아스코르브산(AA), 도파민(DA), 요산(UA)은 생체 내 존재하는 중요한 생리활성 물질로서, 각 물질의 농도는 괴혈병, 파킨슨병, 알츠하이머병, 통풍 등과 같은 다양한 질병과 밀접한 관련이 있다. 따라서, 이들의 농도를 실시간으로 정밀하게 측정하는 것은 질병의 조기 진단 및 치료에 있어 매우 중요하다.
현재 AA, DA, UA는 라만 산란, 크로마토그래피, 형광 분석, 전기화학 분석 등의 다양한 분석 기법으로 검출이 가능하며, 이 중 전기화학적 방법은 고감도, 빠른 응답 속도, 경제성 등의 이점으로 인해 휴대 가능한 생체 분자 센싱 플랫폼으로 주목받고 있다.
하지만, 전기화학적 방법은 AA, DA, UA가 비슷한 산화 전위를 가지고 있어 산화 신호가 중첩되며 선택적 검출이 어렵다는 단점이 있다. 이러한 한계를 극복하기 위해 금속 나노입자, 그래핀, 탄소나노튜브 등과 같은 나노소재를 활용한 전극 개질 기법이 적용되고 있으며, 이들 소재는 전극의 유효 표면적을 증가시켜 감도를 향상시키고, 산화 전위 분리도를 확보하여 선택성을 높이는 데 효과적이다.
본 연구에서는, 시트르산으로 기능화된 다중벽 탄소나노튜브(CA-MWCNT)를 유리탄소 전극(GCE) 표면에 드롭캐스팅 방식으로 고정하고, 금 나노입자(AuNPs)를 단순 담금 방식을 통해 환원시켜 개질 전극(AuNPs/CA-MWCNT/GCE)을 제작하였다.
제작된 전극은 주사전자현미경(SEM), 에너지 분산형 X선 분광법(EDS), 퓨리에 변환 적외선 분광법(FT-IR), 접촉각 분석(WCA)을 통해 표면 구조 및 원소 조성을 확인하였으며, 순환 전압전류법(CV)을 통해 유효 표면적 증가를 정량적으로 분석하였다. 펄스 차이 전압전류법(DPV) 측정을 통해 AA, DA, UA를 각각 -0.088 V, 0.148 V, 0.276 V에서 명확하게 구분된 산화 신호로 검출할 수 있었으며, 검출 한계는 각각 300 μM, 1.5 μM, 6.0 μM으로 측정되었다. 또한, pH에 따른 반응 메커니즘 분석, 간섭 물질에 대한 선택성 평가, 반복 측정을 통한 안정성 검증을 통해 고감도, 고선택성을 확인하였다.
이러한 결과를 바탕으로, 본 연구에서의 전극 제작 방식은 간단한 공정으로 대량 생산이 가능하며, 나아가서는 유연 기판 적용을 통해 웨어러블 센서 플랫폼으로도 확장 가능성을 보인다.

Ascorbic acid (AA), dopamine (DA), and uric acid (UA) are biologically important molecules whose concentrations are closely related to several diseases such as Parkinson’s disease, Alzheimer’s disease, and gout. Therefore, real-time and accurate monitoring of these analytes is crucial for early diagnosis and treatment. Although various analytical methods such as Raman scattering, chromatography, fluorescence, and electrochemical techniques have been developed, electrochemical sensing is particularly attractive due to its high sensitivity, fast response, and cost-effectiveness, making it suitable for portable biosensing platforms.
However, electrochemical detection of AA, DA, and UA faces challenges due to their overlapping oxidation potentials, which hinders selective analysis. To overcome this, electrode surface modification using nanomaterials—such as metal nanoparticles, graphene, and carbon nanotubes—has been proposed to enhance sensitivity and enable peak separation.
In this study, we fabricated a modified glassy carbon electrode (GCE) by drop-casting citric acid-functionalized multi-walled carbon nanotubes (CA-MWCNTs) and reducing gold ions (Au³⁺) in situ using citric acid to form gold nanoparticles (AuNPs). The resulting AuNPs/CA-MWCNT/GCE was characterized by SEM and EDS, and its electrochemical properties were investigated by cyclic voltammetry (CV) and differential pulse voltammetry (DPV). The modified electrode exhibited distinct oxidation peaks for AA, DA, and UA at −0.088 V, 0.148 V, and 0.276 V, respectively, with detection limits of 300 μM, 1.5 μM, and 6.0 μM.
The electrode also showed pH-dependent oxidation behavior, high selectivity against common interferents, and excellent long-term electrochemical stability. This simple fabrication approach is suitable for large-scale production and can be extended to flexible substrates, offering potential for wearable electrochemical sensors.
