Background and Objectives: Methionine restriction (MetR) is currently a commonly used adjuvant treatment in cancer therapy. Nevertheless, existing studies on MetR have paid little attention to its potential implications for fibrosis. Hypertrophic scar (HTS) is an abnormal fibrotic response during wound healing process, resulting from the excessive activation of fibroblasts. Due to the absence of a fully effective pharmacological treatment, HTS frequently causes great annoyance on patients as a common sequela of burns. To date, the effects of MetR on HTS fibroblasts (HTSFs) remain unclear, which is precisely the objective of our study.
Methods: We isolated HTSFs from post-burn HTS tissues, and cultured the experimental group cells in specially prepared MetR medium. Cell images and immunocytochemistry staining images were captured separately through light and fluorescence microscope. Cell proliferation was evaluated through CellTiter-Glo Luminescent Cell Viability Assay Kit. mRNA and protein expression were detected through quantitative real-time polymerase chain reaction and western blot.
Results and Conclusions: In HTSFs, MetR reduced cellular inflammation and downregulates multiple signaling pathways, including TGF-β-SMADs, STATs, and AKT/mTOR, as well as upregulating the MAPKs. Further, MetR arrested cell cycle, promotes apoptosis, suppressed cell proliferation and migration, finally reduced extracellular matrix protein secretion, thereby exerting multi-faceted inhibitory effects on HTS formation.
