    This dissertation explores molecular mechanisms underlying sarcopenia, an age-related skeletal muscle disorder. It focuses on the emerging multiple roles of primary cilia such as muscle fusion, contraction, and myelination. A key theme is how ciliogenesis is regulated, and what function cilia serve in modulating critical muscle cell processes.
    First, it investigates how decreased expression of ADSSL1, a muscle-specific enzyme, contributes to sarcopenia. The findings suggest ADSSL1 induces autophagy-mediated actin depolymerization, facilitating myoblast fusion via primary cilia formation. Next, it reveals ACOX3 enzyme activity likely coordinates actin remodeling and ciliogenesis to influence muscle contraction. Finally, it demonstrates muscle cell primary cilia communicate with Schwann cells through extracellular matrix proteins like FBLN5 to support nerve myelination and muscle function.
    Together, these studies position primary cilia as crucial integration hubs connecting signals from diverse biochemical, structural and functional pathways to coordinate muscle cell fusion, force generation and nerve interactions.     우리 모두는 나이가 들어 감에 따라 자연스러운 신체적 변화를 겪게 된다. 이러한 변화는 근육 감소, 뼈 밀도 감소, 인지 기능 저하 등의 형태로 나타날 수 있는데, 그 중 근육의 감소는 60세 이후부터 더욱 가속되어 1년에 2~3%의 근육 감소를 보인다. 이렇게 노화에 의한 근육량의 급격한 감소와 근육 기능의 저하되는 상태를 근감소증(sarcopenia)이라고 한다. 노인성 근감소증은 독립보행의 장애를 일으킬 뿐만 아니라 기초대사율을 저하시켜 인슐린 저항성을 높이고 당뇨, 비만, 심혈관 질환 등 2차 노인성 만성질환을 유발한다. 노인성 근질환은 노화에 의한 만성염증 상태로 인해 근원세포의 융합이 억제되어 근육의 항상성이 유지되지 못하고 근섬유가 빠르게 소실돼 치료가 더욱이 어렵다. Sarcopenia의 근본적인 치료법은 알려져 있지 않으며, 현재의 치료는 각 개인에게 나타나는 특정 증상의 완화 및 보조기구의 사용 정도임Sarcopenia의 분자적 기전 규명과 해당 질환에 대한 더 깊은 이해를 바탕으로 한 근본적인 치료 방안이 몹시 필요한 실정이다.
    앞서 다른 연구진들은 동물 모델을 사용한 노화 유전자 발현 연구에서는 최대 75%의 유전자 발현이 노화와 관련될 수 있다는 것을 발견했다. 연구에 따르면 후성유전학적 시계(Epigenetic Clocks)가 빠른 사람은 생물학적으로 더 빨리 노화를 겪는다고도 알려진 것을 본다면, 근감소증과 관련된 유전자 발현의 변화를 이해하고, 더 나아가 노화유전자를 발굴하고 기전을 이해하는 것은 근감소증 치료에 새로운 패러다임을 제시할 것이라는 것을 시사한다.
