    바이오센서를 활용한 단일세포의 전기적 분석은 세포 전체 집단에서 특이한 거동을 보이는 일부 하위 집단 세포를 구별하거나 세포의 복잡한 생리학적 상태를 연구하는데 중요한 부분이다. 따라서 본 연구는 바이오센서를 제작하고 시간에 따른 정전용량 변화를 측정하여 NIH/3T3 단일 세포의 정전용량을 분석하고자 하였다. 특히 세포가 성장하면서 정전용량이 증가하는 구간을 집중적으로 분석하였으며 이를 위해 세포의 개체 수를 카운팅하여 시간에 따라 세포의 증가 정도를 확인하고 측정된 정전용량과 비교하여 세포 성장 전반부(0 ~ 2.33시간)와 후반부(2.33 ~ 48시간) 사이 정전용량 측정에 대한 메커니즘과 단일 세포 정전용량을 제시하였다.
    결과적으로 전반부는 배지에 떠있는 세포가 가라앉는 단계이기 때문에 현미경 이미지 상에서 세포가 증가하는 정도는 빠르지만 센서에 부착된 세포의 면적이 작아 단일 세포 정전용량은 낮은 수치(300 kHz에서 0.67 fF)를 보이며, 후반부에서는 대부분의 세포가 성장하는 단계이기 때문에 세포 분열에 따라 세포 수가 증가하므로 전반부에 비해 세포 증가 속도는 느리면서 센서와 세포 사이 부착면적은 넓기 때문에 단일 세포 정전용량은 높은 수치 (300 kHz에서 1.98 fF)를 보이는 것을 확인하였다. 이러한 경향성은 다른 여러 주파수에서 측정된 정전용량에서도 확인되었으며, 측정을 반복하여 주파수별로 전반부와 후반부의 단일세포 정전용량을 제시하였다.
    바이오센서는 데이터 확보 효율성의 증가와 공정 변수 및 측정 환경에 대한 변수 조절, 분석에 필요한 검체 수 감소의 관점에서 지속적인 발전이 필요하다. 이를 위해서 기존의 단일 센서를 멀티-웰 바이오센서로 발전시켜 디자인을 확립하고 센서를 제작하여 기초 데이터를 얻고자 하였다. 이때 패턴을 어레이하고 분석에 필요한 검체 수를 감소시킬 수 있는 바이오센서의 제작을 위해서는 기존 센서의 패턴을 축소시키는 것이 중요하므로, 우선 센서의 패턴 사이즈 변화가 정전용량 측정에 미치는 영향이 없는지 분석하였다. 기존 센서의 패턴을 기준으로 1.25배, 0.75배, 0.5배 한 패턴을 설계하고 단일 센서를 제작하여 측정을 수행한 결과 세포의 성장 및 사멸에 따른 정전용량의 증가, 감소 개형이 패턴 사이즈에 관계없이 100 kHz 이후에서 올바르게 측정되는 경향성이 나타났다. 따라서 패턴 사이즈의 변화가 정전용량 측정에 영향을 미치지 않는 것으로 판단하고, 축소된 패턴을 2 × 2 형태로 배열하여 센서를 설계하였다. 그 후 동일한 측정을 수행하여 정전용량 개형의 재현성과 웰 간의 균일성을 확인하였으며, 이를 통해 추후에는 웰 사이 조건을 변경하여 독립적인 데이터를 동시에 얻거나 배열하는 패턴의 수를 늘리는 등 멀티-웰 바이오센서를 지속적으로 발전시킬 예정이다.

    The use of biosensors to analyze single cells is crucial for studying complex physiological states or identifying subpopulations of cells with unusual behavior. In this study, we analyzed the capacitance of NIH/3T3 single cells by fabricating biosensors and measuring changes in capacitance of groups of cells over time. Our focus was on the section where capacitance increases as cells grow, particularly in comparing the early stage (0-2.33 hours) to the later stage (2.33-48 hours) of cell growth. To accomplish this, we tracked the cell count over time and compared it with the measured capacitance.
    As a result, we found that in the early stage, the cells floating in the medium are sinking, so the appearance of cell in the microscopic image is fast, but the area of the cell attached to the sensor is small, so the single cell capacitance is low (0.67 fF at 300 kHz), and in the late stage, most of the cells are growing, so the number of cells increases by cell division, therefore the appearance of cell is slow compared to the early stage, but the area of attachment between the sensor and the cell is large, so the single cell capacitance is high (1.98 fF at 300 kHz). This trend was also confirmed for capacitance measured at several other frequencies, and the measurements were repeated to present the single cell capacitance per frequency in early and later stage.
    Biosensors should continue to evolve by increasing data acquisition efficiency, controlling process variables and measurement environment, and reducing the number of samples required for analysis. To achieve this goal, we expanded the single sensor into a multi-well biosensor. In order to create a biosensor capable of pattern arraying and reducing the number of samples needed for analysis, we needed to decrease the size of the original sensor pattern. Therefore, We designed patterns that were 1.25x, 0.75x, and 0.5x the size of the original sensor to determine whether altering the pattern size of the sensor would affect capacitance measurement. We fabricated a single sensor to perform measurements and found that the increase and decrease in capacitance due to cell growth and death were measured correctly after 100 kHz, regardless of the pattern size. Therefore, we concluded that the change in pattern size did not affect the capacitance measurement, and we designed a sensor by arranging the reduced patterns in a 2 × 2 format. The measurements were repeated to confirm the reproducibility of the capacitance shape and the uniformity between wells. This information will be used to develop the multi-well biosensor by altering the conditions between wells to obtain independent data simultaneously or increasing the number of arranged patterns.
