Cancer is the ability of cancer cells that grow by attracting newly formed blood vessels in the human vascular system to travel through the bloodstream and cause metastasis to other organs, greatly reducing the patient's survival rate. Extensive testing of inhibitors and anticancer drugs is important to improve survival. However, prior research is needed to establish a model for testing. In this study, we conducted a study to build a model by evaluating gelatin bioink and silk bioink to evaluate extracellular matrix. The evaluated bioinks were mixed in appropriate ratios to confirm their ability to create various structures through 3D printing. Additionally, we confirmed the suitability of these inks for cell culture through cell survival tests and observed the successful formation of vascular networks by vascular cells within the structures, indicating their potential for vascularization. The second focus of our study was to determine angiogenesis in cancer cells. A model mimicking blood vessels was designed through perfusion culture and its suitability for organ culture was confirmed. Using immunofluorescence staining, we observed vascular cells growing toward cancer cells, indicating that cancer cells induce the formation of new blood vessels. We also observed cancer cells moving toward newly formed blood vessels. Immunofluorescence staining for specific factors between vascular cells and cancer cells revealed the presence of cancer cells within vascular cells. It was confirmed that when new blood vessels fully grow toward cancer cells, cancer cells can move through the newly formed blood vessels. In conclusion, we evaluated bioinks for 3D printing, assessed cell survival within the ink, confirmed vascular network formation within the structures, and assessed interactions between vascular cells and cancer cells through perfusion culture and immunofluorescence staining. This model provides insight into angiogenesis and cancer metastasis through 3D printing, paving the way for future evaluation of factors released by cancer cells in the culture medium. Additionally, we anticipate the potential to test anti-cancer drugs and inhibitors in perfusion culture channels to confirm cancer cell death.
