    중간엽 줄기세포는 중배엽에 속해있고 분화 상 중간 단계에 해당하기 때문에 특정한 세포로의 분화의 방향성을 가지고 있어 체내에서 재생성, 회복성, 분화능을 가지고 있는 비교적으로 안전한 줄기세포이며, 앞으로 세포 치료제로의 활용으로는 이와 같은 세포를 중점적으로 연구를 하는 것이 중요하다.
    첫번째 연구는 중배엽에 해당하는 중간엽 줄기세포(MSCs)을 이용하여 아토피 피부염을 치료하기 위해 세포치료제를 투여한 내용이다. 기존에는 면역조절 치료를 위해 상용화된 다른 상용화된 의약품과 비교하여 개 MSCs의 면역 조절 능의 비교는 잘 연구되지 않았다. 본 연구에서는 개 양막 중간엽 줄기세포 (cAM-MSCs)의 특성과 면역조절 효과를 조사했다. 특히, 활성화된 개 말초 혈액 단핵 세포(PBMCs) 증식으로부터 면역 조절 및 T 림프구의 유전자 발현을 조사했다. 그 결과, cAM-MSCs가 면역 조절 유전자(TGF-β1, IDO1 및 PTGES2)가 증가되고 T 세포의 증식 능력을 억제하는 것을 확인했다. 또한 개 아토피성 피부염(AD) 치료제로 가장 일반적으로 사용되는 JAK(Janus Kinase) 억제제인 OCL (oclacitinib)과 cAM-MSCs의 치료 효과를 마우스 AD 모델을 통해 확인했다. 그 결과, PBS만 처리한 그룹에 비해 MSCs 처리 그룹 (계대 4, 6, 8)을 사용한 cAM-MSCs는 피부과적 징후, 조직 병리학적 변화 및 염증성 사이토카인의 점수가 크게 감소했음을 확인했다. 특히, cAM-MSCs는 상처 기능 장애 회복, 비만 세포 활성 조절 및 면역 조절 단백질 발현 수준에서 OCL보다 효과적 이였다. 흥미롭게도, cAM-MSCs의 피하 주사는 체중이 회복되고 정상군과의 차이가 없었지만, OCL의 경구 투여는 부작용으로 체중 감소를 나타냈다. 결론적으로, 본 연구는 cAM-MSCs가 효과적인 재생과 면역조절을 통해 부작용 없이 안전한 개 아토피 피부염 치료제로 개발될 수 있음을 알 수 있었다.

    In stem cell research, multipotent stem cells differentiate from embryonic and pluripotent stem cells to construct intermediate-stage stem cells of ectoderm, mesoderm, and endoderm. Especially, mesenchymal stem cells (MSCs) regenerate and recover after the differentiation of a specific cell type and tissue. As stem cells have self-renewal and proliferation, it is important to focus on them for stem cell therapy.
    The first section discusses how mesoderm-derived MSCs are a potential treatment option for canine immunological diseases. The properties and immunomodulatory effects of canine amnion membrane (cAM)-MSCs were examined in this study. Gene expression of immune modulation and T lymphocytes were examined from activated canine peripheral blood mononuclear cells (PBMCs) proliferation. As a result, cAM-MSCs significantly upregulated immunomodulatory genes (including TGF-β1, IDO1, and PTGES2) and suppressed T-cell proliferation. Moreover, the therapeutic effect of cAM-MSCs was confirmed by comparison with oclacitinib (OCL), the most commonly used Janus kinase (JAK) inhibitor, as a treatment for canine atopic dermatitis (CAD) using a mouse atopic dermatitis (AD) model. As a result, cAM-MSCs was confirmed with phosphate-buffered saline (PBS) treatment groups (passage 4, 6 and 8) compared to PBS only though scores of dermatologic signs, tissue pathologic changes and inflammatory cytokines were significantly reduced. In particular, cAMMSCs was more effective than OCL in the recovery of wound dysfunction, regulation of mast cell activity and expression level of immune modulation protein. Notably, subcutaneous injection of cAMMSCs induced weight recovery, but oral administration of oclacitinib induced weight loss as a side effect. This study demonstrates that cAM-MSCs possess regenerative and immunomodulatory properties, offering a safe therapeutic option for CAD.
    In the second section, according to the previous study, the administration of cAM-MSCs is a potential therapy for immunological diseases. However, many of AD is a chronic and inflammatory disease. Unfortunately, most related studies involve short-term experiments and acute AD animal models. Thus, studies of repeated subcutaneous injection of cAM-MSCs for ameliorating long-term inflammatory skin disorders have not been established. In this study, the effects of long-term cAM-MSCs and calcineurin inhibitors (CNIs) treatments were evaluated in the mouse AD model for up to 8 weeks. Then, they compared the differences in therapeutic effect through canine PBMCs. Using a mouse DNCB-derived AD model, the therapeutic impact of cAM-MSCs was validated in comparison to pimecrolimus (Pime), the most widely used CNIs, as a therapy for CAD. Based on their results, the treatment with cAM-MSCs group exhibited substantially lower scores for tissue pathologic alterations, inflammatory cytokines, and dermatologic symptoms than the PBS control group. Notably, compared with Pime, cAM-MSCs were more effective at preventing wound dysfunction and regulating mast cell activity. Additionally, immune modulation proteins (TGF-β1, IDO1, and COX-2) were increased in the cAM-MSCs treatment group. Furthermore, the immunoregulatory effect of cAM-MSCs was examined through the proliferation of T lymphocytes from activated canine PBMCs. As a result, cAM-MSCs suppressed the proliferative capacity of effector T-cells from canine PBMCs more effectively than Pime. This study suggested that the cAM-MSCS could be an effective canine treatment for long-term CAD through regeneration and immunomodulation.
    In conclusion, cAM-MSCs are not only suitable for cell therapies in AD, but they are also safe for using companion animals as they have fewer adverse effects. In the future, cAM-MSCs will be a good research direction that can contribute to veterinary research and public health.
