    The mTOR/S6K1 signaling pathway is regulated by various factors such as growth hormones, amino acids, and glucose, and through its downstream pathways, it influences cell growth, proliferation, protein synthesis, and survival, playing a critical role in maintaining cellular function. Studies in mouse models have shown that disruption or dysregulation of the mTOR/S6K1 signaling pathway can impair developmental processes in multiple tissues, including the brain, heart, and pancreas. Furthermore, improper regulation of the mTOR pathway is also known to affect human brain development, contributing to conditions such as autism spectrum disorder and cognitive developmental delays.
    Most research on the mTOR/S6K1 signaling pathway has been conducted in cancer cells, leading to the identification of many downstream effectors involved in cell proliferation, protein synthesis, and autophagy. However, how this pathway influences tissue development and differentiation through specific downstream mechanisms remains poorly understood. In this study, I investigated the role of the S6K1 signaling pathway in differentiation by generating S6K1-depleted human embryonic stem cells via gene editing and applying a stem cell-derived pancreatic beta cell differentiation protocol. I found that the loss of S6K1 suppresses the expression of transcription factors involved in epithelial–mesenchymal transition (EMT), thereby disrupting normal transitions in cytoskeletal organization and cell–cell junctions. This disruption impairs the formation of three-dimensional cellular architecture, reduces cellular diversity, and leads to a loss of cell populations responsible for expressing signaling molecules critical for pancreatic endocrine differentiation.
    Furthermore, in brain organoids derived from human embryonic stem cells carrying the Smith-Kingsmore syndrome genotype, which is associated with mTOR hyperactivation, I observed increased organoid size and abnormal cellular organization around neural rosettes.
    Through these investigations, this study demonstrates that the mTOR/S6K1 signaling pathway regulates cellular differentiation during three-dimensional development by modulating multicellular organization and structural arrangement.

    mTOR/S6K1 신호 전달 체계는 성장 호르몬, 아미노산, 당류 등 다양한 요인에 의해 영향을 받으며, 하위 조절 경로로 세포의 성장, 증식, 단백질 발현 및 생존에 영향을 미쳐, 세포의 기능 조절 유지에 매우 중요하다. mTOR/S6K1 신호 전달 체계가 붕괴되어 잘 조절되지 않았을 때, 뇌, 심장, 췌장 등 다양한 조직에서 발달 과정에 문제가 생긴다는 것이 쥐 실험에서 알려져 있었다. 또한, mTOR 신호 전달 체계의 잘못된 조절이 인간의 뇌 발달 과정에 영향을 미쳐 자폐 스펙트럼이나 인지 발달 장애를 일으킨 다는 점 역시 잘 알려져 있다.
