Gαi 억제로 인해 Gαq 경로가 증폭된다는 관찰에 기반하여, HRH1 수용체를 통한 칼슘 신호를 민감한 readout으로 사용하는 분석법을 설계하였다. 본 플랫폼은 기존의 클러스터링 기반 또는 cAMP 기반 검출법에 비해 높은 특이도, 재현성, 고처리량 분석 적합성을 제공하며, 전사체 분석을 통해 사용된 세포주에서 HRH 계열 중 HRH1이 가장 높게 발현됨을 확인하여 타깃 선정의 근거를 마련하였다. 또한, HRH1이 Gαi와 Gαq 양쪽 모두에 결합할 수 있는 G 단백질 결합 다형성(coupling diversity)을 지닌다는 점이 분석 민감도 향상에 중요한 역할을 하였으며, 이는 수용체 발현 패턴과 신호전달 유연성을 동시에 고려한 진단 응용의 한 예로 작용하였다. 두 번째 연구에서는 Gαs 결합 수용체인 ADORA2B가 삼중음성 유방암(triple-negative breast cancer, TNBC)에서 저산소 적응 및 면역관문 조절에 중심적인 역할을 수행함을 규명하였다. 벌크 및 단일세포 RNA-seq, 3차원 종양 스페로이드 모델링, 기능적 차단 실험을 통합한 결과, ADORA2B–PKA–UBE2S–β-catenin–HIF-1α로 이어지는 자기 강화형 신호축이 확인되었으며, 이는 정상 산소 또는 간헐적 저산소 조건에서도 저산소 전사 프로그램을 지속시키는 기능적 회로로 작용한다. G protein-coupled receptors (GPCRs) constitute one of the most functionally versatile families of membrane proteins, mediating a wide spectrum of physiological responses through coupling with distinct classes of heterotrimeric G proteins. While traditionally studied in isolated signaling contexts, recent findings underscore the dynamic crosstalk and context-dependent modulation among different G protein pathways. This functional plasticity presents both opportunities and challenges in understanding GPCRs in diagnostic and pathological settings. Moreover, since a single ligand can activate multiple GPCR subtypes, accurate characterization of both ligand abundance and receptor expression patterns is essential to deciphering the biological outcome of GPCR signaling in a given context. This dissertation presents two distinct but conceptually connected studies that leverage the contextual nature of GPCR signaling in both diagnostic assay development and oncogenic pathway analysis. The first study establishes a functional calcium-based assay for pertussis toxin (PTx) detection by exploiting the crosstalk between Gαi and Gαq signaling. Building on observations that Gαi inactivation enhances calcium signaling through Gαq-coupled receptors, I developed a sensitive and reproducible assay using HRH1-mediated calcium responses as a quantitative readout. This platform improves upon traditional clustering or cAMP-based assays, offering enhanced specificity, reproducibility, and high-throughput compatibility. Transcriptomic profiling identified HRH1 as the dominantly expressed histamine receptor in the assay system, validating its selection. Equally critical was the recognition that HRH1 can couple to both Gαi and Gαq, enabling the deliberate use of G protein coupling diversity to amplify assay performance—demonstrating how receptor expression and signaling flexibility can be jointly harnessed in diagnostic contexts. The second study investigates the role of ADORA2B, a Gαs-coupled receptor enriched in triple-negative breast cancer (TNBC), in promoting hypoxia-adaptive transcriptional programs and immune checkpoint regulation. Multi-omics analyses—including bulk and single-cell RNA-seq, 3D tumor spheroid modeling, and functional perturbation—identified a self-reinforcing ADORA2B–PKA–UBE2S–β-catenin–HIF-1α axis that sustains hypoxic signaling even under normoxic or fluctuating oxygen conditions. ADORA2B was found to be the most abundantly expressed adenosine receptor in the same model, and its activation is driven by elevated extracellular adenosine under hypoxia. This axis integrates a feed-forward mechanism, where ADORA2B amplifies hypoxia signaling via β-catenin–HIF-1α cooperation, and a positive feedback loop, where HIF-1α transcriptionally upregulates ADORA2B. Disruption of this circuit attenuated hypoxia-responsive gene expression and reduced PD-L1 levels, suggesting a viable therapeutic target for modulating immune evasion. Taken together, these studies illustrate how contextual features of GPCR signaling—ligand availability, receptor expression, and G protein coupling versatility—can be strategically leveraged to develop innovative diagnostic platforms and dissect oncogenic adaptation mechanisms. By bridging assay engineering with tumor biology, this dissertation highlights the broader implications of GPCR functional plasticity in biomedical research and clinical translation.
