Suspended sediment (SS) generated from anthropogenic activities such as marine sand mining and coastal development has emerged as a key physical stressor in marine ecosystems. While SS is often recognized for its turbidity-related effects, its biological consequences have remained underexamined, particularly in terms of developmental sensitivity, trait-based susceptibility, and sublethal physiological disruptions. Given the lack of standardized SS thresholds in sediment quality guidelines, a trait-explicit and stage-informed framework is urgently needed. This study adopted a multi-taxa, multi-endpoint experimental design to examine the ecotoxicological effects of SS across microalgae, bivalves, and fish—three taxonomic groups representing distinct ecological roles and physiological characteristics. By integrating multiple lines of evidence (MLOE), including growth inhibition, developmental toxicity, and physiological impairment, this work aimed to derive robust effect thresholds and provide ecological insight into SS risk assessment.
To quantify sublethal SS effects on primary producers, microalgal bioassays were conducted using SS collected from coastal environments. Growth inhibition was assessed for six microalgal species, and multiple endpoints were measured using flow cytometry and pulse-amplitude modulation (PAM) fluorometry for three species (Dunaliella tertiolecta, Isochrysis galbana, and Phaeodactylum tricornutum). Among these, the median effective concentration (EC50) for growth inhibition of D. tertiolecta (6700 mg L–1) was notably lower compared to the other species, and among several endpoints, esterase activity was the most inhibited. Species-specific responses to SS exposure were identified, with D. tertiolecta exhibiting greater susceptibility across most endpoints. Meanwhile, measurements of Fo’, Fm’, and Y(NPQ) in P. tricornutum using PAM fluorometry revealed greater sensitivity. Based on the results of this study and review, the tentative predicted no-effect concentration (PNEC) was calculated as 12.1 mg L–1. Overall, this study provides novel insights into SS ecotoxicity, establishing a crucial baseline for future investigations.
To evaluate developmental sensitivity across benthic invertebrates, SS-induced biological responses were experimentally assessed in six bivalve species spanning four ontogenetic stages: egg, larva, juvenile, and adult. Sub-lethal and lethal endpoints were quantified, revealing strong size-dependent tolerance. In Ruditapes philippinarum, the median lethal concentration (LC50) values increased nearly 40-fold from larva to adult, and across all species, relative body size positively correlated with threshold concentrations (r = 0.898). Species sensitivity distributions (SSD) showed substantial variation depending on stage inclusion. The hazardous concentration for 5% of species (HC5) dropped from 671 mg L–1 (juvenile–adult) to 220 mg L–1 when early stages were incorporated, yielding a PNEC of 22 mg L–1. These results highlight the vulnerability of early stages and demonstrate the need for ontogeny-informed SS risk assessments to derive protective sediment guidelines.
To further explore SS effects in higher trophic organisms, the physiological responses of four marine fish species (Lateolabrax japonicus, Sebastes schlegelii, Platichthys stellatus, and Paralichthys olivaceus) were assessed under SS exposure generated by marine sand mining. Using oxygen consumption rate (OCR), osmolality, and mortality as endpoints, the effects of SS concentrations ranging from 0 to 10,000 mg L–1 were evaluated. L. japonicus exhibited the highest sensitivity in OCR, with an EC50 of 15,812 mg L–1, responding more rapidly to SS exposure. In contrast, S. schlegelii demonstrated the highest mortality, with an LC50 of 340,921 mg L–1. SS exposure caused diel variations in OCR across species, highlighting the necessity of accounting for such differences. Based on the results of this study and review, species sensitivity to SS is influenced by habitat type, activity patterns, and developmental stage, emphasizing the importance of incorporating species-specific traits to accurately evaluate the ecological impacts of suspended sediment on marine ecosystems.
Collectively, this thesis emphasizes the importance of integrating biological traits and developmental stages into assessments of SS impacts. Trait-based comparisons across taxa revealed that susceptibility to SS was strongly influenced by diel behavior and ecological niche of each species. SSD-based modeling showed that inclusion of early developmental stages significantly reduced protective thresholds, thereby challenging sediment guidelines established using data from adults alone. In addition, physiological endpoints such as oxygen consumption and esterase activity served as sensitive early-warning indicators of SS exposure across various organisms. These findings underscore the need for ecologically realistic and trait-explicit sediment management strategies and reaffirm the effectiveness of a MLOE approach in improving the predictive accuracy of ecological risk assessments under intensifying sediment-related disturbances.
