    Accurate and early detection of neurodegenerative diseases (NDs) remains a critical need in clinical neuroscience. This thesis systematically evaluates two promising biomarkers candidate ubiquitin (Ub) and melatonin—across a spectrum of NDs through quantitative meta-analysis. Focused conditions include Alzheimer's disease (AD), Parkinson's disease (PD), Huntington's disease (HD), Creutzfeldt-Jakob disease (CJD), frontotemporal dementia (FTD), vascular dementia (VAD), and mild cognitive impairment (MCI). Ubiquitin, a protein involved in cellular proteostasis through targeted degradation of misfolded proteins, was consistently elevated in cerebrospinal fluid (CSF) of individuals with AD [RoM = 1.68, 95% CI (1.30 to 2.06)] and showed the most prominent increase in CJD [RoM = 4.51, 95% CI (3.71 to 5.31)]. In contrast, AD patients exhibited reduced blood Ub levels, highlighting biofluid-specific biomarker dynamics. These findings support Ub's role as a pathophysiological and diagnostic marker of neurodegeneration. Melatonin, a hormone regulating circadian rhythms, demonstrated disease-specific alterations, particularly in PD and HD. PD patients exhibited decreased melatonin amplitude [RoM = 0.76] and increased Area under the curve [RoM = 1.06], with significant modulation by age and medication status. Denovo patients had higher nighttime and morning melatonin levels, while those on dopaminergic treatment showed lower levels. Notably, patients with sleep disturbances had markedly different melatonin profiles. In HD, progressive melatonin declines from early to advanced stages contrast with an unexpected elevation in the premanifest stage, suggesting that circadian hormone regulation may precede clinical onset. Collectively, these findings underscore the biomarker potential of both ubiquitin and melatonin across different stages and types of neurodegenerations. Ubiquitin may serve as a general indicator of neurodegeneration severity, particularly in AD and CJD, while melatonin offers promise for early diagnosis in PD and monitoring disease progression in HD. These molecular markers could complement existing diagnostic tools and inform targeted early interventions in clinical practice.
