에보디아민(evodiamine), 루테카르핀(rutaecarpine)과 같은 알칼로이드를 함유하고 있으며, 에보디아민은 강력한 항암 활성을, 루테카르핀은 항염증, 항비만, 심혈관 보호 효과를 나타내는 것으로 보고되었다. 그러나 이들 화합물은 주로 쉬나무속 오수유의 과실에서 추출되며, 환경 요인에 의해 함량 변동이 크고, 반복적인 채취 시 자원 고갈 우려가 있다. 따라서, 유용한 성분을 지속적이고 안정적으로 생산할 수 있는 시스템의 구축이 필요하다. 본 연구는 캘러스 배양을 통해 에보디아민과 루테카르핀의 함량을 증대시킬 수 있는 최적 배양 조건을 규명하고, 조직별 함량 차이를 정량적으로 비교하고자 하였다. 쉬나무의 기내배양 식물체 조직(잎, 엽병)과 성체 조직(잎, 엽병, 과실)을 Murashige and Skoog (MS), Woody Plant Medium (WPM), Driver and Kuniyuki Walnut (DKW), Schenk and Hildebrandt (SH) 배지에 치상하고, 명조건(16 h light/8 h dark) 및 암조건(24 h dark)에서 캘러스를 유도하였다. 유도된 캘러스와 조직은 Liquid Chromatography-Mass Spectrometry (LC-MS)를 통해 두 성분의 함량을 정량 분석하였다. 캘러스 유도 결과, 기내배양 식물체 조직에서 유도율이 높았고, WPM 배지에서는 조직과 광조건에 관계없이 100%의 유도율을 보였다. DKW 배지는 재분화율이 높았으며, SH 배지는 유도율과 생체량이 전반적으로 낮았다. 정량 분석 결과, MS 배지 명조건에서 배양한 기내배양 잎 유래 캘러스는 에보디아민 6.45 mg/kg, 루테카르핀 140.83 mg/kg으로, 가장 높은 함량을 나타냈으며, 미성숙 과실 대비 각각 1.2배, 5.7배 높은 함량을 나타냈다. 기내배양 잎 유래 캘러스를 명조건에서 배양한 경우, 암조건 대비 루테카르핀 함량이 약 4.2배 높았고, 광 조건이 루테카르핀 축적에 유의미한 영향을 미침을 확인하였다. 반면, 성체 조직은 캘러스 유도 과정에서 오염률이 높았고, 두 화합물의 함량도 미성숙 과실 대비 낮았다. 본 연구는 쉬나무에서 조직의 생리적 상태, 배지, 광 조건에 따른 캘러스 유도 특성 및 알칼로이드 축적 경향을 최초로 규명하였다. 특히 기내배양 식물체 잎을 MS 배지의 명조건에서 배양한 캘러스가 두 화합물의 함량 증대에 가장 효과적임을 입증하였다. 향후 질소원 조절, 전구체 공급, 고생산성 세포주 선별, 생합성 경로 유전자 발현 분석 등을 통해 조직배양 기반 쉬나무 알칼로이드의 대량생산 시스템으로 확장할 수 있을 것으로 기대된다.

    Tetradium daniellii (Benn.) T.G. Hartley, native to Korea and China, is valued as a melliferous tree due to its abundant nectar production and has also been traditionally utilized in herbal medicine. This species contains indole alkaloids such as evodiamine and rutaecarpine, with evodiamine exhibiting potent anticancer activity and rutaecarpine demonstrating anti-inflammatory, anti-obesity, and cardioprotective effects. However, these metabolites are primarily isolated from immature fruits of related Tetradium species, and their content fluctuates significantly depending on harvest time and environmental conditions, and repeated collection raises concerns regarding resource sustainability. Therefore, it is necessary to establish a system that can continuously and stably produce useful compounds. In this study, we aimed to optimize callus culture conditions to enhance the yields of evodiamine and rutaecarpine and to quantitatively compare their content across tissue types. Callus induction was performed using leaf and petiole explants from both in vitro-grown plantlets and mature trees, cultured on four basal media: Murashige and Skoog (MS), Woody Plant Medium (WPM), Driver and Kuniyuki Walnut (DKW), and Schenk and Hildebrandt (SH), under both light (16 h light/8 h dark) and dark (24 h dark) conditions. The induced calli and tissues (leaf, petiole, fruit) were analyzed for alkaloid content using liquid chromatography-mass spectrometry (LC-MS). Callus induction was most successful in explants derived from in vitro-grown plantlets, with no contamination observed. WPM medium achieved 100% induction regardless of tissue type or light conditions. DKW medium resulted in frequent redifferentiation, while SH medium exhibited generally low induction rates and limited proliferation of callus biomass. Quantitative analysis showed that callus derived from in vitro-grown leaves cultured on MS medium under light conditions accumulated the highest levels of both compounds: evodiamine at 6.45 mg/kg and rutaecarpine at 140.83 mg/kg. These values represent 1.2-fold and 5.7-fold increases, respectively, compared to those in immature fruit. For in vitro leaf-derived calli, exposure to light yielded approximately 4.2 times higher rutaecarpine content compared to dark conditions, indicating a significant effect of illumination on rutaecarpine accumulation. In contrast, callus induction from adult tissues resulted in higher contamination rates (30–60%) and lower alkaloid levels than those observed in immature fruits. This study provides the first comprehensive assessment of how tissue physiological state, basal medium, and light conditions affect callus induction and indole alkaloid accumulation in T. daniellii. The results demonstrate that callus derived from in vitro-grown leaves cultured on MS medium under light is the most effective for producing evodiamine and rutaecarpine. These findings provide a foundation for developing a tissue culture-based production system for functional alkaloids from T. daniellii and suggest future directions, including nitrogen source modulation, precursor supplementation, high-yield cell line selection, and gene expression analysis of the biosynthetic pathway.
