    Divergent β-adrenergic receptor signaling across species : implications for brown adipocyte thermogenesis

    G protein-coupled receptors (GPCRs) can elicit multiple intracellular signaling pathways by primarily coupling with G proteins (Gα, Gβ and Gγ subunits) or β-arrestin upon activation. Beta-adrenergic receptors(β-ARs), a subset of GPCRs, are well known for their regulatory roles in brown adipose tissue. Under cold exposure or ligand stimulation, β-ARs mediated coupling to Gαs and enhanced cAMP production, leading to thermogenesis. And little is known about the potential coupling of β-ARs to non-Gαs proteins. Recently, a comprehensive understanding of downstream signaling with Gα subunits has received attention as a broad approach to discover novel signaling partners beyond established physiological mechanisms. Here, using BRET-based biosensors known as TRUPATH, we dissected the ability of the endogenous agonist norepinephrine and the synthetic agonist isoproterenol to couple 14 different Gα subunits through human and rodent β-ARs. We demonstrated that (i) endogenous and synthetic agonist activated distinct Gα protein signaling repertoires, (ii) humans and mice β-ARs engage different signaling partners, with a broader range of signaling observed in mice, and (iii) novel Gαi coupling, especially in mice, affects the brown adipose tissue thermogenesis. Our study highlights the importance of capturing the full range of Gα protein partners of GPCRs to gain a complete understanding of downstream signaling and physiological mechanisms. Furthermore, the observed species-specific differences or ligand-dependent effects in G protein coupling between rodents and humans highlight the necessity of careful ligand selection and validation in preclinical animal studies. Lastly, exploring the unique physiological roles and mechanisms of the novel Gαi/o family coupling, particularly in mice, will be an interesting area for further research.

    G 단백질 결합 수용체 (GPCR)는 활성화 시 G 단백질 또는 β-arrestin과 결합하여 다양한 세포 내 하위 신호 전달 경로를 유도한다.
